Studies on the Differentiation of Mouse Erythroleukemia (MEL) Cells Induced by Erythroid Differentiantion Factor ( EDF)

Abstract

本文研究EDF的分离纯化方法及其对MEL细胞的诱导分化。 以哺乳类动物(兔)为材料,按费氏法分离纯化EDF,即给新西兰兔 注射盐酸苯肼致贫血,收集网织红细胞,用冻融法裂解细胞,乙醇、 氯仿变性除去血红蛋白,过SephadexG-100,超滤浓缩后再过 DEAE——纤维素和CM——纤维素柱,获得纯化30,000倍的活性物质EDF, SDS-PAGE电泳为单一蛋白带,分子量15KD,产率9.6%。另将 过sephadexG-100后的活性浓缩法直接过CM-柱,也得到活性组 分,其结果与费氏法相似。或将上述浓缩液直接等电聚焦层析;也得 到相近的结果。 观察EDF(纯品)对MEL细胞...This research includes two parts: 1) the isolation and purification of EDF and its method improvement; 2) the differentiation of MEL cells treated by EDF.
