    ESTADO ATUAL DA LAGOQUILASCARÍASE HUMANA NO BRASIL

    Helmintos do gênero Lagochilascaris causam uma parasitose emergente, rara e potencialmente grave. Na literatura, encontram-se descritas cinco espécies do gênero, sendo Lagochilascaris minor a única associada à infecção humana. No Brasil são observados mais de 90% dos registros de lagochilascaríase humana mundial. As principais formas de contaminação é a ingestão de carne crua ou mal cozida de animais silvestres contendo larvas encistadas do parasito e/ou ingestão de larvas infectantes, juntamente com água ou alimentos contaminados com fezes de hospedeiros silvestres. Neste trabalho foi realizada uma revisão integrativa de literatura sobre o parasita Lagochilascaris minor. A busca nos bancos de dados foi realizada com a utilização do portal Capes, nas bases ScieLO, ScienceDirect e LILACS indexados nos últimos 100 anos. Um dos critérios de inclusão foi seleção de artigos com acesso ao texto completo. A busca retornou 122 publicações e depois de aplicados os critérios de inclusão e exclusão, 35 trabalhos foram selecionados para participar da pesquisa, denotando a escassez de publicações sobre o tema. A lagochilascaríase é uma doença de evolução crônica cujo processo infeccioso pode persistir por vários anos, sobretudo quando o verme se aloja no tecido subcutâneo do pescoço, seios paranasais e mastoide. O parasita realiza autoinfecção no ser humano e isso leva à cronificação da doença. A migração do parasito, por meio dos tecidos, origina lesões secundárias próximas ou distantes do abscesso inicial, formando verdadeiros túneis nos tecidos comprometidos, demonstrando que o parasito apresenta intensa capacidade de lisar os tecidos dos hospedeiros definitivos. Apesar dos avanços na pesquisa científica, esta ainda é uma parasitose emergente e pouco conhecida, até mesmo por profissionais da saúde. É necessário o conhecimento para que sejam tomadas medidas no sentido de reduzir a contaminação dos ambientes, evitando assim a disseminação dessa parasitose nos possíveis hospedeiros envolvidos

    Genome-wide methylation profile of mitochondrial DNA across bovine preimplantation development

    This study characterized variations in the methylation profile of mitochondrial DNA (mtDNA) during initial bovine embryo development and correlated the presence of methylation with mtDNA transcription. Bovine oocytes were obtained from abattoir ovaries and submitted to in vitro culture procedures. Oocytes and embryos were collected at various stages (immature oocyte, IM; mature oocyte, MII; zygote, ZY; 4-cells, 4C; 16-cells, 16C and blastocysts, BL). Total DNA (including mtDNA) was used for Whole Genome Enzymatic Methyl Sequencing and for quantification of mtDNA copy number. Extracted RNA was used for quantification of mitochondrial transcripts using Droplet Digital PCR. We selected ND6, CYTB, tRNA-Phe and tRNA-Gln based on their location in the mitochondrial genome, functionality and/or previous literature associating these regions with cytosine methylation. The number of mtDNA copies per oocyte/embryo was found to be similar, while methylation levels in mtDNA varied among stages. Higher total methylation levels were found mainly at 4C and 16C. In specific gene regions, higher methylation levels were also observed at 4C and 16C (ND6, CYTB and tRNA-Phe), as well as an inverse correlation with the quantity of transcripts for these regions. This is a first description of epigenetic changes occurring in mtDNA during early embryonic development. Our results indicate that methylation might regulate the mtDNA transcription at a local level, particularly around the time of embryonic genome activation

    A Closer Look at Small-Scale Magnetic Flux Ropes in the Solar Wind at 1 AU: Results from Improved Automated Detection

