Assessing evaluation procedures for individual researchers: the case of the Italian National Scientific Qualification
The Italian National Scientific Qualification (ASN) was introduced as a
prerequisite for applying for tenured associate or full professor positions at
state-recognized universities. The ASN is meant to attest that an individual
has reached a suitable level of scientific maturity to apply for professorship
positions. A five member panel, appointed for each scientific discipline, is in
charge of evaluating applicants by means of quantitative indicators of impact
and productivity, and through an assessment of their research profile. Many
concerns were raised on the appropriateness of the evaluation criteria, and in
particular on the use of bibliometrics for the evaluation of individual
researchers. Additional concerns were related to the perceived poor quality of
the final evaluation reports. In this paper we assess the ASN in terms of
appropriateness of the applied methodology, and the quality of the feedback
provided to the applicants. We argue that the ASN is not fully compliant with
the best practices for the use of bibliometric indicators for the evaluation of
individual researchers; moreover, the quality of final reports varies
considerably across the panels, suggesting that measures should be put in place
to prevent sloppy practices in future ASN rounds
Five-minute Apgar score and educational outcomes: retrospective cohort study of 751 369 children
Background: The Apgar score is used worldwide for assessing the clinical condition and short-term prognosis of newborn infants. Evidence for a relationship with long-term educational outcomes is conflicting. We investigated whether Apgar score at 5 min after birth was associated with additional support needs (ASN) and educational attainment.
Methods: Data on pregnancy, delivery and later educational outcomes for children attending Scottish schools between 2006 and 2011 were collated by linking individual-level data from national educational and maternity databases. The relationship between Apgar score and overall ASN, type-specific ASN and educational attainment was assessed using binary, multinomial and generalised ordinal logistic regression models, respectively. Missing covariate data were imputed.
Results: Of the 751 369 children eligible, 9741 (1.3%) had a low or intermediate Apgar score and 49 962 (6.6%) had ASN. Low Apgar score was independently associated with overall ASN status (adjusted OR for Apgar ≤3, OR 1.52 95% CI 1.35 to 1.70), as well as ASN due to cognitive (OR 1.26, 95% CI 1.09 to 1.47), sensory (OR 2.49 95% CI 1.66 to 3.73) and motor (OR 3.57, 95% CI 2.86 to 4.47) impairments. There was a dose-response relationship between Apgar score and overall ASN status: of those scoring 0–3, 10.1% had ASN, compared with 9.1% of those scoring 4–7 and 6.6% of those scoring 7–10. A low Apgar score was associated with lower educational attainment, but this was not robust to adjustment for confounders.
Conclusions: Apgar scores are associated with long-term as well as short-term prognoses, and with educational as well as clinical outcomes at the population level
Quantitative Analysis of the Italian National Scientific Qualification
The Italian National Scientific Qualification (ASN) was introduced in 2010 as
part of a major reform of the national university system. Under the new
regulation, the scientific qualification for a specific role (associate or full
professor) and field of study is required to apply to a permanent professor
position. The ASN is peculiar since it makes use of bibliometric indicators
with associated thresholds as one of the parameters used to assess applicants.
