As leishmanioses constituem um conjunto de doen??as negligenciadas que afetam cerca
de 12 milh??es de indiv??duos em 98 pa??ses. Os medicamentos utilizados no controle da
infec????o est??o associados ?? baixa efic??cia, alta toxicidade, dificuldade de administra????o,
altos custos e crescente resist??ncia. A procura por novas alternativas de tratamento ??,
portanto, uma prioridade. Este trabalho tem como objetivo avaliar a atividade
leishmanicida de novas hidroxietilpiperazinas sint??ticas e de Momordica charantia
(Cucurbitaceae). Uma s??rie de oito hidroxietilpiperazinas foi sintetizada e testada, a que
demonstrou ser mais ativa e com menor citotoxidade, PMIC4, foi selecionada.
Promastigotas de Leishmania amazonensis foram cultivadas em diferentes
concentra????es da PMIC4, que inibiu acentuadamente a multiplica????o do parasito, com
IC50 de 11,6 ??M. Em amastigotas, os resultados mostraram que macr??fagos tratados
com 20 ??M de PMIC4 apresentaram uma redu????o de 82% nos n??veis de infec????o, com
IC50 de 1,8 ??M. Uma vez que hidroxietilpiperazinas s??o utilizadas como precursores na
s??ntese dos inibidores de protease do HIV, foram realizados ensaios para detectar se a
PMIC4 seria capaz de inibir a atividade de asp??rtico proteases em promastigotas de L.
amazonensis. Concentra????es de at?? 200 ??M de PMIC4 n??o foram capazes de inibir esta
atividade enzim??tica. Para avaliar se houve diferen??a na composi????o dos ester??is de
membrana entre L. amazonensis tratadas com a PMIC4 e n??o tratadas, os lip??deos
neutros foram analisados por cromatografia de camada fina. Foi poss??vel notar uma
diferen??a significativa entre os grupos tratados e o grupo controle. Para avaliar o efeito
desta hidroxietilpiperazina no controle da infec????o por L. amazonensis em
camundongos BALB/c, os animais foram tratados com a PMIC4, que mostrou possuir
atividade leishmanicida in vivo por via oral. Paralelamente, tamb??m foi demonstrada a
atividade leishmanicida de fra????es de Momordica charantia em promastigotas e
amastigotas intracelulares de L. amazonensis. Ao longo do trabalho, foi realizado o
fracionamento do extrato bruto das partes a??reas da planta, guiado pela atividade
leishmanicida, at?? a obten????o de tr??s fra????es semi-purificadas ativas. Em amastigotas
intracelulares, as fra????es ativas MS??Ac e F7 apresentaram IC50 de 1,69 ??g/mL e 1,08
??g/mL, respectivamente. Este ?? o primeiro relato tanto da atividade de uma
hidroxietilpiperazina quanto de Momordica charantia contra Leishmania amazonensis.
Os extratos, fra????es e subst??ncias aqui apresentados podem ser considerados candidatos
potenciais para o desenvolvimento de novos fitoter??picos e/ou quimioter??picos contra a
leishmaniose cut??nea.Leishmaniasis is a group of neglected diseases that affect approximately 12 million
people in 98 countries. The drugs used to control the disease are associated with low
efficacy, high toxicity, difficulties of administration, high costs and increasing
resistance. The search for new alternatives of treatment is a priority. This study aims to
evaluate the leishmanicidal activity of new synthetic hidroxyethylpiperazines and of
Momordica charantia (Cucurbitaceae). A series of eight hidroxyethylpiperazines was
synthesized and the PMIC4 showed to be more active and less cytotoxic. Leishmania
amazonensis promastigotes were cultivated in different concentrations of PMIC4, which
strongly inhibited the multiplication of the parasite, with IC50 of 11.6 ??M. In
amastigotes, the results demonstrated that macrophages treated with 20 ??M of PMIC4
presented 82% reduction in infection levels, with IC50 of 1.8 ??M. Once
hidroxyethylpiperazines are used as precursors in the synthesis of protease inhibitors of
HIV, we tested whether the PMIC4 could inhibit the activity of aspartic proteases in
promastigotes of L. amazonensis. Concentrations up to 200 ??M of PMIC4 were not able
to inhibit this enzyme activity. To determine differences in the composition of
membrane sterols between L. amazonensis treated with PMIC4 and untreated, the
neutral lipids were analyzed by thin layer chromatography. It was possible to observe a
significant difference between treated and control groups. To evaluate the effect of this
hidroxyethylpiperazine in controlling infection by L. amazonensis in BALB/c mice,
animals were treated with PMIC4, which showed in vivo leishmanicidal activity orally.
Moreover, we also demonstrated the leishmanicidal activity of fractions of Momordica
charantia in promastigotes and intracellular amastigotes of L. amazonensis. Throughout
the work, we performed the fractionation of crude extract of aerial parts of the plant,
guided by the leishmanicidal activity until we get three very active fractions. In
intracellular amastigotes, the active fractions MS??Ac and F7 showed IC50 of 1,69
??g/mL and 1,08 ??g/mL, respectively. This is the first report of the activity of
hidroxyethylpiperazines and Momordica charantia against Leishmania amazonensis.
The extracts, fractions and substances presented here can be considered potential
candidates for the development of new phytotherapy and/or chemotherapy against
cutaneous leishmaniasis