    Small-scale interplanetary magnetic flux ropes (SMFRs) are similar to ICMEs in magnetic structure, but are smaller and do not exhibit ICME plasma signatures. We present a computationally efficient and GPU-powered version of the single-spacecraft automated SMFR detection algorithm based on the Grad-Shafranov (GS) technique. Our algorithm is capable of processing higher resolution data, eliminates selection bias caused by a fixed \avg{B} threshold, has improved detection criteria demonstrated to have better results on an MHD simulation, and recovers full 2.5D cross sections using GS reconstruction. We used it to detect 512,152 SMFRs from 27 years (1996 to 2022) of 3-second cadence \emph{Wind} measurements. Our novel findings are: (1) the radial density of SMFRs at 1 au (∼1{\sim}1 per \si{10^6\kilo\meter}) and filling factor (∼{\sim}35\%) are independent of solar activity, distance to the heliospheric current sheet (HCS), and solar wind plasma type, although the minority of SMFRs with diameters greater than ∼{\sim}0.01 au have a strong solar activity dependence; (2) SMFR diameters follow a log-normal distribution that peaks below the resolved range (≳104\gtrsim 10^4 km), although the filling factor is dominated by SMFRs between 10510^5 to 10610^6 km; (3) most SMFRs at 1 au have strong field-aligned flows like those from PSP measurements; (4) in terms of diameter dd, SMFR poloidal flux ∝d1.2\propto d^{1.2}, axial flux ∝d2.0\propto d^{2.0}, average twist number ∝d−0.8\propto d^{-0.8}, current density ∝d−0.8\propto d^{-0.8}, and helicity ∝d3.2\propto d^{3.2}. Implications for the origin of SMFRs and switchbacks are briefly discussed. The new algorithm and SMFR dataset are made freely available

    LUMINOS-102: Lerapolturev with and without α-PD- 1 in unresectable α-PD- 1 refractory melanoma

    Lerapolturev (lera, formerly PVSRIPO) is a novel poliovirus based intratumoral immunotherapy that infects both cancer cells and antigen-presenting cells (APCs) via CD155, the poliovirus receptor. Lera has direct anticancer effects while also generating type I/III interferon-dominated inflammation and anti-tumor T-cell priming and activation via infection of local APCs. LUMINOS-102 (NCT04577807) is a multi-center, open-label, two-arm randomized Phase 2 study investigating the efficacy and safety of lera ± α-PD- 1 in patients with unresectable melanoma who failed prior α-PD- 1 therapy. Cross-over to the α-PD- 1 arm is permitted after progression, PR for ≥6 mo or 6 mo on treatment with SD. The maximum initial lera dose was 6x108 TCID50 /visit every 3 or 4 weeks (Q3/4 W). As of March 2022, the maximum lera dose was increased to 1.6 x 109 TCID50/visit, every week (QW) for 7 weeks (induction), followed by Q3/4 W dosing (maintenance). As of 20-Jun- 2022, 21 participants (10 male, 11 female, median 64 yrs) received lera (n = 14 at initial dose, Q3/4 W; n = 4 at increased dose, Q3/4 W; n = 3 at increased dose, QW) ± αPD-1. Five patients are currently on treatment. With the initial regimen, no objective responses and a CBR of 7% were observed. However, with the higher dose regimen, 1 complete response and a CBR of 71% (5/7) has been observed. Two of 4 participants with stable disease have evidence of response (1 with resolution of uninjected lung metastasis, 1 with decreased PET signal in injected and uninjected lesions receiving combination therapy). The only treatment related AE in \u3e1 pt was fatigue (19%, all grade 1 or 2). No dose-limiting toxicities or treatment-related SAEs were reported. Multiplex-IF analysis of on-treatment tumor biopsies will be presented. Lera ± αPD-1 is well tolerated, with early signs of efficacy at the higher dose level. Enrollment and randomization are ongoing

