NF-κB p65 Subunit Is Modulated by Latent Transforming Growth Factor-β Binding Protein 2 (LTBP2) in Nasopharyngeal Carcinoma HONE1 and HK1 Cells
NF-kappa B is a well-characterized transcription factor, widely known as a key player in tumor-derived inflammation and cancer development. Herein, we present the functional and molecular relevance of the canonical NF-kappa B p65 subunit in nasopharyngeal carcinoma (NPC). Loss-and gain-of-function approaches were utilized to reveal the functional characteristics of p65 in propagating tumor growth, tumor-associated angiogenesis, and epithelial-to-mesenchymal transition in NPC cells. Extracellular inflammatory stimuli are critical factors that trigger the NF-kappa B p65 signaling; hence, we investigated the components of the tumor microenvironment that might potentially influence the p65 signaling pathway. This led to the identification of an extracellular matrix (ECM) protein that was previously reported as a candidate tumor suppressor in NPC. Our studies on the Latent Transforming Growth Factor-beta Binding Protein 2 (LTBP2) protein provides substantial evidence that it can modulate the p65 transcriptional activity. Re-expression of LTBP2 elicits tumor suppressive effects that parallel the inactivation of p65 in NPC cells. LTBP2 was able to reduce phosphorylation of p65 at Serine 536, inhibit nuclear localization of active phosphorylated p65, and impair the p65 DNA-binding ability. This results in a consequential down-regulation of p65-related gene expression. Therefore, the data suggest that the overall up-regulation of p65 expression and the loss of this candidate ECM tumor suppressor are milestone events contributing to NPC development.published_or_final_versio
Divulgação de informação sobre o governo das sociedades em Portugal: Extensão e determinantes
Tese de Doutoramento em ContabilidadeO objetivo geral desta pesquisa é identificar os determinantes do nível da divulgação da
informação sobre o governo das sociedades em Portugal, considerando o modelo de
divulgação comply or explain. Na primeira fase, foi dada atenção à pressão social exercida
pelo contexto macroeconómico e, numa segunda fase, pusemos a ênfase nos mecanismos
de governo das sociedades para explicar as diferenças no nível de divulgação de
informação. Metodologicamente, utilizamos uma amostra de empresas portuguesas
cotadas na Euronext Lisbon, entre 2005 e 2011. Na análise estatística utilizou-se um
modelo de regressão ordinal logit, enquadrado pelas teorias da agência, sinalização,
stewardship, stakeholders e institucional. Através da análise de conteúdo da informação
contida nos relatórios e contas anuais e nos relatórios de governo societário, construiu-se
um índice de divulgação global, com as seguintes categorias: conselho de administração
e estrutura de gestão, comissões especializadas, auditoria e gestão de riscos, estrutura de
propriedade, conformidade e responsabilidade empresarial e transparência financeira. Os
mesmos itens de informação foram agrupados em duas outras categorias: voluntária e
recomendada ou obrigatória. Os resultados mostram uma evolução positiva do nível de
divulgação de informação sobre o governo das sociedades. Indicam também quais as
categorias de informação mais e menos divulgadas. Os resultados sugerem que os fatores
explicativos da divulgação sobre o governo das sociedades em Portugal são o sistema
legal, o desenvolvimento económico, a concentração da propriedade, a participação dos
administradores no capital, a participação dos investidores qualificados estrangeiros no
capital, o tamanho do conselho de administração, a independência do conselho de
administração, a estrutura de liderança, a qualidade da auditoria externa, o grau de
internacionalização e o nível de endividamento. Os resultados mantém-se constantes
quando consideramos o índice de divulgação voluntária, mas ao considerar-se o índice de
divulgação recomendada ou obrigatória, a participação dos administradores no capital, a
independência do conselho de administração e o grau de internacionalização deixam de
se mostrar explicativos. Finalmente, este estudo fornece um enquadramento teórico para a
análise da divulgação de informação sobre o governo das sociedades.The central purpose of this study is to identify the determinants of the level of corporate
governance disclosure by Portuguese listed companies, based on a “comply or explain”
model. First we analyze how the social pressure at the macroeconomic level influences
corporate governance disclosure; second, we emphasize the corporate governance
mechanisms to explain the differences of the level of corporate governance disclosure.
The proposed approach was based on data of a sample of Portuguese companies listed on
Euronext Lisbon from 2005 to 2011. An ordinal logistic regression model was used for
statistical analysis, framed by agency theory, signaling theory, stakeholder’s theory,
stewardship theory and institutional theory. The construction of the global corporate
governance disclosure index rely on the data extracted from the firm’s corporate
governance and annual reports using content analysis. The disclosure index was classified
into the following categories: board of directors and management structure, corporate
committees, audit and risk management, ownership concentration, corporate
responsibility and compliance and financial transparency. The same information items
were gathered in two other disclosure indexes: voluntary and mandatory or
recommended. Our results reveal that the level of corporate governance disclosure
evolved positively, during the period 2005–2011. The categories of information that
presents the highest and lowest level of disclosure are also specified. The results also
suggest that the legal system, economic development, ownership concentration, board of
director’s ownership, qualified foreign investor’s ownership, board size, board
independence, leadership structure, external audit quality, internationalization degree and
debt are the determinants of the level of corporate governance disclosure. The results
remain the same when we consider the voluntary disclosure index, but once we consider
the recommended or mandatory disclosure index, the variables board ownership, board
independence and internationalization degree do not remain significant. Finally, this
