    On capillary viscosity measurements: how far do surface tension effects go?

    Viscosity is a fundamental thermophysical property of liquids making it very important particularly in the industry. Capillary viscometers have been widely used for viscosity measurements in different applications, the most relevant being the definition of viscosity standards, traceable to the primary water viscosity standard, by metrological institutions and industrial applications, mostly for quality control. Practical viscometry is based on the internationally accepted primary standard value for the kinematic viscosity of water at 20ºC and atmospheric pressure, which has been measured using capillary viscometers [1]. However, due to the water surface tension, viscosity measurements which have been related to water as a primary standard, can be significantly affected. It is difficult to rigorously assess the surface tension effects on capillary viscometers, and the practical way to avoid this problem is to use long capillaries, which are not appropriate for routine measurements [1-3]. After several experimental studies, using different types of viscometers, the usual procedure to correct surface tension effects in capillary viscosity measurements adopted by different authors, is to employ an empirical expression [1-4]. Additionally, other types of problems exist as the need to perform a kinetic energy correction which must also be taken into consideration [1]. The main goal of this work was to perform the calibration of a suspended-level, or Ubbelohde, capillary viscometer, which is not a long capillary viscometer, as well as the study of corrections to be used for the measurements performed with it. The experimental work covers the calibration of that Ubbelohde capillary viscometer, the evaluation of the uncertainty of the corresponding viscometer constant and the overall uncertainty of the measurements performed with it. This study includes the evaluation of the necessary corrections for kinetic energy and surface tension effects and, finally, the analysis of the case of a set of measurements performed with n-tetradecane. The ultimate purpose of this work is to obtain the lowest uncertainty for the Ubbelohde capillary viscometer 541 01/Ia, and to understand the need for the corrections that must be considered when using capillary viscometers and how they should be applied.N/

    Inbreeding levels in Northeast Brazil: Strategies for the prospecting of new genetic disorders

    A new autosomal recessive genetic condition, the SPOAN syndrome (an acronym for spastic paraplegia, optic atrophy and neuropathy syndrome), was recently discovered in an isolated region of the State of Rio Grande do Norte in Northeast Brazil, in a population that was identified by the IBGE (Brazilian Institute of Geography and Statistics) as belonging to the Brazilian communities with the highest rates of “deficiencies” (Neri, 2003), a term used to describe diseases, malformations, and handicaps in general. This prompted us to conduct a study of consanguinity levels in five of its municipal districts by directly interviewing their inhabitants. Information on 7,639 couples (corresponding to about 40% of the whole population of the studied districts) was obtained. The research disclosed the existence of very high frequencies of consanguineous marriages, which varied from about 9% to 32%, suggesting the presence of a direct association between genetic diseases such as the SPOAN syndrome, genetic drift and inbreeding levels. This fact calls for the introduction of educational programs for the local populations, as well as for further studies aiming to identify and characterize other genetic conditions. Epidemiological strategies developed to collect inbreeding data, with the collaboration of health systems available in the region, might be very successful in the prospecting of genetic disorders

    Application of flow cytometry for the identification of Staphylococcus epidermidis by peptide nucleic acid fluorescence in situ hybridization (PNA FISH) in blood samples

    Staphylococcus epidermidis is considered to be one of the most common causes of nosocomial bloodstream infections, particularly in immune-compromised individuals. Here, we report the development and application of a novel peptide nucleic acid probe for the specific detection of S. epidermidis by fluorescence in situ hybridization. The theoretical estimates of probe matching specificity and sensitivity were 89 and 87%, respectively. More importantly, the probe was shown not to hybridize with closely related species such as Staphylococcus aureus. The method was subsequently successfully adapted for the detection of S. epidermidis in mixed-species blood cultures both by microscopy and flow cytometry.This work was supported by the Portuguese Institute Fundacao para a Ciencia e Tecnologia (PhD Fellowship SFRH/BD/29297/2006 and Post-Doc Fellowship SFRH/BPD/42208/2007)

