    The evolution of the spatial and sectoral patterns in Ile-de-France over 1978-1997

    The aim of this paper is to analyze the intra-urban spatial distribution of employment in the agglomeration of Ile-de-France in 1978 and 1997. In that purpose, exploratory spatial data analysis is used in order to identify employment centers and a sectoral analysis of the CBD and the subcenters is performed. Our results highlight a suburbanization process of employment between 1978 and 1997 in Ile-de-France. A more polarized space emerges in 1997 compared to 1978 with several employment centers specialized in different activities. Moreover, even if the spatial influence of the CBD is diminishing during the study period, the CBD preserves its economic leadership by concentrating a large variety of high-order producer services.     A Novel 3-Methyladenine DNA Glycosylase from Helicobacter pylori Defines a New Class within the Endonuclease III Family of Base Excision Repair Glycosylases

    The cloning, purification, and characterization of MagIII, a 3-methyladenine DNA glycosylase from Helicobacter pylori, is presented in this paper. Sequence analysis of the genome of this pathogen failed to identify open reading frames potentially coding for proteins with a 3-methyladenine DNA glycosylase activity. The putative product of the HP602 open reading frame, reported as an endonuclease III, shares extensive amino acid sequence homology with some bacterial members of this family and has the canonic active site helix-hairpin-helix-GPD motif. Surprisingly, this predicted H. pylori endonuclease III encodes a 25,220-Da protein able to release 3-methyladenine, but not oxidized bases, from modified DNA. MagIII has no abasic site lyase activity and displays the substrate specificity of the 3-methyladenine-DNA glycosylase type I of Escherichia coli (Tag) because it is not able to recognize 7-methylguanine or hypoxanthine as substrates. The expression of the magIII open reading frame in null 3-methyladenine glycosylase E. coli (tag alkA) restores to this mutant partial resistance to alkylating agents. MagIII-deficient H. pylori cells show an alkylation-sensitive phenotype. H. pylori wild type cells exposed to alkylating agents present an adaptive response by inducing the expression of magIII. MagIII is thus a novel bacterial member of the endonuclease III family, which displays biochemical properties not described for any of the members of this group until now.Fil: O'Rourke, Eyleen J..     Development of a locus-specific, co-dominant SCAR marker for assisted-selection of the Sw-5 (Tospovirus resistance) gene cluster in a wide range of tomato accessions

    The best levels of broad-spectrum Tospovirus resistance reported in tomatoes thus far are conferred by the Sw-5 locus. This locus contains at least five paralogues (denoted Sw-5a through Sw-5e), of which Sw-5b represents the actual resistance gene. Here we evaluated a panel of seven PCR primer pairs matching different sequences within a genomic region spanning the Sw-5a and Sw-5b gene cluster. Primer efficiency evaluation was done employing tomato isolines with and without the Sw-5 locus. One primer pair produced a single and co-dominant polymorphism between susceptible and resistant isolines. Sequence analysis of these amplicons indicated that they were specific for the Sw-5 locus and their differences were due to insertions/deletions. The polymorphic SCAR amplicon encompass a conserved sequence of the promoter region of the functional Sw-5b gene, being located in the position -31 from its open reading frame. This primer pair was also evaluated in field assays and with a collection of accessions known to be either susceptible or resistant to tospoviruses. An almost complete correlation was found between resistance under greenhouse/field conditions and the presence of the marker. Therefore, this primer pair is a very useful tool in marker-assisted selection systems in a large range of tomato accessions

    Control sustentable poscosecha: extractos de Larrea cuneifolia mediados por nades frente a Botrytis cinerea

    Botrytis cinerea es un hongo ubicuo que ocasiona la podredumbre gris, una de las principales enfermedades de fruta en poscosecha, siendo responsable de pérdidas económicas. Debido a la creciente preocupación por los efectos adversos del uso de pesticidas, la búsqueda de alternativas se presenta como una meta prioritaria. En este contexto, los extractos de plantas representan una rica fuente de biocompuestos con atractivas propiedades antimicrobianas. Recientemente, los solventes eutécticos naturales (NADES) se han propuesto como agentes extractantes sustentables de compuestos bioactivos a partir de plantas. En el presente estudio, se evaluó un bioextracto de L. cuneifolia basado en NADES hacia B. cinerea. Para este propósito, se usó un NADES compuesto por ácido láctico, glucosa y agua (LGH) como agente de extracción y se comparó con solventes tradicionales en términos de capacidad antioxidante y contenido fenólico total. Además, la actividad antimicrobiana del bioextracto se evaluó in vitro e in vivo en uvas inoculadas artificialmente. A una concentración del 2% el bioextracto fue capaz de inhibir el crecimiento miceliar de B. cinerea en un 92%. Interesantemente, L. cuneifolia mostró un excelente rendimiento para el control de la podredumbre gris en uvas, demostrando su potencial como alternativa sustentable a los fungicidas sintéticos.Botrytis cinerea is a ubiquitous fungus causing gray mold, the main postharvest disease in fruit, which implies important economic losses in agriculture. With growing concern over health and environmental effects of pesticides, the search for eco-friendly alternatives is a clear priority. Plant extracts represent a rich source of biocompounds with attractive antimicrobial properties. In the last decade, Natural Deep Eutectic Solvents (NADES) has emerged as an auspicious green extraction media to achieve bioextract for a sustainable postharvest control. In the present study, a novel L. cuneifolia NADES-based bioextract was evaluated against B. cinerea. To this purpose, a NADES composed by lactic acid, glucose and water (LGH) was used as extracting agent and compared with traditional solvents in terms of antioxidant capacity and total phenolic content. Furthermore, the bioextract antifungal activity was tested in vitro and also in vivo on artificially inoculated grapes, in order to obtain preliminary data about the efficacy on gray mold development. The antimicrobial activity of the bioextract was assessed using agar diffusion method against B. cinerea, inhibition of 92% was achieved with the bioextract at 2%. Notably, L. cuneifolia bioextract showed an excellent performance for gray mold control on grapes, supporting their potential as alter-native green fungicide.Fil: Boiteux, Joana Jaqueline.
