    Influence of synoptic meteorology on airborne allergenic pollen and spores in an urban environment in Northeastern Iberian Peninsula

Six pollen types (Platanus, Cupressaceae, Olea, Poaceae, Urticaceae and Amaranthaceae), and one fungal spore (Alternaria) were selected for their high allergenic effect in sensitive people. Six synoptic meteorological patterns were identified through cluster analysis of sea level pressure fields as the main responsible of the weather conditions in the Iberian Peninsula. The local meteorological conditions in Barcelona associated with each one of the synoptic types were also stablished. Different statistical methods were applied to analyse possible relationships between concentrations and timing of the recorded aerobiological particles and specific synoptic types. The study, focused in the 19-year period 2001-2019, shows that one of the scenarios, frequent in winter and linked to high stability and air-mass blockage, registered the highest mean and median values for Platanus and Cupressaceae, but it was not very relevant for the other taxa. It was also this scenario that turned out to be the most influent on the pollination timing showing a significant influence on the start occurrence of Urticaceae flowering and on the peak date of Platanus. On the other hand, the most frequent synoptic type in the period, relevant in spring and summer, was linked to sporadic episodes of levels considered to be of high risk of allergy to Platanus, Poaceae, and Urticaceae pollen, and Alternaria fungal spore. This synoptic pattern, characterized by the presence of the Azores anticyclone and the Atlantic low located in the north of the United Kingdom, was associated with high temperatures, low relative humidity and moderate winds from the NW in Barcelona. The identification of an interaction between synoptic meteorology and pollen/spore dynamics will allow better abatement measures, reducing adverse health effects on sensitive population

    Maximal tear secretion evoked by controlled stimulation of corneal sensory nerves in healthy individuals and dry eye subjects

    [Purpose] To measure, the tear flow changes evoked in healthy subjects and dry eye disease (DED) patients by controlled sensory stimulation of the eye surface with i-Onion‚ĄĘ, a new stimulation device.[Methods] Sensory corneal nerves were stimulated with an instrument (i-Onion‚ĄĘ) that ejects puffs of CO2 gas (99.9%) at 200 ml¬∑min‚ąí1 for 3s, delivered 5 mm from the cornea. Using Schirmer test strips, tear volumes were measured over 3 min in the cornea of one eye before (basal tear volume -BTV) and in the other eye after stimulation of the sensory nerves with CO2 (stimulated tear volume -STV). These measurements were obtained from a control group of adults of either sex (17 students aged 20‚Äď30 and 29 subjects without signs of dry eye aged 25‚Äď61), a cohort of DED patients (aged 34‚Äď75) that included 12 asymptomatic, suspected DED subjects (Schirmer 10 mm, the remaining 70% responded vigorously to stimulation, even those with a BTV >18 mm. Asymptomatic DED subjects displayed weaker responses to CO2 stimulation, with lower STVs.     CartoCell, a high-content pipeline for 3D image analysis, unveils cell morphology patterns in epithelia

    Decades of research have not yet fully explained the mechanisms of epithelial self-organization and 3D packing. Single-cell analysis of large 3D epithelial libraries is crucial for understanding the assembly and function of whole tissues. Combining 3D epithelial imaging with advanced deep-learning segmentation methods is essential for enabling this high-content analysis. We introduce CartoCell, a deep-learning-based pipeline that uses small datasets to generate accurate labels for hundreds of whole 3D epithelial cysts. Our method detects the realistic morphology of epithelial cells and their contacts in the 3D structure of the tissue. CartoCell enables the quantification of geometric and packing features at the cellular level. Our single-cell cartography approach then maps the distribution of these features on 2D plots and 3D surface maps, revealing cell morphology patterns in epithelial cysts. Additionally, we show that CartoCell can be adapted to other types of epithelial tissues

    Early diarrhoea under sorafenib as a marker to consider the early migration to second-line drugs

