Florian Kaiser: Studien zur Totalsynthese von Mumbaistatin und anderen komplexenBenzophenonen als potentielle Antidiabetes-WirkstoffeDie vorliegende Arbeit beschreibt die Erschließung von Synthesestrategien zur Darstellung vonmedizinisch interessanten aromatischen Naturstoffen mit hochsubstituiertem Benzophenon-Gerüst.Zielstrukturen waren das neuartige Antibiotikum Pestalon und vor allem das Anthrachinon-PolyketidMumbaistatin, das als stärkster bisher gefundener natürlicher Glucose-6- PhosphatTranslocase-Inhibitor eine Leitstruktur für neue Antidiabetes- Wirkstoffe darstellt.In einer hochkonvergenten Synthese wurde zunächst ein Mumbaistatin-Analogon hergestellt, dem dieBenzoesäure-Funktionalität des Naturstoffes fehlte. Der Aufbau des Anthrachinon-Gerüsts erfolgtedurch eine Arin-Phthalid- Anellierungsreaktion. Die zentrale Benzophenon-Struktur wurde durch einenucleophile Kupplung eines Aryllithium-Dianions mit einem Anthracenaldehyd und nachfolgenderMulti-Oxidation aufgebaut.Die weiteren Untersuchungen konzentrierten sich auf die Synthese von 3- Deoxymumbaistatin unterAusnutzung der entwickelten Strategie. Die benötigten aromatischen Synthesebausteine wurden u. a.durch selektive Bromierungs- und ortho-Lithiierungs-Reaktionen dargestellt. Für den Aufbau derhochsubstituierten Anthrachinon-Teilstruktur wurden unterschiedliche Zugänge evaluiert, darunterverschiedene Arin-Phthalid-Anellierungen und eine anionische Cyclisierung.Die zentrale CC-Verknüpfung zum tetra-ortho-substituierten Benzophenon, die zuvor anModellsystemen optimiert worden war, konnte aufgrund der unerwarteten Instabilität der beteiligtenVerbindungen nicht erreicht werden.In einem separaten Teil der Arbeit wurde ein erster Zugang zu dem marinen Naturstoff Pestalonuntersucht. Ausgehend von einfachen Resorcin-Derivaten wurden die beiden aromatischenMolekülteile zunächst durch Halogenierungs- und Alkylierungsreaktionen funktionalisiert und dannwiederum durch eine nucleophile Kupplung einer Aryllithiumverbindung an einen Benzaldehyd undnachfolgende Oxidation zum Benzophenon verknüpft. So konnte Deformyl- Pestalon in 10 Stufen undhoher Gesamtausbeute synthetisiert werden. Die erarbeiteten Synthesesequenzen und -Strategiensollten sich in Zukunft durch Weiterentwicklung und Optimierung auch zur Darstellung der beidenNaturstoffe selbst nutzen lassen. This work describes the development of synthetic strategies for the preparation of new aromaticnatural products with highly substituted, sterically hindered benzophenone scaffolds which showedinteresting medicinal properties. The target structures were the marine antibiotic pestalone andparticularly the anthraquinone polyketide mumbaistatin. As the strongest naturally occurring inhibitorof the enzyme glucose-6- phosphate translocase, mumbaistatin represents a lead structure for thedevelopment of new antidiabetic drugs.In a highly convergent approach a close mumbaistatin analog was synthesized which lacks the benzoicacid functionality of the natural product. The anthraquinone scaffold was prepared utilizing anaryne-phthalide annulation reaction. The elaboration of the central benzophenone structure wasachieved via nucleophilic coupling of a lithiated arene dianion with an anthracene carbaldehydefollowed by multiple oxidation.The following studies concentrated on the synthesis of 3-deoxymumbaistatin applying the developedstrategies. The preparation of the functionalized arene intermediates was achieved exploiting highlyregioselective bromination and ortho-lithiation reactions. For the construction of the denselysubstituted anthraquinone core various approaches were evaluated, among them differentaryne-phthalide-annulations and an anionic cyclization. The key connection of the building blocks tothe tetra-ortho substituted benzophenone, which was previously optimized on model substrates, couldnot be achieved due to the unexpected instability of the involved compounds.In a separate part of the thesis a first synthetic access towards the marine natural product pestalone waselaborated. Starting from simple resorcinol derivatives, the aromatic building blocks were firstfunctionalized by halogenation- and alkylation reactions and then again connected to a benzophenonestructure by nucleophilic coupling followed by oxidation. In this manner, the derivativedeformyl-pestalone was prepared in 10 steps and high overall yield.In the future, it should be possible to optimize and further develop the elaborated synthetic sequencesand strategies for the preparation of the natural products themself.
