    A model relating transpiration for Japanese cedar and cypress plantations with stand structure

    Previous studies have revealed that changes in forest structure due to management (e.g., thinning, aging, and clearcutting) could affect the forest water balance. However, there are unexplained variability in changes in the annual water balance with changing structure among different sites. This is the case even when analyzing data for specific species/regions. For a more advanced and process-based understanding of changes in the water balance with changing forest structure, we examined transpiration (E) observed using the sap-flux method for 14 Japanese cedar and cypress plantations with various structure (e.g., stem density and diameter) in Japan and surrounding areas and developed a model relating E with structural parameters. We expressed E using the simplified Penman–Monteith equation and modeled canopy conductance (G[c]) as a product of reference G[c] (G[cref]) when vapor pressure deficit is 1.0 kPa and functions expressing the responses of G[c] to meteorological factors. We determined G[cref] and parameters of the functions for the sites separately. E observed for the 14 sites was not reproduced well by the model when using mean values of G[cref] and the parameters among the sites. However, E observed for the sites was reproduced well when using G[cref] determined for each site and mean values of the parameters of the functions among the sites, similar to the case when using G[cref] and the parameters of the functions determined for each site. These results suggest that considering variations in G[cref] among the sites was important to reproduce variations in E, but considering variations in the parameters of the functions was not. Our analysis revealed that G[cref] linearly related with the sapwood area on a stand scale (A) and that Alinearly related with stem density (N) and powers of the mean stem diameter (d[m]). Thus, we proposed a model relating E with A (or N and d[m]), where G[cref] was calculated from A (or N and d[m]) and the parameters of the functions were assumed to be the mean values among the sites. This model estimates changes in Ewith changing structure from commonly available data (N and d[m]), and therefore helps improve our understanding of the underlying processes of the changes in the water balance for Japanese cedar and cypress plantations

    Current status of a helicopter transportation system on remote islands for patients undergoing mechanical thrombectomy

    Background: Mechanical thrombectomy (MT) is standard treatment for acute ischemic stroke (AIS) with large-vessel occlusion within 6 h of symptom onset to treatment initiation (OTP). Recent trials have extended the therapeutic time window for MT to within 24 h. However, MT treatment remains low in remote areas. Nagasaki Prefecture, Japan has many inhabited islands with no neurointerventionalists. Our hospital on the mainland is a regional hub for eight island hospitals. We evaluated clinical outcomes of MT for patients with AIS on these islands versus on the mainland. Methods: During 2014–2019, we reviewed consecutive patients with AIS who received MT at our hospital. Patients comprised the Islands group and Mainland group. Patient characteristics and clinical outcomes were compared between groups. Results: We included 91 patients (Islands group: 15 patients, Mainland group: 76 patients). Seven patients (46.7%) in the Islands group versus 43 (56.6%) in the Mainland group achieved favorable outcomes. Successful recanalization was obtained in 11 patients (73.3%) on the islands and 67 (88.2%) on the mainland. The median OTP time in the Islands was 365 min. In both the Islands and Mainland groups, the OTP time and successful recanalization were associated with functional outcome. The modified Rankin Scale (mRS) score at 90 days ≤2 was obtained in two patients and mRS = 3 in four patients among eight patients with OTP time >6 h. Conclusions: Few patients with AIS on remote islands have received MT. Although patients who underwent MT on the islands had longer OTP, the clinical outcomes were acceptable. OTP time on remote islands must be shortened, as this is related to functional outcome. In some cases with successful recanalization, a favorable outcome can still be obtained even after 6 h. Even if OTP exceeds 6 h, it is desirable to appropriately select patients and actively perform MT

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target