    Background: Despite atezolizumab and bevacizumab (A + B) is currently the first-line treatment for hepatocellular carcinoma (HCC) patients, some patients will not be adequate for this combination. In the setting of sorafenib some adverse events have been proposed as prognostic factors. Objective: To characterize the early diarrhoea development as prognostic factor in 344 HCC patients. Methods: The development of early diarrhoea in sorafenib treatment defined as patients who developed diarrhoea and needed dose modification within the first 60 days of treatment (e-diarrhoea) and 3-grouping variables were analysed: Patients with e-diarrhoea, patients who developed diarrhoea after the first 60 days of treatment (L-diarrhoea) and patients that never developed diarrhoea (never diarrhoea). Results: The median overall survival in sorafenib treated patients was significantly different across groups (6.8 months for e-diarrhoea, 26.7 months for L-diarrhoea and 13.3 months for never-diarrhoea). The emergence of e-diarrhoea was associated with poor outcomes (hazard ratio [HR] 1.84 [95%CI 1.15-2.95]), while there was no increased/decreased risk of dismal evolution in patients with L-diarrhoea (HR 0.66 [95%CI 0.42-1.03]). Conclusion: The emergence of e-diarrhoea in HCC patients treated with sorafenib is an early predictor of dismal evolution under this therapy. Thus, prompt identification of these non-responders may be useful for an early switch to second-line therapies

    Comparative study between the effects of autumn and winter rainfall on aerobiological variables in the NE of the Iberian Peninsula

    The correlation analysis between precipitation indices and aerobiological variables could lead to different results if seasonal rainfall is considered. The number of significant correlations, as well as the ratio of positive/negative correlations, is higher when previous autumn rainfall is taken into account than for annual rainfall. However, the percentage of significant correlations is only 5.1 % of the total possible correlations. The autumn indices showing more correlations are related to rainfall duration and persistence, while those for winter were related to rainfall amount and intensity. Since the influence of moderately extreme rainfall seems to be low, the reported probable increase, due to climate change, of the occurrence of intense and short rainy events in the Mediterranean area in autumn does not seem that will have an appreciable effect by itself on the subsequent pollen production.Peer ReviewedPostprint (published version