Overall, more than 59000 applications were submitted, and the results have been
made publicly available for a short period of time, including the values of the
quantitative indicators for each applicant. The availability of this wealth of
information provides an opportunity to draw a fairly detailed picture of a
nation-wide evaluation exercise, and to study the impact of the bibliometric
indicators on the qualification results. In this paper we provide a first
account of the Italian ASN from a quantitative point of view. We show that
significant differences exist among scientific disciplines, in particular with
respect to the fraction of qualified applicants, that can not be easily
explained. Furthermore, we describe some issues related to the definition and
use of the bibliometric indicators and thresholds. Our analysis aims at drawing
attention to potential problems that should be addressed by decision-makers in

Comparison of nitrification inhibitors to restrict nitrate leaching in a maize crop irrigated under mediterranean conditions
The aim of this paper was to compare dicyandiamide (DCD) and 3,4 dimethylpyrazole phosphate (DMPP) as inhibitors of ammonium oxidation and nitrate leaching after applying fertilizer to a maize (Zea mays L.) crop grown under Mediterranean conditions. The effects of nitrification inhibitors were also compared to those of N fertilization without inhibitors and with split N application. In plots fertilized with ammonium sulphate nitrate (ASN), either DCD or DMPP lengthened ammonium presence in soil and produced lower soil NO3- concentrations (30% lower than in plots with no inhibitor). The use of DCD or DMPP achieved significant reductions in nitrate leaching. DCD showed excellent properties for controlling nitrate leaching, taking into account the fact that grain yield and N accumulated by plant were similar for the ASN-DCD and ASN treatments applied at the same N doses. The split N treatment did not offer any advantages in terms of leached nitrate, either with the use of single ammonium sulphate nitrate (ASN) or with single application of nitrification inhibitors. The nitrification inhibitors did not increase the yield but did not reduce it either. The drainage rate was the most important component of nitrate leaching. The low drainage values of the first year resulted in a sharp decline of nitrate leaching. However, the experiment of the second year, showed clear differences in nitrate leaching between treatments due to the greater drainage
Competitive pathways for peptide deamidation
Asparagine (Asn) residues spontaneously – yet non-enzymatically – deamidate to form aspartate under physiological conditions, causing time-dependent changes in the conformation of proteins, limiting their lifetime [1]. The 'molecular clocks' hypothesis [2], suggests that deamidation is a biological molecular timing mechanism that could be set to any desired time interval by genetic control of the protein structure and the immediate environment of the Asn residue. The fact that deamidation occurs over a wide range of biologically relevant time intervals suggests that different mechanisms may be at play. To date deamidation is believed to occur over a succinimide-mediated pathway [3]. A novel route leading to the succinimide intermediate via tautomerization of the Asn side chain amide functionality was recently proposed [4,5]. The current study introduces a new 'competing' route for the deamidation of asparagine residues. The aim is to comparatively analyze the feasibility of this new mechanism against the traditional succinimide route, taking into account the catalytic effect of the solvent environment. For this purpose, QM dynamics and meta-dynamics calculations were performed on a model peptide placed in a periodic water box. These results will identify the lowest energy pathway for asparagine deamidation and will serve as a stepping stone for QM/MM calculations of Asn deamidation in proteins
Syndecan-4 knockout leads to reduced extracellular transglutaminase-2 and protects against tubulointerstitial fibrosis
Transglutaminase type 2 (TG2) is an extracellular matrix crosslinking enzyme with a pivotal role in kidney fibrosis. The interaction of TG2 with the heparan sulfate proteoglycan syndecan-4 (Sdc4) regulates the cell surface trafficking, localization, and activity of TG2 in vitro but remains unstudied in vivo. We tested the hypothesis that Sdc4 is required for cell surface targeting of TG2 and the development of kidney fibrosis in CKD. Wild-type and Sdc4-null mice were subjected to unilateral ureteric obstruction and aristolochic acid nephropathy (AAN) as experimental models of kidney fibrosis. Analysis of renal scarring by Masson trichrome staining, kidney hydroxyproline levels, and collagen immunofluorescence demonstrated progressive fibrosis associated with increases in extracellular TG2 and TG activity in the tubulointerstitium in both models. Knockout of Sdc-4 reduced these effects and prevented AAN-induced increases in total and active TGF-b1. In wild-type mice subjected to AAN, extracellular TG2 colocalized with Sdc4 in the tubular interstitium and basement membrane, where TG2 also colocalized with heparan sulfate chains. Heparitinase I, which selectively cleaves heparan sulfate, completely abolished extracellular TG2 in normal and diseased kidney sections. In conclusion, the lack of Sdc4 heparan sulfate chains in the kidneys of Sdc4-null mice abrogates injury-induced externalization of TG2, thereby preventing profibrotic crosslinking of extracellular matrix and recruitment of large latent TGF-b1. This finding suggests that targeting the TG2- Sdc4 interaction may provide a specific interventional strategy for the treatment of CKD
