    Approximately 6000 specific DNA deletion events occur during development of the somatic macronucleus of the ciliate Tetrahymena. The eliminated Tlr1 element is 13 kb or more in length and has an 825 bp inverted repeat near the rearrangement junctions. A functional analysis of the cis-acting sequences required for Tlr1 rearrangement was performed. A construct consisting of the entire inverted repeat and several hundred base pairs of flanking DNA on each side was rearranged accurately in vivo and displayed junctional variability similar to the chromosomal Tlr1 rearrangement. Thus, 11 kb or more of internal element DNA is not required in cis for DNA rearrangement. A second construct with only 51 bp of Tetrahymena DNA flanking the right junction underwent aberrant rearrangement. Thus, a signal for determination of the Tlr1 junction is located in the flanking DNA, 51 bp or more from the right junction. Within the Tlr1 inverted repeat are 19 bp tandem repeats. A construct with the 19mer repeat region deleted from the right half of the inverted repeat utilized normal rearrangement junctions. Thus, despite its transposon-like structure, Tlr1 is similar to other DNA rearrangements in Tetrahymena in possessing cis-acting sequences outside the deleted DNA

Methods: To examine the relationship between these inflammatory markers and unfavourable outcomes in COVID-19 pneumonia, we carried out an 18-month observational research (February 2021 to August 2022) with 100 COVID-19-positive patients. We gathered demographic information, clinical information, and marker levels. Chi-squared tests and logistic regression were used in the statistical analysis. Results: Patients with COVID-19 pneumonia had elevated levels of CRP, ESR, LDH, ferritin, and D-dimer. Increased levels of these markers were strongly linked to unfavourable results. The odds ratios, which varied from 2.12 to 2.59, showed that patients with increased markers were more likely to experience negative outcomes. Conclusion: The clinical importance of tracking inflammatory markers in COVID-19 pneumonia patients is highlighted by our study, in its conclusion. Elevated levels of CRP, ESR, LDH, ferritin, and D-dimer may act as early markers of illness development and predictors of unfavourable results. During the ongoing COVID-19 epidemic, these markers may help with risk stratification and clinical decision-making, thereby enhancing patient care.     BACKGROUND: We have devised a simple and efficient fluorescence-based method to track antigen uptake and processing in human B lymphoblastoid cells (B-LCL). Fluorescein labelled subtilisin was used to optimize antigen uptake conditions and identify processed peptides from human cell lines. RESULTS: Fluorescein labelled subtilisin conjugates had 0.06 to 2 moles of fluorescein per subtilisin molecule. High performance liquid chromatography and mass spectrometry (NanoESI-LC/MS/MS) analysis identified fluorescein conjugated to K141, K256, and the N terminus. Conjugates retained antigenic specificity to subtilisin specific antibodies and could be processed by whole cell extracts into low molecular weight fragments at pH 5.2. Maximal antigen uptake and processing occurred when PMSF (phenylmethylsulfonyl fluoride) inhibited subtilisin conjugate was incubated with cells at 100–200 μg/ml for 16 to 24 hr. Once optimal uptake conditions were established, processed subtilisin peptides were isolated and identified from human cell lines. CONCLUSION: Our studies show that FITC-conjugation provides an efficient tool to track the uptake and processing of this protease antigen and to facilitate identification of processed antigenic peptides from human cell lines

    Background: In hepatitis B virus (HBV), reverse transcriptase (RT) region of the polymerase P gene and surface S gene (HBsAg) are largely overlapped. Mutations in surface S gene may cause escape variants. In the present study, we aimed to study the prevalence and pattern of the typical HBsAg escape mutations and concomitant nucleos(t)ide analogue resistance mutation patterns in patients with chronic hepatitis B (CHB) in Indian population.Methods: The present observational study was carried out from January 2021 to June 2022 with 156 known cases of CHB infection. Hepatitis B viral load quantitation was done followed by HBV genotyping and drug resistance detection by PCR and sequencing.Results: Out of 156 cases of CHB, HBsAg escape mutations were found in 50 (32.05%) patients. Genotype D was predominant (90%). Median viral load was 4.43×105 copies/ml. Total 128 HBsAg escape mutations of 46 different patterns were observed with overall prevalence of 29.49% (46/156) in CHB infected patients. The most common substitutions were sP127T (16.67%), sA128V (14.74%), sR122K (5.13%), sY134N (3.85%), sK141R (2.56%), sS143L (2.56%) and sT126INST (1.92%). Concomitant RT mutations were detected in 20 (40%) patients. Total 68 (43.59%) RT mutations of 18 different mutation characteristics were found conferring possible or confirmed resistance to nucleos(t)ide analogues.Conclusions: The emergence of drugs resistant mutants with alteration in ‘aa’ determinant of the S protein is of some concern. The development of novel nucleos(t)ide analogues with a high barrier to resistance is warranted.  National surveillance networks should be set up

    In a recent paper, Gordon, Muratov, and Shvartsman studied a partial differential equation (PDE) model describing radially symmetric diffusion and degradation in two and three dimensions. They paid particular attention to the local accumulation time (LAT), also known in the literature as the mean action time, which is a spatially dependent timescale that can be used to provide an estimate of the time required for the transient solution to effectively reach steady state. They presented exact results for three-dimensional applications and gave approximate results for the two-dimensional analogue. Here we make two generalizations of Gordon, Muratov, and Shvartsman’s work: (i) we present an exact expression for the LAT in any dimension and (ii) we present an exact expression for the variance of the distribution. The variance provides useful information regarding the spread about the mean that is not captured by the LAT. We conclude by describing further extensions of the model that were not considered by Gordon,Muratov, and Shvartsman. We have found that exact expressions for the LAT can also be derived for these important extensions..
