    TRA-1 ChIP-seq reveals regulators of sexual differentiation and multilevel feedback in nematode sex determination

    Sex-determining genes have been identified in many animals, but how they impose sex specificity on development is poorly understood. We ask how the nematode sex-determining transcription factor Transformer 1 (TRA-1) regulates sex by identifying where in the genome TRA-1 binds and which nearby genes may be affected by this binding. We find that TRA-1 promotes female development primarily by preventing the expression of genes involved in male development. Among the genes repressed by TRA-1 are a number that control the timing of developmental events and also several that function upstream of TRA-1 in the global sex-determination pathway. The suite of TRA-1 targets presented here provides a resource to continue uncovering the basis of sex-specific development

    Insulin-like signalling to the maternal germline controls progeny response to osmotic stress

    In 1893 August Weismann proposed that information about the environment could not pass from somatic cells to germ cells, a hypothesis now known as the Weismann barrier. However, recent studies have indicated that parental exposure to environmental stress can modify progeny physiology and that parental stress can contribute to progeny disorders. The mechanisms regulating these phenomena are poorly understood. We report that the nematode Caenorhabditis elegans can protect itself from osmotic stress by entering a state of arrested development and can protect its progeny from osmotic stress by increasing the expression of the glycerol biosynthetic enzyme GPDH-2 in progeny. Both of these protective mechanisms are regulated by insulin-like signalling: insulin-like signalling to the intestine regulates developmental arrest, while insulin-like signalling to the maternal germline regulates glycerol metabolism in progeny. Thus, there is a heritable link between insulin-like signalling to the maternal germline and progeny metabolism and gene expression. We speculate that analogous modulation of insulin-like signalling to the germline is responsible for effects of the maternal environment on human diseases that involve insulin signalling, such as obesity and type-2 diabetes

    Chandra X-ray constraints on the candidate Ca-rich gap transient SN 2016hnk

    We present a Chandra observation of SN 2016hnk, a candidate Ca-rich gap transient. This observation was specifically designed to test whether or not this transient was the result of the tidal detonation of a white dwarf by an intermediate-mass black hole. Since we detect no X-ray emission 28 d after the discovery of the transient, as predicted from fall-back accretion, we rule out this model. Our upper limit of similar to 10M(circle dot) does not allow us to rule out a neutron star or stellar-mass black hole detonator due limits on the sensitivity of Chandra to soft X-rays and unconstrained variables tied to the structure of super-Eddington accretion discs. Together with other Chandra and multiwavelength observations, our analysis strongly argues against the intermediate-mass black hole tidal detonation scenario for Ca-rich gap transients more generally

    Analytical representation of spatial and temporal variations of the geomagnetic field in the Indian region

    The magnetic measurements of declination (D), horizontal (H) and vertical (Z) components of earth's magnetic field, collected from ground surveys between 1962 and 1966, are used to develop an analytical model of geomagnetic field variations over Indian region for the epoch 1965. In order to reflect spatial features with wavelengths of approximately 1000 km, sixth degree polynomial as a function of differential latitude and longitude is calculated by the method of least squares. The root mean square fit of the model to the input data is better than that accounted by the International Geomagnetic Reference Field for 1965.0. Isomagnetic charts drawn for D, H, Z and total force (F) reflect more details than that shown on world magnetic charts. Further, the values of the field at common repeat stations recorded between 1962 and 1974, after eliminating the field values for the epoch 1965.0, are used to get the secular variation as well as its spatial dependence again by means of polynomial which now includes coefficients which are functions of time and of geographical locations. The accuracy of coefficients is tested against the behaviour of secular variation at permanent magnetic observatories. The merits and limitations of the model are discussed

    Influence of mesoscale eddies on a commercial fishery in the coastal waters of the Andaman and Nicobar Islands, India

    Mesoscale eddies enhance the productivity in a stratified coastal environment by upwelling. The seas around the Andaman and Nicobar Islands have been found to have frequent mesoscale eddy activity. Commercial fishing grounds coincide with upwelling areas associated with cyclonic and anticyclonic eddies and also with areas between two adjacent eddies. There are different eddy zones supporting different types of fishing gears and fish. The current study aims at identifying the different zones of mesoscale eddies in the Andaman Sea and compares the productivity and fishing activity in each of them. Data collected from 454 commercial fishing trips in the Andaman Sea along with maps of sea level anomaly and Moderate Resolution Imaging Spectroradiometer (MODIS) global level 3 mapped thermal infrared (IR) daytime sea surface temperature (SST) from the Aqua and Terra satellites were used for the study. Known upwelling areas such as the periphery of anticyclonic and the core of cyclonic eddies showed higher catches in longlines, ring seines among the fishing gears, and among all the fish species groups. Downwelling areas such as the periphery of cyclonic and the core of anticyclonic eddies showed lower catches with ring seines and the fish species groups. Areas in between adjacent eddies were explored in this study and the fish captures in such areas were found to be different with types of fishing and the target fish group. The study shows results that link eddy activity with the performance of a fishery

    Potential conservation of circadian clock proteins in the phylum Nematoda as revealed by bioinformatic searches

    Although several circadian rhythms have been described in C. elegans, its molecular clock remains elusive. In this work we employed a novel bioinformatic approach, applying probabilistic methodologies, to search for circadian clock proteins of several of the best studied circadian model organisms of different taxa (Mus musculus, Drosophila melanogaster, Neurospora crassa, Arabidopsis thaliana and Synechoccocus elongatus) in the proteomes of C. elegans and other members of the phylum Nematoda. With this approach we found that the Nematoda contain proteins most related to the core and accessory proteins of the insect and mammalian clocks, which provide new insights into the nematode clock and the evolution of the circadian system.Fil: Romanowski, Andrés.
