Answers all parts of this question.   (a) Discuss the necessity to sterilize fermentation medium. What are the available methods for media sterilization?  (10 marks)  (b) Table 1 (Appended) shows the Del values for Bacillus stearothermophilus spores during the heating process for sterilization. Assume that the spore destruction only occurs at temperatures beyond 100 ºC and that the heating and cooling cycle is linear. For a medium containing 1012 cells that was used, the overall Del value was 35.5.   Calculate the holding time at  121 ºC for the sterilization process where the medium has been heated from 100 ºC to 121 ºC in 25 minutes and cooled from  121 ºC to 100 ºC in 15 minutes.            (7 marks) (c) If the operation was scaled up from 1000 litres to 10000 litres using the same medium, what is the new Del value?                (3 marks)2. Write an essay on the use and production of bioethanol. (20 marks)3. Discuss how the principles of biotechnology can be applied to improve the production process of local fermented food. Use appropriate examples to illustrate your answer.                   (20 marks)4. Answers all parts of this question.   (a) Explain the preparation of inoculum for a 120,000 litre bacterial fermentation. (10 marks) (b) You have been asked to formulate a medium for the production of single cell protein from Candida utilis with a yield of at least 10g /l, using a continuous stirred tank reactor (CSTR) operated batch-wise. The carbon sources available are glucose (Cell yield coefficient of 0.54) and n-alkane (Cell yield coefficient of 1.03).    (i) What is your choice (glucose or n-alkane) as carbon source? Why? (4 marks)  (iii) If you are using glucose as the carbon source, will you add it in all at once into the fermenter/medium at the start of the fermentation? Explain your answer . (3 marks)5. Jawab EMPAT soalan sahaja.  1. Jawab semua bahagian soalan ini.   (a) Bincangkan kepentingan untuk mensterilkan medium fermentasi. Apakah kaedah-kaedah yang terdapat untuk pensterilan medium?    (10 markah)  (b) Jadual 1 (dilampirkan) menunjukkan nilai Del untuk spora Bacillus stearothermophilus semasa proses pemanasan untuk pensterilan.  Andaikan kemusnahan spora hanya berlaku pada suhu melebihi 100 ºC dan kitaran proses pemanasan dan penyejukan adalah linear. Untuk medium yang diguna yang mengandungi bilangan sel sebanyak 1012, nilai Del keseluruhan proses ialah 35.5.   Kirakan masa penahanan pada suhu 121 ºC untuk satu proses pensterilan  yang  mana  medium   tersebut  telah  dipanaskan  dari  100 ºC  ke 121 ºC dalam masa 25 minit dan disejukkan dari 121 ºC ke 100 ºC dalam masa 15 minit.             (7 markah) (c) Sekiranya operasi tersebut dibesarkan daripada 1000 liter kepada 10000 liter dengan menggunakan medium yang sama, apakah nilai Del yang baru?      (3 markah)2. Tuliskan satu karangan mengenai penggunaan dan penghasilan bioethanol.   (20 markah)3. Bincangkan  bagaimana prinsip bioteknologi  dapat diaplikasi untuk meningkatkan proses penghasilan makanan terfermen tempatan. Berikan contoh-contoh yang sesuai untuk menjelaskan jawapan anda.                             (20 markah)4. Jawab semua bahagian soalan ini.    (a) Terangkan cara penyediaan inokulum untuk sesuatu fermentasi bakteria berskala 120,000 liter. Sumber karbon yang tersedia adalah glukosa (Pekali yil sel = 0.54) dan n-alkane (Pekali yil sel = 1.03).     (i) Apakah pilihan (glukosa atau n-alkane) yang akan anda guna sebagai sumber karbon? Kenapa?           (3 markah)  (ii) Berapa banyakkah sumber karbon yang diperlukan untuk mencapai sasaran hasil dalam fermentasi 100 liter?  (4 markah)  (iii) Sekiranya anda menggunakan glukosa, adakah anda akan memasukkan kesemua glukosa ini kedalam fermenter sekali gus pada permulaan fermentasi?  Huraikan jawapan anda.