    A partir de uma experiência pessoal somada à construção de conhecimento acadêmico, incitou-se o desejo de ampliar os conhecimentos acerca da atuação do psicólogo hospitalar fundamentado na Abordagem Centrada na Pessoa (ACP), no Brasil. Nesse sentido, esta pesquisa, com abordagem qualitativa, tem como objetivo compreender as formas de atuação destes profissionais, como também, analisar as discussões e apontamentos que realizam durante as suas práticas. Para isso, foi realizada uma pesquisa documental de caráter exploratório, que utilizou como metodologia a Revisão Sistemática de Literatura (RSL). A coleta de dados se deu nos bancos SciELO, Periódicos CAPES, BVS Brasil, Revista Gestáltica, NUFEN, Redalyc. Foi utilizado também da Literatura Cinzenta para mapear outras fontes de dados, como o portal eletrônico Encontro ACP e incluir referências indicadas por pesquisadores dessa abordagem, os quais são nomeados como especialistas. Fizemos uso da análise descritiva qualitativa, bem como do referencial teórico da ACP para compreensão dos resultados. Estes, delineiam a conduta do psicólogo fundamentado na ACP em contexto de saúde/hospitalar, onde visualiza ser possível atuar em diferentes espaços dentro da instituição, com diferentes públicos e formatos psicoterapêuticos, possibilitando, assistir aos envolvidos no contexto de maneira a assegurar uma atuação humanizada, a partir dos pressupostos da ACP.From a personal experience added to the construction of academic knowledge, the desire to expand knowledge about the work of the hospital psychologist based on the Person-Centered Approach (PCA), in Brazil. In this sense, this research, with a qualitative approach, aims to understand the ways of acting of these professionals, as well as to analyze the discussions and notes they make during their practices. For this, an exploratory documentary research was carried out, which used as methodology the Systematic Literature Review (RSL). Data were collected in the databases SciELO, Periódicos CAPES, BVS Brasil, Revista Gestáltica, NUFEN, Redalyc. Grey Literature was also used to map other data sources, such as the electronic portal Encontro ACP and to include references indicated by researchers of this approach, who are appointed as experts. We used the qualitative descriptive analysis, as well as the theoretical framework of the PCA to understand the results. These outline the conduct of the psychologist based on the PCA in the health/hospital context, where he sees it being possible to act in different spaces within the institution, with different audiences and psychotherapeutic formats, making it possible to assist those involved in the context to ensure a humanized performance, based on the assumptions of the PCA.A partir de una experiencia personal añadida a la construcción del conocimiento académico, se alentó el deseo de ampliar el conocimiento sobre el trabajo del psicólogo hospitalario basado en el Enfoque Centrado en la Persona (ECP), en Brasil. En este sentido, esta investigación, con un enfoque cualitativo, tiene como objetivo comprender las formas de actuar de estos profesionales, así como analizar las discusiones y notas que hacen durante sus prácticas. Para ello, se realizó una investigación documental exploratoria, que utilizó como metodología la Revisión Sistemática de la Literatura (RSL). Los datos fueron recolectados en las bases de datos SciELO, Periódicos CAPES, BVS Brasil, Revista Gestáltica, NUFEN, Redalyc. También se utilizó la Literatura Gris para mapear otras fuentes de datos, como el portal electrónico Encontro ACP e incluir referencias indicadas por investigadores de este enfoque, que son designados como expertos. Se utilizó el análisis descriptivo cualitativo, así como el marco teórico del ECP para comprender los resultados. Estos describen la conducta del psicólogo basada en la ECP en el contexto salud/hospitalario, donde ve posible actuar en diferentes espacios dentro de la institución, con diferentes audiencias y formatos psicoterapéuticos, permitiendo asistir a los involucrados en el contexto para garantizar un desempeño humanizado, basado en los supuestos de la ECP

    Parker Solar Probe Encounters the Leg of a Coronal Mass Ejection at 14 Solar Radii

