Its purpose is an assessment of the structure in material and determination of static and fatigue characteristics. In the theoretical part of thesis, there is explained, how to divide and mark kinds of steel. There is also a mention about basic parameters of material and the main attention is paid to fatigue and its laws. The metallographic analysis, tensile test and the fatigue test were realized in the experimental part of thesis. Experimentally determined values were compared with data in the literature.

The theoretical part is divided into three parts. The first one defines the austenitic stainless steels with accent to their chemical composition and microstructure. Second part is dedicated to deformation-induced martensite formation. The third part shows the mechanisms and stages of damage due to cyclic straining. The experimental part is focused especially on the observing of microstructural changes by methods of colour metallography and their relation to stress-strain response of material under selected conditions of fatigue loading. Series of samples of 304 and 316L steels were prepared by colour etching and their microstructures were documented using optical microscopy. Then the semi-quantitative measurements of presence and morphology of deformation-induced martensite were done and the results were discussed and correlated with fatigue damage level.

    Der demographische Wandel und die dadurch bedingte Zunahme des Anteils alter und multimorbider Menschen in unserer Gesellschaft stellt die Medizin vor neue Herausforderungen. Insbesondere in der Geriatrie spielt der ganzheitliche Blick auf die Patient*innen und ihre Gesundheitsprobleme eine entscheidende Rolle. Das ICF-Modell erm├Âglicht diesen ganzheitlichen Blick, indem es f├╝nf Komponenten der Funktionsf├Ąhigkeit, Behinderung und Gesundheit definiert, die in ihrer Gesamtheit eine Erkl├Ąrung f├╝r ein Gesundheitsproblem liefern k├Ânnen. Mobilit├Ątseinschr├Ąnkungen haben bei geriatrischen Patient*innen eine hohe Pr├Ąvalenz. Sie sind mit einer erh├Âhten Morbidit├Ąt und Mortalit├Ąt sowie mit einer geringeren Lebensqualit├Ąt assoziiert. In dieser Arbeit wurde untersucht, inwiefern die motorische Funktion von geriatrischen Patient*innen sowie ihre Ver├Ąnderung nach einer fr├╝hrehabilitativen geriatrischen Komplexbehandlung durch das ICF-Modell erkl├Ąrt werden kann. Das Gesamtmodell erwies sich als statistisch signifikant f├╝r die motorische Funktion von geriatrischen Patient*innen zum Aufnahmezeitpunkt. Von den einzelnen Faktoren konnten Sturzangst und motorische Symptome einen signifikanten Einfluss aufweisen. Diese Ergebnisse zeigen, dass sich das ICF-Modell in seiner ganzheitlichen Betrachtung f├╝r die Erkl├Ąrung von Gesundheitsproblemen eignet. Die Ver├Ąnderung der motorischen Funktion konnte durch das ICF-Modell nicht erkl├Ąrt werden. Hierf├╝r verantwortlich k├Ânnte die begrenzte Messbarkeit dieser Ver├Ąnderung durch die SPPB sein, die bereits in vorherigen Untersuchungen einen Deckeneffekt zeigte. F├╝r die dezidierte Betrachtung der Ver├Ąnderung der motorischen Funktion bedarf es daher genauerer Messinstrumente wie zum Beispiel Gang- und Gleichgewichtsparameter anhand Sensor-basierter Daten

    Cellular O-Glycome Reporter/Amplification to explore O-glycans of living cells

    Protein O-glycosylation has key roles in many biological processes, but the repertoire of O-glycans synthesized by cells is difficult to determine. Here we describe an approach termed Cellular O-Glycome Reporter/Amplification (CORA), a sensitive method used to amplify and profile mucin-type O-glycans synthesized by living cells. Cells convert added peracetylated benzyl-╬▒-N-acetylgalactosamine to a large variety of modified O-glycan derivatives that are secreted from cells, allowing for easy purification for analysis by HPLC and mass spectrometry (MS). Relative to conventional O-glycan analyses, CORA resulted in an Ôł╝100-1,000-fold increase in sensitivity and identified a more complex repertoire of O-glycans in more than a dozen cell types from Homo sapiens and Mus musculus. Furthermore, when coupled with computational modeling, CORA can be used for predictions about the diversity of the human O-glycome and offers new opportunities to identify novel glycan biomarkers for human diseases

    Treatment of vancomycin overdose using high-efficiency dialysis membranes

    Two children underwent acute hemodialysis using high-efficiency dialysis membranes for vancomycin intoxication (plasma levels 238 ┬Ág/ml and 182 ┬Ág/ml). During a 3-h treatment, plasma vancomycin removal was on average 60%, with a calculated vancomycin half-life ( t 1/2 ) of 2 h. This is in contrast to a recent report using charcoal hemoperfusion for vancomycin intoxication (plasma level of 137 ┬Ág/ml), which resulted in a 40% relative plasma clearance and a calculated vancomycin t 1/2 of 12.5 h for a 4-h treatment. The choice of optimal modality for clearing a toxin should take into account the availability of equipment, protein or lipid binding of the toxin, and inherent risks of charcoal hemofiltration (large extracorporeal circuit, reversible hypocalcemia, heat loss, reversible coagulation defects) versus risks of high-efficiency hemodialysis (large extracorporeal circuit).Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/42299/1/467-13-9-773_90130773.pd