    Bas√°ndonos en los principios vertebradores de la innovaci√≥n como pilares para la actualizaci√≥n y la excelencia educativa, resulta relevante destacar la necesidad de generar nuevas formas de llevar a cabo los procesos formativos con el fin de satisfacer los intereses del alumnado de la era digital. En esta l√≠nea, una de las novedades m√°s vanguardistas generadas por la revoluci√≥n tecnol√≥gica fue el metaverso, un mundo virtual en el cual, a trav√©s de dispositivos digitales, se brinda al usuario una experiencia inmersiva y una interacci√≥n total con sus elementos. Debido a su reciente aparici√≥n, las posibilidades del metaverso en la educaci√≥n a√ļn se encuentran en proceso de exploraci√≥n. El objetivo del proyecto se centr√≥ en la creaci√≥n de un metaverso educativo con el fin de desarrollar los contenidos did√°cticos de diferentes materias curriculares para llevar el proceso de ense√Īanza y aprendizaje a un nivel m√°s elevado de inmersi√≥n e innovaci√≥n. Para lograrlo, se llev√≥ a cabo el dise√Īo e implementaci√≥n de una metodolog√≠a did√°ctica espec√≠fica para la formaci√≥n en el metaverso, as√≠ como la elaboraci√≥n e implementaci√≥n de un plan de formaci√≥n de competencias para la creaci√≥n y uso de aulas virtuales. De esta manera, fue posible generar y transferir conocimiento en el metaverso, partiendo de la mejora de las habilidades psicoeducativas de los alumnos, y promover la educaci√≥n inclusiva y la mejora de la atenci√≥n a la diversidad. La metodolog√≠a que sigui√≥ el proyecto se bas√≥ en el trabajo colaborativo y el principio de autonom√≠a para llevar a cabo de manera efectiva un aprendizaje basado en proyectos y problemas, con el fin de impartir los contenidos did√°cticos y alcanzar los objetivos de aprendizaje. Todo esto se sustent√≥ en una estrategia sistem√°tica de seguimiento y evaluaci√≥n del desarrollo del proyecto y su impacto. Los resultados obtenidos una vez implementado el proyecto de innovaci√≥n fueron los siguientes: -Se logr√≥ un alto nivel de competencias en el profesorado para la creaci√≥n y utilizaci√≥n de aulas virtuales en el metaverso. -Se logr√≥ un alto nivel de competencias en el alumnado para un √≥ptimo desenvolvimiento en el metaverso. -Se alcanz√≥ un desarrollo efectivo, √≥ptimo y eficaz de la docencia, lo cual contribuy√≥ a la consecuci√≥n de aprendizajes significativos en las distintas titulaciones y asignaturas involucradas en el proyecto. -Se incentiv√≥ la mejora de destrezas y habilidades psicoeducativas en el alumnado, como la motivaci√≥n, la autonom√≠a, la participaci√≥n activa, la atenci√≥n e inter√©s, la interacci√≥n entre estudiantes y con los docentes, y la facilidad de acceso a los contenidos. -Se foment√≥ la educaci√≥n inclusiva y se mejor√≥ la atenci√≥n a la diversidad mediante el desarrollo del proceso formativo en el metaverso, adaptando el entorno a las necesidades de los estudiantes. -Se logr√≥ un impacto significativo en t√©rminos de alcance hacia la comunidad educativa, lo que permiti√≥ promover la utilizaci√≥n del metaverso como una propuesta pedag√≥gica innovadora en las diferentes etapas educativas obligatorias y postobligatorias, as√≠ como en diversas ramas de conocimiento. La metodolog√≠a docente adoptada se bas√≥ en los principios del trabajo colaborativo, la autonom√≠a y la atenci√≥n a la diversidad, con el objetivo de implementar de manera efectiva un aprendizaje basado en proyectos y problemas, aprovechando las posibilidades ofrecidas por la tecnopedagog√≠a. En consecuencia, se llevaron a cabo las siguientes acciones para iniciar la implementaci√≥n del metaverso educativo con los estudiantes: -Se proporcionaron gafas de realidad virtual a los estudiantes y se les presentaron breves p√≠ldoras formativas sobre su funcionamiento, como resumen de la formaci√≥n recibida en la primera fase del proyecto. -El docente present√≥ el entorno virtual creado para llevar a cabo el proceso formativo. -Los estudiantes crearon sus avatares en el entorno virtual. -Se realiz√≥ una prueba piloto del entorno virtual mediante la impartici√≥n de ciertos contenidos previamente digitalizados por los docentes. -Se resolvieron los problemas surgidos durante la prueba piloto. Despu√©s de completar las acciones anteriores, fue posible desarrollar un proceso instructivo en el cual tanto estudiantes como docentes estaban familiarizados con el nuevo modelo de aprendizaje, lo que condujo a una mayor eficacia de todo el proceso, minimizando las contingencias en la medida de lo posible

    Cisplatin-induced Cell Death Increases the degradation of the MRE11-RAD50-NBS1 Complex Through the Autophagy/lysosomal Pathway

    Cisplatin and other platinum-based anticancer agents are among the most widely used chemotherapy drugs in the treatment of different types of cancer. However, it is common to find patients who respond well to treatment at first but later relapse due to the appearance of resistance to cisplatin. Among the mechanisms responsible for this phenomenon is the increase in DNA damage repair. Here, we elucidate the effect of cisplatin on the MRN (MRE11-RAD50-NBS1) DNA damage sensor complex. We found that the tumor suppressor FBXW7 is a key factor in controlling the turnover of the MRN complex by inducing its degradation through lysosomes. Inhibition of lysosomal enzymes allowed the detection of the association of FBXW7-dependent ubiquitylated MRN with LC3 and the autophagy adaptor p62/SQSTM1 and the localization of MRN in lysosomes. Furthermore, cisplatin-induced cell death increased MRN degradation, suggesting that this complex is one of the targets that favor cell death.
