Studies on the Properties and the Activity Regulation of N-Acetyl-β-D-glucosaminidase from Green Crab (Scylla serrata)

Abstract

以锯缘青蟹(Scyllaserrata)内脏为材料,通过含0.2mol/LNaCl的0.01mol/LTris-HCl(pH7.5)缓冲液抽提、硫酸铵分级沉淀、SephadexG-100凝胶柱过滤层析、DE-32离子交换柱层析,获得了比活力为7,990U/mg的电泳单一纯N-乙酰-β-D-氨基葡萄糖苷酶制剂(NAGase,EC3.2.1.52)。该酶的等电点为4.68,分子量约为132kD,每个酶蛋白分子含有约1097个氨基酸残基,其中非极性占39.1%,碱性占7.1%,酸性占24.0%,极性不带电荷的占29.8%。该酶含两个分子量约为65.8kD的同型亚基,肽链间不含二硫键而肽链内含有二硫键...N-Acetyl-β--D-glucosaminidase (NAGase, EC 3.2.1.52) was purified from viscera of green crab (Scylla serrata), by extraction with 0.01 mol/L Tris-HCl buffer (pH 7.5) containing 0.2 mol/L NaCl and ammonium sulfate fractionation, then chromatography on Sephadex G-100 and DEAE-cellulose (DE-32).
