    Designing and Evaluation of Skin Extract Agar for Isolation of Microflora from Raw Buffalo Hide

    Abstract Present study was aimed to design nutrient medium most suitable for isolation and enumeration of microbial flora associated with raw buffalo hide. Skin extract agar (SEA) was designed and standardized on the basis of its chemical analysis. SEA and nutrient medium supplemented with skin extract was inoculated with buffalo hide wash. Total viable count as well as diversity of microbial colonies were enumerated on SEA as well as on nutrient agar and standard plate count agar both supplemented with skin extract (1% v/v). Bacterial strains forming diverse types of colonies on the media tested were identified on the basis of their 16S rRNA  gene sequences. The SEA was found to yield higher number of bacteria and to support growth of Acinetobacter, Exiguobacterium and Stenotrophomonas which otherwise difficult to selectively isolate from buffalo hide using nutrient agar and standard plate count agar. Diversity of microbial colonies formed on SEA was significantly higher than that observed on nutrient agar or standard plate count agar. Feasibility of utilizing SEA as a microbiological medium for isolation and identification of microflora from raw buffalo hide was successfully demonstrated. Use of skin extract medium can maximize recovery of taxonomically distinct bacteria from raw buffalo hide. This basic study, with proper manipulations could lead to development of product for enumeration and isolation of bacteria from buffalo hides especially cattle pathogens related to skin diseases

    Protease production and enzymatic soaking of salt-preserved buffalo hides for leather processing

    Response surface methodological (RSM) optimization of protease by Pseudomonas aeruginosa MCM B327, increased 1.3-fold activity with 1% inoculum having cell density of 27.57 x 109 cells mL-1 at pH 7, 300C and 72 h of incubation. Protease enzyme recovered from P. aeruginosa showed characteristic activities against diverse proteins of hide. Enzyme was found to be active with substrates e.g. casein, Bovine serum albumin, gelatin, elastin, haemoglobin but inactive against keratin and collagen. During leather manufacturing, non-collagenase and non-keratinase activities have advantageous in a quality leather and hair saving process, respectively. Increased proteolytic enzyme concentration (0.1-0.5%) in soaking process showed increased water penetration because of hydrolysis of albumin and elastin proteins as indicated by opened fibers in histopathological sections. These findings suggest, protease secreted by P. aeruginosa may have application in soaking operation of leather processing for minimizing harmful deharing chemicals and processing time

    Effect of Various Nutritional Supplements on Hyaluronic Acid Production

    Effect of various nutritional supplementations on production of hyaluronic acid (HA) using Streptococcus equi subsp. zooepidemicus ATCC 39920 was evaluated at shake flask. Among the different carbon sources used, maltodextin showed HA of 0.9 g/L followed by 0.7 g/L for dextrose. L-arginine HCl at 2.0 g/L increased the yield of HA from 0.7 g/L to 0.85 g/L. L-arginine and L-glutamine showed the yield of HA of 0.7 g/L and 1.0 g/L, respectively at 2.0 g/L concentration. L-glutamine (2.0 g/L) and uridine (0.75 g/L) showed HA yield of 0.8 and 1.045 g/L, respectively. Addition of sodium glucuronate at 0.25 g/L concentration at 8 h, the yield of HA was increased from 0.7 g/L to 0.85 g/L. Pantothenic acid at concentration of 8.33 mg/L supplemented to the P10 medium, yield of 0.87 g/L of HA was obtained. Active dry yeast at 5.0 g/L increased yield of HA from 0.7 g/L to 0.85 g/L. Tween 80 at higher concentration reduced HA production. The addition of MgSO4.7H2O at 1.0 g/L improved the production of HA. Other metal ions like manganese, ferrous, copper, and zinc have no significant effect on HA production. Thus, it has been found that various nutritional supplements have effect on production of HA using S. equi subsp. zooepidemicus ATCC 39920

    It has been shown that initial conditions for bacterial cultivation are extremely important for the successful production of hyaluronic acid (HA) by fermentation. We investigated several physical parameters that affect productivity of HA under shake flask. i.e. transfer criteria of seed, agitation and aeration of fermentation flasks. Among the various physical parameters studied, inoculum age of 8-10 h, pH 6.4, optical density (600 nm) 2.0 and 3% level inoculum transfer found to be optimum. After inoculating with Streptococcus equi subsp. zooepidemicus ATCC 39920, the temperature 37 0C and 90 rpm found optimum during growth as well as for the HA production. The fractional factorial design of six factors with two levels showed yeast extract, potassium dihydrogen phosphate and sodium bicarbonate as significant model terms. The factor potassium dihydrogen phosphate was relatively more significant than yeast extract.

