    Leveraging upon standards to build the Internet of things

    Smart embedded objects will become an important part of what is called the Internet of Things. However, the integration of embedded devices into the Internet introduces several challenges, since many of the existing Internet technologies and protocols were not designed for this class of devices. In the past few years, there were many efforts to enable the extension of Internet technologies to constrained devices. Initially, this resulted in proprietary protocols and architectures. Later, the integration of constrained devices into the Internet was embraced by IETF, moving towards standardized IP-based protocols. Long time, most efforts were focusing on the networking layer. More recently, the IETF CoRE working group started working on an embedded counterpart of HTTP, allowing the integration of constrained devices into existing service networks. In this paper, we will briefly review the history of integrating constrained devices into the Internet, with a prime focus on the IETF standardization work in the ROLL and CoRE working groups. This is further complemented with some research results that illustrate how these novel technologies can be extended or used to tackle other problems.The research leading to these results has received funding from the European Union's Seventh Framework Programme (FP7/2 007-2013) under grant agreement n°258885 (SPITFIRE project), from the iMinds ICON projects GreenWeCan and O’CareCloudS, and a VLI R PhD scholarship to Isam Ishaq

    p185(BCR/ABL) has a lower sensitivity than p210(BCR/ABL) to the allosteric inhibitor GNF-2 in Philadelphia chromosome-positive acute lymphatic leukemia

    Background: The t(9;22) translocation leads to the formation of the chimeric breakpoint cluster region/c-abl oncogene 1 (BCR/ABL) fusion gene on der22, the Philadelphia chromosome. The p185(BCR/ABL) or the p210(BCR/ABL) fusion proteins are encoded as a result of the translocation, depending on whether a "minor" or "major" breakpoint occurs, respectively. Both p185(BCR/ABL) and p210(BCR/ABL) exhibit constitutively activated ABL kinase activity. Through fusion to BCR the ABL kinase in p185(BCR/ABL) and p210(BCR/ABL) "escapes" the auto-inhibition mechanisms of c-ABL, such as allosteric inhibition. A novel class of compounds including GNF-2 restores allosteric inhibition of the kinase activity and the transformation potential of BCR/ABL. Here we investigated whether there are differences between p185(BCR/ABL) and p210(BCR/ABL) regarding their sensitivity towards allosteric inhibition by GNF-2 in models of Philadelphia chromosome-positive acute lymphatic leukemia. Design and methods: We investigated the anti-proliferative activity of GNF-2 in different Philadelphia chromosome-positive acute lymphatic leukemia models, such as cell lines, patient-derived long-term cultures and factor-dependent lymphatic Ba/F3 cells expressing either p185(BCR/ABL) or p210(BCR/ABL) and their resistance mutants. Results: The inhibitory effects of GNF-2 differed constantly between p185(BCR/ABL) and p210(BCR/ABL) expressing cells. In all three Philadelphia chromosome-positive acute lymphatic leukemia models, p210(BCR/ABL)-transformed cells were more sensitive to GNF-2 than were p185BCR/ABL-positive cells. Similar results were obtained for p185(BCR/ABL) and the p210(BCR/ABL) harboring resistance mutations. Conclusions: Our data provide the first evidence of a differential response of p185(BCR/ABL)- and p210(BCR/ABL)- transformed cells to allosteric inhibition by GNF-2, which is of importance for the treatment of patients with Philadelphia chromosome-positive acute lymphatic leukemia

    Allosteric inhibition enhances the efficacy of ABL kinase inhibitors to target unmutated BCR-ABL and BCR-ABL-T315I