    We use Parker Solar Probe (PSP) observations to report the first direct measurements of the particle and field environments while crossing the leg of a coronal mass ejection (CME) very close to the Sun (∼14 Rs). An analysis that combines imaging from 1 au and PSP with a CME model, predicts an encounter time and duration that correspond to an unusual, complete dropout in low-energy solar energetic ions from H–Fe, observed by the Integrated Science Investigation of the Sun (IS⊙IS). The surrounding regions are populated with low-intensity protons and heavy ions from 10s to 100 keV, typical of some quiet times close in to the Sun. In contrast, the magnetic field and solar wind plasma show no similarly abrupt changes at the boundaries of the dropout. Together, the IS⊙IS energetic particle observations, combined with remote sensing of the CME and a dearth of other “typical” CME signatures, indicate that this CME leg is significantly different from the magnetic and plasma structure normally assumed for CMEs near the Sun and observed in interplanetary CMEs farther out in the solar wind. The dropout in low-energy energetic ions may be due to the cooling of suprathermal ions at the base of the CME leg flux tube, owing to the rapid outward expansion during the release of the CME

    [Italiano]: La ricerca si colloca nel settore scientifico disciplinare del disegno focalizzando l’attenzione sulle problematiche dell’autismo e sulla possibilità di guidare la percezione dello spazio architettonico attraverso il ricorso alla realtà virtuale. Lo studio intende ribadire la necessità che le discipline della rappresentazione possano contribuire in maniera determinante a un ambito di ricerca finora dominato dalle sole discipline mediche, pedagogiche e tecnologiche costituendosi, non solo come interprete della sintassi spaziale, ma anche come medium interdisciplinare per osservare con una nuova ottica i dati di output della sperimentazione. La trattazione, per il suo carattere polifunzionale, integra le visioni congiunte di più ambiti teorici; oltre a quello più specifico del disegno, anche il settore ergonomico, pedagogico, museale e l'area delle neuroscienze. L’autrice si propone di colmare alcune lacune, riscontrate nell’ambito di studi indagato, sulla risposta percettiva legata alla rappresentazione in VR spostando il focus d’attenzione, dagli aspetti tecnologici dello strumento digitale, all’utente, con lo scopo di incrementare il potenziale della realtà virtuale come medium educativo e inclusivo. Partendo da questi presupposti, lo studio affronta una profonda analisi delle risposte percettive nel rapporto uomo-spazio architettonico, sia in ambito neurotipico che nello scenario autistico, per approfondire il ruolo della disciplina della rappresentazione come guida nell’interpretazione delle morfologie spaziali per persone con compromissioni percettive. Dal confronto dei dati di output delle ricerche studiate, scaturisce l'esigenza di linee guida per la progettazione delle ambientazioni virtuali che, superando le lacune delle precedenti sperimentazioni, tenga conto di tutti i fattori analizzati nelle fasi di studio preliminari, fattori umani ed ergonomici, percettivi, educativi, di inclusività e in cui la disciplina della rappresentazione, ponendosi come tramite interdisciplinare, fornisca gli strumenti operativi più opportuni per il raggiungimento degli obiettivi prefissati. Attraverso un’adeguata interpretazione dei codici spaziali, guidata dagli strumenti di analisi e comunicazione delle discipline ICAR 17, lo studio approda alla definizione di uno strumento d’indirizzo per la progettazione di ambienti virtuali autism friendly, dove il patrimonio architettonico diventi medium di inclusione e comunicazione del suo valore culturale. Il “ri-disegno” di uno spazio configurato sulle specificità di un’utenza più debole ha, come ulteriore ricaduta, l’esito di estendere in modo indiscriminato la possibilità di accesso alla cultura, dal momento che migliorare il rapporto con lo spazio, può aiutare a migliorare anche il rapporto con noi stessi e con gli altri. / [English]: The research belongs to the disciplinary field of drawing disciplines, focusing attention on autism’s needs, and aiming at guiding the architectural space perception using virtual reality. The drawing disciplines, providing an interpretation of spatial syntax, become an interdisciplinary medium to observe with a new perspective the output data of the experimentation, this way incisively contributing to a field of research hitherto dominated only by medical, pedagogical, and technological disciplines. The interdisciplinary research methodology integrates the joint visions of multiple theoretical fields; in addition to the more specific one of drawing, also the ergonomic, pedagogical, museum sector and the neuroscience area. The author aims to fill some gaps, found in the investigated context, about human perceptual answers to the VR representation, shifting the focus of attention, from the technological aspects of the digital tool to the user. This way, the research increases the potential of virtual reality as an educational and inclusive medium. Starting from these assumptions, the study deals with a profound analysis of the perceptive responses in the relationship between man and architectural space, both in the neurotypical context and in the autistic scenario, to explore the role of the drawings discipline as a guide in the interpretation of spatial morphologies for people with perceptual impairments. Comparing the analytical phase’s output data, the research develops the main design objectives aimed at developing the guidelines tool for the autism-friendly virtual environment design. This way it overcomes the gaps of previous experiments considering all the factors analyzed in the preliminary study phases, human and ergonomic factors, perceptive, educational, and inclusive. The drawing disciplines, acting as an interdisciplinary medium, provide the most appropriate operational tools for achieving the preset objectives. The "re-design" of a space customized to a weaker user's needs has, as a further consequence, the outcome of indiscriminately extending the possibility of access to culture, since improving the relationship with the space can also help improve our relationship with ourselves and with others