    Synergistically Active and Safe Fixed Dose Combination of Meropenem and Sulbactam

Emergence of meropenem resistance has been reported due to development of mutant plasmid mediated metallo-β-lactamases (IMP-6) and AmpC β–lactamases. Sulbactam, a stable β-lactamase inhibitor, increase antimicrobial activity of meropenem by inhibiting the enzyme β-lactamase. Fixed dose combination of Meropenem-sulbactam in the proportion of 1:1, 1:2, 1:3, 2:1 and 3:1 were evaluated for the antimicrobial activity. Combination of meropenem and sulbactam in the ratio of 2:1 exhibited the synergistic activity. This combination was checked for the subchronic toxicity on wistar rats and no change in biochemical and physiological parameters was observed

    Acupuncture is a very ancient form of healing which predates recorded history. The philosophy is rooted in the Taoist tradition which goes back over 8000 years. To date this practice although used world-wide to treat addictive behaviors has not been generally accepted or scientifically proven. There are however many positive reports in the literature in its effectiveness in reducing symptoms of withdrawal from alcohol, heroin and cocaine. In addition, due to known neuro-chemical mechanisms associated with acupuncture, especially its role in pain relief and endogenous opioid function, clinical benefits in reward dependence behaviors seem parsimonious. In this review article we are hypothesizing synergy between acupuncture/auriculotherapy and natural activation of mesolimbic dopaminergic pathways to reduce drug hunger and prevent relapse. Following a review of the literature involving both of these modalities the scientific and clinical community is encouraged to carry out translational research in large randomized double-blinded placebo controlled investigations coupling these two anti-craving treatment modalities

    Virtual Quantification of Protein Stability Using Applied Kinetic and Thermodynamic Parameters

    Protein stability, the most important aspect of molecular dynamics and simulations, requires sophisticated instrumentations of molecular biology to analyze its kinetic and thermodynamic background. Sequence- and structure-based programs on protein stability exist which relies only on single point mutations and sequence optimality. The energy distribution conferred by each hydrophobic amino acid in the protein essentially paves way for understanding its stability. To the best of our knowledge, Protein Stability is a first program of its kind, developed to explore the energy requirement of each amino acid in the protein sequence derived from various applied kinetic and thermodynamic quantities. The algorithm is strongly dependent both on kinetic quantities such as atomic solvation energies and solvent accessible surface area and thermodynamic quantities viz. enthalpy, entropy, heat capacity, etc. The hydrophobicity pattern of protein was considered as the important component of protein stabilization

    In Vivo Study of Nutritive Value of Oil Seeds from New Varieties of Sunflower, Safflower and Groundnut

    The nutritive value of new varieties of oil seeds has been assessed in a series of feeding trial with 34 days old 18 white rats supported moderate rat growth (male albino). The nutritive values of five new varieties of oil seeds, viz sunflower ( Helianths annuus L.) variety LSF -11 and LSF-8 , safflower (Carthamus tinctorius L.) variety PBNS-12 and PBNS-40, and  groundnut (Arachis hypogaea L.)  variety JL-24 were determined and compared in terms of the total feed intake, faeces voided, feed utilization, percent of feed utilization, nitrogen intake, nitrogen voided, nitrogen utilization, percent of nitrogen utilization, protein efficiency ratio and feed efficiency ratio per rat per day were reported in ranged between 6.56 to 8.03, 1.20 to 1.48, 5.26 to 6.55, 80.15 to 82.70, 0.25 to 0.32, 0.02 to 0.028, 0.23 to 0.30, 90.75 to 92.89, (+)1.37 to  (+)1.51 and (+)0.35 to (+)0.37  percent respectively. These new varieties of oil seeds were found non-toxic for rats and supported moderate rat growth.