    Background: Chronic myelogenous leukemia (CML) and Philadelphia chromosome-positive (Ph+) acute lymphatic leukemia (Ph + ALL) are caused by the t(9;22), which fuses BCR to ABL resulting in deregulated ABL-tyrosine kinase activity. The constitutively activated BCR/ABL-kinase "escapes" the auto-inhibition mechanisms of c-ABL, such as allosteric inhibition. The ABL-kinase inhibitors (AKIs) Imatinib, Nilotinib or Dasatinib, which target the ATP-binding site, are effective in Ph + leukemia. Another molecular therapy approach targeting BCR/ABL restores allosteric inhibition. Given the fact that all AKIs fail to inhibit BCR/ABL harboring the 'gatekeeper' mutation T315I, we investigated the effects of AKIs in combination with the allosteric inhibitor GNF2 in Ph + leukemia. Methods: The efficacy of this approach on the leukemogenic potential of BCR/ABL was studied in Ba/F3 cells, primary murine bone marrow cells, and untransformed Rat-1 fibroblasts expressing BCR/ABL or BCR/ABL-T315I as well as in patient-derived long-term cultures (PDLTC) from Ph + ALL-patients. Results: Here, we show that GNF-2 increased the effects of AKIs on unmutated BCR/ABL. Interestingly, the combination of Dasatinib and GNF-2 overcame resistance of BCR/ABL-T315I in all models used in a synergistic manner. Conclusions: Our observations establish a new approach for the molecular targeting of BCR/ABL and its resistant mutants using a combination of AKIs and allosteric inhibitors

    EC-IoT : an easy configuration framework for constrained IoT devices

    Connected devices offer tremendous opportunities. However, their configuration and control remains a major challenge in order to reach widespread adoption by less technically skilled people. Over the past few years, a lot of attention has been given to improve the configuration process of constrained devices with limited resources, such as available memory and absence of a user interface. Still, a major deficiency is the lack of a streamlined, standardized configuration process. In this paper we propose EC-IoT, a novel configuration framework for constrained IoT devices. The proposed framework makes use of open standards, leveraging upon the Constrained Application Protocol (CoAP), an application protocol that enables HTTP-like RESTful interactions with constrained devices.     This review supplies the reader with a detailed overview on the utilization of intramolecular processes for a design and synthesis of prodrugs. It is well known that a respected number of drugs suffer from low bioavailability, toxicity, unpleasant taste and presystemic first-pass metabolism which result in drug inactivation. The classical prodrug approach in which the linkage attaching the parent drug to its non-toxic linker and cleaved by in vivo enzyme’s catalyzed reactions has proven its success in solving toxicity and bioavailability related issues. On the other hand, prodrugs based on chemical interconversion in which the prodrug releases the corresponding active parent drug via inter or intramolecular chemical process in the absence of an enzyme is considered as a better alternative approach since the prodrug cleavage is not dependent in the efficiency or quantity of the enzyme catalyzes the interconversion of the prodrug. Examples of successful prodrugs using the chemical approach via intramolecular processes such as cyclization reactions are illustrated as well. In addition, another part of this review is devoted to cover reported studies on enzyme models and their utilization for the design and synthesis of a variety of novel prodrugs. In this approach, computational calculations using DFT and MM methods were exploited and correlations between experimentally determined and computed values of the rate-limiting step in the studied intramolecular processes were utilized in the prodrugs design. Selected examples of the designed prodrugs include aza-nucleosides for the treatment of myelodysplastic syndromes, the anti-Parkinson’s agent dopamine, the anti-viral acyclovir, the anti-malarial atovaquone, and statins for lowering cholesterol levels in the blood, the antihypertensive atenolol, the antibacterial cefuroxime, the anti-bleeding tranexamic acid, the decongestant phenylephrine, and the pain killer paracetamol

    Suicidal Behavior and Psychological Distress in University Students: A 12-Nation Study. Archives of Suicide Research

    This study investigated the prevalence of suicidal behavior and psychological distress in university students across 12 nations. A total of 5572 university students from 12 countries were surveyed about suicide ideation, suicide attempts and psychological distress by means of a self-administered questionnaire. Almost 29% of the samples reported having contemplated suicide and 7% reported attempting suicide. Of the total sample, 51.1% scored above the General Health Questionnaire-12 ≥ 3 cut-off point, 41.6% above the GHQ-12 ≥ 4 cut-off point, and 33.8% scored above the GHQ-12 ≥ 5 cut-off point. While odds of suicide ideation were elevated in Austria and the UK, reduced ORs were detected for China, Italy, Saudi Arabia, Tunisia, and Turkey. Similarly, while odds of suicide attempt were high in Jordan, Palestine, Saudi Arabia, and to some extent in Turkey, reduced ORs were observed for Austria, China, Italy, Japan and the USA. Elevated ORs for psychological distress were seen in Japan, Jordan, Palestine, Saudi Arabia, Tunisia and Turkey but reduced ORs were noted in Austria, China, Iran, Italy and the USA. Psychological distress was strongly associated with reports of suicide ideation and attempts. Suicide ideation, suicide attempt and psychological distress are common in university students but their rates vary depending on the sociocultural context. Due attention should be devoted to the mental health needs of young adults enrolled in higher educational institutions and more cross-cultural research is warranted to better understand the etiology of the observed intersocietal variations in suicidal behavior and psychological distress

