Serological profile of chronically infected mice with the cystogenic ME-49 strain of Toxoplasma gondii before and after specific treatment

v. 38, n. 4, p. 261-266, out./dez. 2009Neste trabalho, foi avaliado o perfil sorológico experimental, após terapêutica específica, em camundongos Balb/c infectados com cepa cistogênica ME-49. Foram inoculados, individual e intraperitonealmente, 45 animais com dez cistos da cepa ME-49. Os camundongos foram divididos em três grupos de 15 animais e um grupo controle com cinco animais não infectados e não tratados, todos acompanhados ao mesmo tempo, diariamente, durante 20 dias. Após este período, foram subdivididos em dois grupos – A e B – e submetidos a esquemas terapêuticos específicos: grupo A – pirimetamina e ácido folínico; grupo B – pirimetamina, ácido folínico e sulfadiazina por dez dias consecutivos. A partir do quinto dia de tratamento, um camundongo de cada grupo foi anestesiado para a coleta de sangue por punção cardíaca, fazendo-se a análise do soro pela técnica de imunofluorescência indireta. Os animais tratados apresentaram diminuição significativa nos títulos de anticorpos (IgG) em relação ao grupo de infectados e não tratados. Entre as amostras analisadas com diferentes esquemas terapêuticos não houve diferença significativa. Isso mostra que o tratamento precoce da toxoplasmose reduz rapidamente os níveis de IgG, sugerindo uma possível cura desta parasitose. _______________________________________________________________________________________________________________________ ABSTRACT _______________________________________________________________________________________________________________________ This study evaluated the serological profile after experimental specific therapy of BALB / c mice infected with the cystogenic strain ME-49. Forty-five animals were inoculated individually, intraperitoneally with ten cysts of strain ME-49. Mice were divided into three groups of fifteen animals each, and a control group with five non-infected and non-treated animals. The infected animals and controls were simultaneously monitored daily for twenty days. After this period, they were divided into two groups: A and B and subjected to specific treatment regimens as follows: group A: pyrimethamine plus folic acid, group B: pyrimethamine, folinic acid and sulfadiazine for ten consecutive days. From the fifth day of treatment on, one mouse of each group was anesthetized and blood was collected by cardiac puncture; theserum was analyzed by the indirect immunofluorescence test. Treated animals showed a significant decrease in the titers of IgG antibodies, compared to the infected and untreated group. There was no significant difference among samples with different treatment regimes, showing that early treatment reduces rapidly the levels of IgG anti T. gondii and suggesting a possible cure of the infection
