    Molecular characterization of a mosaic locus in the genome of 'Candidatus Liberibacter asiaticus'

    <p>Abstract</p> <p>Background</p> <p>Huanglongbing (HLB) is a highly destructive disease of citrus production worldwide. '<it>Candidatus </it>Liberibacter asiaticus', an unculturable alpha proteobacterium, is a putative pathogen of HLB. Information about the biology and strain diversity of '<it>Ca</it>. L. asiaticus' is currently limited, inhibiting the scope of HLB research and control.</p> <p>Results</p> <p>A genomic region (CLIBASIA_05640 to CLIBASIA_05650) of '<it>Ca</it>. L. asiaticus' showing hyper-sequence variation or locus mosaicism was identified and investigated using 262 bacterial strains (188 from China and 74 from Florida). Based on the characteristic electrophoretic profiles of PCR amplicons generated by a specific primer set, eight electrophoretic types (E-types) were identified, six E-types (A, B, C, D, E, and F) in China and four E-types (A, C, G, and H) in Florida. The '<it>Ca</it>. L. asiaticus' strains from China consisted predominately of E-type A (71.3%) and E-type B (19.7%). In contrast, the '<it>Ca</it>. L. asiaticus' strains from Florida was predominated by E-type G (82.4%). Diversity of '<it>Ca</it>. L. asiaticus' in China was also evidenced. Strains from the high altitude Yunnan Province consisted of five E-types with E-type B being the majority (62.8%), whereas strains from the low altitude coastal Guangdong Province consisted of only two E-types with E-type A as the majority (97.0%). Sequence analyses revealed that variation of DNA amplicons was due to insertion/deletion events at CLIBASIA_05650 and the downstream intergenic region.</p> <p>Conclusions</p> <p>This study demonstrated the genomic mosaicism of '<it>Ca</it>. L. asiaticus' resulted from active DNA insertion/deletion activities. Analyses of strain variation depicted the significant inter- and intra-continent diversity of '<it>Ca</it>. L. asiaticus'.</p

    Advancements in the Fenton Process for Wastewater Treatment

    Fenton is considered to be one of the most effective advanced treatment processes in the removal of many hazardous organic pollutants from refractory/toxic wastewater. It has many advantages, but drawbacks are significant such as a strong acid environment, the cost of reagents consumption, and the large production of ferric sludge, which limits Fentonā€™s further application. The development of Fenton applications is mainly achieved by improving oxidation efficiency and reducing sludge production. This chapter presents a review on fundamentals and applications of conventional Fenton, leading advanced technologies in the Fenton process, and reuse methods of iron containing sludge to synthetic and real wastewaters are discussed. Finally, future trends and some guidelines for Fenton processes are given

    Discovery and Survey of a New Mandarivirus Associated with Leaf Yellow Mottle Disease of Citrus in Pakistan.

    During biological indexing for viruses in citrus trees, in a collection of Symons sweet orange (SSO) (Citrus sinensis L. Osbeck) graft inoculated with bark tissues of citrus trees from the Punjab Province in Pakistan, several SSO trees exhibited leaf symptoms of vein yellowing and mottle. High-throughput sequencing by Illumina of RNA preparation depleted of ribosomal RNAs from one symptomatic tree, followed by BLAST analyses, allowed identification of a novel virus, tentatively named citrus yellow mottle-associated virus (CiYMaV). Genome features of CiYMaV are typical of members of the genus Mandarivirus (family Alphaflexiviridae). Virus particles with elongated flexuous shape and size resembling those of mandariviruses were observed by transmission electron microscopy. The proteins encoded by CiYMaV share high sequence identity, conserved motifs, and phylogenetic relationships with the corresponding proteins encoded by Indian citrus ringspot virus (ICRSV) and citrus yellow vein clearing virus (CYVCV), the two current members of the genus Mandarivirus. Although CYVCV is the virus most closely related to CiYMaV, the two viruses can be serologically and biologically discriminated from each other. A reverse-transcription PCR method designed to specifically detect CiYMaV revealed high prevalence (62%) of this virus in 120 citrus trees from the Punjab Province, Pakistan, where the novel virus was found mainly in mixed infection with CYVCV and citrus tristeza virus. However, a preliminary survey on samples from 200 citrus trees from the Yunnan Province, China failed to detect CiYMaV in this region, suggesting that the molecular, serological, and biological data provided here are timely and can help to prevent the spread of this virus in citrus-producing countries

