Salicylic acid (SA) also induces stomatal closure accompanied by peroxidase-mediated extracellular ROS production and intracellular ROS accumulation but not by [Ca2l叩 osciIlation，while yeast elicitor (YEL) and chitosan (CHT) induce stomatal closure accompani凶 byperoxidase-mediated extracellular ROS production， intracellular ROS accumulation， and [Ca2+]cyI oscillation. Mitogen・activatedprotein kinases (MAPKs)， MPK9叩 dMPK12， redundantly function as positive regulators downstream of ROS production and [Ca2l叩 osci1lationin ABA signaling pathwaドHowever，it remains to be cIarified whether MPK9 and MPKl2 function in MeJA， SA， YEし叩dCHT signaling in guard cels. 1 investigated MeJA-， SA-， YEL・， and CH下inducedstomatal closure using Arabidopsis wild type， ecotype Columbia-erecta (Col-er)， mpk9， mpk12， mpk9 mpk12 and the恥1APKkinase (MAPKK)凶libitors，PD98059 and U0126. Production of intracellular ROS， extracellular ROS， and alkalization were measured using 2'，7'-dichlorodihydrofluorescein diacetate (H2DCF・DA)，3，3' -diaminobenzidine tetrahydrochloride hydrate (DAB)， mtro blue tetrazolium (NBT)， 叩 d2'，7'・bis-(2・carboxyethyl)・5，(6)-carboxyfluoresceinacetoxymethyl(BCECF-AM)， respectively. The wild-type血 dmpk9 mpk12 plants expressing yellow cameleon 3.6 were used for the measurement of guard cel [Ca2+]c戸 oscillation.S-type anion channel cu汀entswere measured using a whole-cell patch-clamp technique. MeJA-， SA-， and CHT-induced stomatal cIosure in wild type were inhibited by PD98059 and were impaired in mpk9 mpk12 but not in mpk9 or mpk12. MeJA， SA， YEL， and CHT induced ROS production and cytosolic alkalization in al mutants and wild type. MeJA， YEL，加dCHT induced [Ca2+]cyt oscillation in wild type and mpk9 mpk12. MeJA induced activation of S-type anion channel in wild type but not in mpk9 mpk12. SA did not induce [Ca2+]cyI oscillation in either wild type or mpk9 mpk12. YEL・inducedstomatal cIosur，e inwild type was inhibited by PD98059 and U0126 and was impaired in mpk9 mpk12 but not in mpk9 or mpk12. mpk12. YEL inhibited light-induced stomatal opening in wild type but not in mpk9 mpk12. Inhibition of Iight-induced stomatal opening by YEL was inhibited by PD98059 and UOl26 in wild type. In conclusion， MPK9 and MPK12 redundantly function downstre創nof ROS production and cytosolic alkalization in MeJA， SA， YEL， and CHT signaling in Arabidopsis guard cels. 論文審査結果の要旨本論文は、陸上植物にとって重要な気孔閉口の機構における MAPキナーゼの関与を明らかにしようとしたものである。ジャスモン酸メチルが誘導する気孔閉口において、孔辺細胞に発現する MAPキナー ゼ(加1PK9、MPK12)がジャスモン酸メチルシグナル伝達経路の活性酸素種産生や細胞内カルシウム変動、細胞内アルカリ化の下流で機能していることを明らかにした。また、 MAPキナーゼキナーゼの阻害剤によって、ジャスモン酸メチル誘導気孔閉口が阻害されたことより、ジャスモン酸メチル信号伝達経路に MAPキナーゼカスケードが関与していることを明らかにした。傷害や菌感染に深く関与するサリチル酸、酵母エリシタ一、キトサンが誘導する気孔閉口においても、その信号伝達経路において MPK9と恥伊K12が、活性酸素種産生や細胞内カルシウム変動、細胞内アルカリ化の下流で機能していることを明らかにした。以上の結果から、ジャスモン酸メチル、サリチル酸、酵母エリシタ一、キトサンが誘導する気孔閉口には、 MAPキナーゼ (MPK9とMPK12)が信号伝達因子として関与していることを明らかにした。本研究内容は、学術的な価値のみならず、気孔運動に着目した生産制御のための技術の基礎となるものである。従って、本審査委員会は本論文が博士(学術)の学位論文に値すると判断した。
