Malaria continues to be a global health burden and one of the most common infectious diseases worldwide. Treatment consists of artemisinin-based combination regimens that utilize pharmacokinetic principles to ensure optimal efficacy. Many pharmacokinetic considerations exist for these agents, including use in special populations and coadministration with agents prone to drug–drug interactions. This review provides an overview of pharmacokinetic considerations of artemisinin-based combinations. Key topics discussed include pharmacokinetic rationale for combinations of artemisinin agents with partner drugs, implications for dosing considerations with children, pharmacokinetic changes associated with pregnancy and known clinical pharmacokinetic interactions between antimalarials and other commonly used medications.</jats:p