    Experimental evaluation of unicast and multicast CoAP group communication

    The Internet of Things (IoT) is expanding rapidly to new domains in which embedded devices play a key role and gradually outnumber traditionally-connected devices. These devices are often constrained in their resources and are thus unable to run standard Internet protocols. The Constrained Application Protocol (CoAP) is a new alternative standard protocol that implements the same principals as the Hypertext Transfer Protocol (HTTP), but is tailored towards constrained devices. In many IoT application domains, devices need to be addressed in groups in addition to being addressable individually. Two main approaches are currently being proposed in the IoT community for CoAP-based group communication. The main difference between the two approaches lies in the underlying communication type: multicast versus unicast. In this article, we experimentally evaluate those two approaches using two wireless sensor testbeds and under different test conditions. We highlight the pros and cons of each of them and propose combining these approaches in a hybrid solution to better suit certain use case requirements. Additionally, we provide a solution for multicast-based group membership management using CoAP

    Effect of Erythropoietin on Haematological Parameters in Chronic Renal Failure Patients Undergoing Dialysis in Malaysia

    Background: The worldwide rise in the number of patients with chronic renal failure (CRF). Chronic Renal Failure is a major health problem resulting in considerable increase in mortality and morbidity, decreased quality of life and heavy cost of therapies. . Objective: This study aims to evaluate the haematological parameters and iron status during monitoring of Chronic Renal Failure patients and to study the effects of subcutaneously and intravenously administered erythropoietin (EPO) in treating anaemia in Chronic Renal Failure patients. Methods: A retrospective record review study was conducted among the CRF patients treated at Hospital Pulau Pinang between 2005 and 2009. A total of 45 patients were randomly selected by using simple random sampling. The parameters were taken one to two weeks prior to EPO treatment and at 1 month and 3 months after EPO therapy was started. Data analyses were performed by using paired t test and independent t test via SPSS Version. 18.0.0. Results: After one month post-EPO, there was a significant difference of mean serum iron between continuous ambulatory peritoneal dialysis (CAPD) and haemodialysis groups where the mean value of serum iron was higher in CAPD group. After three months post-EPO, there were significant differences of mean white blood cells and serum iron between the two groups. Conclusions: Chronic renal failure patients undergoing dialysis responded to EPO treatment and subcutaneous EPO administration appeared to give more favourable results compared to intravenous EPO.The authors of this study wish to thank the Ministry of Health, Malaysia, for thier giving us perminssion to conduct in this study and health staff of the Hospital Pulau Pinang, for their assistance during this study period.     Hexavalent Chromium Removal and Reduction to Cr (III) by Polystyrene Tris(2-aminoethyl)amine

    A commercially available chelating polymer, polystyrene tris(2-aminoethyl)amine, was used for the removal of chromium from aqueous solution. The influence of pH, contact time, adsorbent dosage and initial Cr (VI) concentration on adsorption was studied. The optimum pH for the removal of Cr (VI) was at pH 5, while optimum contact time and adsorbent dosage were 120 minutes and 10 g/L, respectively. Total chromium and Cr (VI) concentrations were analyzed by ICP-MS and UVVisible. Adsorption isotherms using Langmuir and Freundlich isotherm models revealed that the data fitted Langmuir isotherm model better than Freundlich with a maximum adsorption capacity of 312.27 mg/g. FTIR spectroscopy, Scanning electron microscopy (SEM) and Energy Dispersive Spectrometry (EDS) analyses were performed on the adsorbent before and after binding Cr (VI). All analyses confirmed the complexation of Cr (VI) to the adsorbent. Desorption experiments using KCl solution indicated 89.3% release of chromium, rendering this method of high potential for adsorbent regeneration.We wish to thank Professor Dr. Z.