    The ILIAD Safety Stack: Human-Aware Infrastructure-Free Navigation of Industrial Mobile Robots

    Safe yet efficient operation of professional service robots within logistics or production in human-robot shared environments requires a flexible human-aware navigation stack. In this manuscript, we propose the ILIAD safety stack comprising software and hardware designed to achieve safe and efficient motion specifically for industrial vehicles with nontrivial kinematics The stack integrates five interconnected layers for autonomous motion planning and control to enable short- and long-term reasoning. The use-case scenario tested requires an autonomous industrial forklift to safely navigate among pick-and-place locations during normal daily activities involving human workers. Our test-bed in the real world consists of a three-day experiment in a food distribution warehouse. The evaluation is extended in simulation with an ablation study of the impact of different layers to show both the practical and the performance-related impact. The experimental results show a safer and more legible robot when humans are nearby with a trade-off in task efficiency, and that not all layers have the same degree of impact in the system

    Reactivity to allergenic food contaminants: A study on products on the market

    Abstract Background The frequency and severity of reactions in food‐allergic consumers exposed to unintentional food allergen contamination during production is unknown. To warn allergic consumers, it has been suggested for pre‐packaged foods to be precautionary labelled when the food allergen contamination may exceed the amount to which 1%–5% of the population could react (ED01–ED05). ED01 for hazelnut and milk have been estimated at 0.1 and 0.2 mg, respectively, by the Voluntary Incidental Trace Allergen Labelling (VITAL) initiative. The respective reference doses recommended by the FAO/WHO Codex consultation are 3 and 2 mg. We evaluated the reactivity to potential traces of milk and hazelnut allergens in allergen‐free pre‐packaged products by children affected by severe allergies to milk and hazelnuts. Methods Oral Food Challenges with commercially available hazelnut‐free wafer biscuits and milk‐free chocolate pralines were administered to patients with severe food allergies to hazelnut and cow's milk, respectively. Contamination levels of milk or hazelnut allergens were measured using chromatographic separation interfaced with triple quadrupole mass spectrometry. Results No hazelnut allergic patient showed allergic reactions to exposure to biscuits, nor any milk allergic patient displayed allergic reactions to the dark chocolate praline. While no hazelnut trace was detected in biscuits, the praline was found to be contaminated by milk at concentrations ranging between 8 and 35 mg total protein/kg food. In our dose model, these amounts exceeded 1.5–10 times the VITAL ED01 and reached the threshold suggested by the FAO/WHO Codex consultation. Conclusions Upon the consumption of food products available on the market, many patients with severe food allergies tolerate significantly higher doses of allergen than reference doses indicated in the VITAL system used for precautionary allergen labelling. These doses support the safety of the FAO/WHO recommended reference doses