    Spectroscopic variability of two Oe stars

    The Oe stars HD45314 and HD60848 have recently been found to exhibit very different X-ray properties: whilst HD60848 has an X-ray spectrum and emission level typical of most OB stars, HD45314 features a much harder and brighter X-ray emission, making it a so-called gamma Cas analogue. Monitoring the optical spectra could provide hints towards the origin of these very different behaviours. We analyse a large set of spectroscopic observations of HD45314 and HD60848, extending over 20 years. We further attempt to fit the H-alpha line profiles of both stars with a simple model of emission line formation in a Keplerian disk. Strong variations in the strengths of the H-alpha, H-beta, and He I 5876 emission lines are observed for both stars. In the case of HD60848, we find a time lag between the variations in the equivalent widths of these lines. The emission lines are double peaked with nearly identical strengths of the violet and red peaks. The H-alpha profile of this star can be successfully reproduced by our model of a disk seen under an inclination of 30 degrees. In the case of HD45314, the emission lines are highly asymmetric and display strong line profile variations. We find a major change in behaviour between the 2002 outburst and the one observed in 2013. This concerns both the relationship between the equivalent widths of the various lines and their morphologies at maximum strength (double-peaked in 2002 versus single-peaked in 2013). Our simple disk model fails to reproduce the observed H-alpha line profiles of HD45314. Our results further support the interpretation that Oe stars do have decretion disks similar to those of Be stars. Whilst the emission lines of HD60848 are explained by a disk with a Keplerian velocity field, the disk of HD45314 seems to have a significantly more complex velocity field that could be related to the phenomenon that produces its peculiar X-ray emission.Comment: Accepted for Publication in A&

    In search of gravity mode signatures in main sequence solar-type stars observed by Kepler

    Gravity modes (g modes), mixed gravito-acoustic modes (mixed modes), and gravito-inertial modes (gi modes) possess unmatched properties as probes for stars with radiative interiors. The structural and dynamical constraints that they are able to provide cannot be accessed by other means. While they provide precious insights into the internal dynamics of evolved stars as well as massive and intermediate-mass stars, their non-detection in main sequence (MS) solar-type stars make them a crucial missing piece in our understanding of angular momentum transport in radiative zones and stellar rotational evolution. In this work, we aim to apply certain analysis tools originally developed for helioseismology in order to look for g-mode signatures in MS solar-type stars. We select a sample of the 34 most promising MS solar-type stars with Kepler four-year long photometric time series. All these stars are well-characterised late F-type stars with thin convective envelopes, fast convective flows, and stochastically excited acoustic modes (p modes). For each star, we compute the background noise level of the Fourier power spectrum to identify significant peaks at low frequency. After successfully detecting individual peaks in 12 targets, we further analyse four of them and observe distinct patterns of surrounding peaks with a low probability of being noise artifacts. Comparisons with the predictions from reference models suggest that these patterns are compatible with the presence of non-asymptotic low-order pure g modes, pure p modes, and mixed modes. Given their sensitivity to both the convective core interface stratification and the coupling between p- and g-mode resonant cavities, such modes are able to provide strong constraints on the structure and evolutionary states of the related targets.     Deciphering Adipose Tissue Extracellular Vesicles Protein Cargo and Its Role in Obesity

    The extracellular vesicles (EVs) have emerged as key players in metabolic disorders rising as an alternative way of paracrine/endocrine communication. In particular, in relation to adipose tissue (AT) secreted EVs, the current knowledge about its composition and function is still very limited. Nevertheless, those vesicles have been lately suggested as key players in AT communication at local level, and also with other metabolic peripheral and central organs participating in physiological homoeostasis, and also contributing to the metabolic deregulation related to obesity, diabetes, and associated comorbidities. The aim of this review is to summarize the most relevant data around the EVs secreted by adipose tissue, and especially in the context of obesity, focusing in its protein cargo. The description of the most frequent proteins identified in EVs shed by AT and its components, including their changes under pathological status, will give the reader a whole picture about the membrane/antigens, and intracellular proteins known so far, in an attempt to elucidate functional roles, and also suggesting biomarkers and new paths of therapeutic action