    Quantitative Imaging of Single, Unstained Viruses with Coherent X-rays

    Since Perutz, Kendrew and colleagues unveiled the structure of hemoglobin and myoglobin based on X-ray diffraction analysis in the 1950s, X-ray crystallography has become the primary methodology used to determine the 3D structure of macromolecules. However, biological specimens such as cells, organelles, viruses and many important macromolecules are difficult or impossible to crystallize, and hence their structures are not accessible by crystallography. Here we report, for the first time, the recording and reconstruction of X-ray diffraction patterns from single, unstained viruses. The structure of the viral capsid inside a virion was visualized. This work opens the door for quantitative X-ray imaging of a broad range of specimens from protein machineries, viruses and organelles to whole cells. Moreover, our experiment is directly transferable to the use of X-ray free electron lasers, and represents a major experimental milestone towards the X-ray imaging of single macromolecules.Comment: 16 pages, 5 figure

    Catalytic Ozone Oxidation of Petrochemical Secondary Effluent: Mechanism, Application and Future Development

    Petrochemical secondary effluent contains toxic and refractory organic compounds, which are difficult to be further treated by traditional biological process. In China, most of the advanced treatment units have been built recently by catalytic ozone oxidation process to achieve the high-quality effluent. In this chapter, the mechanism and reaction process of catalytic ozone oxidation of petrochemical secondary effluent will be introduced in detail. With the operation of the catalytic ozone oxidation tank, a series of problems which are not taken into account at the beginning of the design have arisen. The chapter will talk about the problems concerning the biological flocs, colloidal macromolecule organic compounds, ozone mass transfer, and catalysts based on practical applications. In the last part of the chapter, the development trends of catalytic ozone oxidation of petrochemical secondary effluent will also be discussed

    Establishment of the Luoping Biota National Geopark in Yunnan, China

    Geoparks in China have been a great success story, with 284 national geoparks and 41 of them accorded UNESCO international status, the highest number for any country in the world. We track the progress of one of the geoparks, Luoping Biota National Geopark in Yunnan Province, from initial plans after its discovery as a key site for the exceptional preservation of Middle Triassic marine fossils in 2007, to acceptance as a National Geopark in 2011. Geoparks combine great scientific importance with accessibility and attraction for tourists. The scientific importance of Luoping is in the fossils, thousands of specimens of marine invertebrates, fishes and reptiles, together with rare elements from land (e.g. insects, plants), representing an important phase in the Mesozoic Marine Revolution, when life was recovering from devastation at the end of the Permian, and 8 million years later, had developed stable ecosystems with a new structure, dominated by predatory fishes and reptiles. The touristic importance of the Luoping Biota Geopark has already been demonstrated by rapid development of facilities and high visitor numbers

    A Sec-dependent effector, CLIBASIA_04425, contributes to virulence in ā€˜Candidatus Liberibater asiaticusā€™

    Citrus Huanglongbing (HLB) is the most destructive citrus disease worldwide, mainly caused by ā€˜Candidatus Liberibacter asiaticusā€™ (CLas). It encodes a large number of Sec-dependent effectors that contribute to HLB progression. In this study, an elicitor triggering ROS burst and cell death in Nicotiana benthamiana, CLIBASIA_04425 (CLas4425), was identified. Of particular interest, its cell death-inducing activity is associated with its subcellular localization and the cytoplasmic receptor Botrytis-induced kinase 1 (BIK1). Compared with CLas infected psyllids, CLas4425 showed higher expression level in planta. The transient expression of CLas4425 in N. benthamiana and its overexpression in Citrus sinensis enhanced plant susceptibility to Pseudomonas syringae pv. tomato DC3000 Ī”hopQ1-1 and CLas, respectively. Furthermore, the salicylic acid (SA) level along with the expression of genes NPR1/EDS1/NDR1/PRs in SA signal transduction was repressed in CLas4425 transgenic citrus plants. Taken together, CLas4425 is a virulence factor that promotes CLas proliferation, likely by interfering with SA-mediated plant immunity. The results obtained facilitate our understanding of CLas pathogenesis
