    Municipal Boundary Demarcation in South Africa: Processes and Effects on Governance in Traditional Rural Areas

    After democracy in 1994, South Africa underwent a period of reform in order to address inequalities and effect broad social change. As part of this, the Municipal Demarcation Board began determining the locations of local government boundaries in 1998. The traditional communal lands and rural villages were often split by municipal boundaries which failed to follow complex social boundaries. Contest between the government and traditional governance over land resulted in escalation of disputes. A duality in governance in traditional rural areas was thus created and rural communities were severely affected. This research adopts a multiple case study approach to understand disputes over municipal boundary   demarcation. The causes of disputes are investigated and the processes of municipal demarcation and boundary dispute resolution are analysed using the goals of good governance in land administration, rights, restrictions and responsibilities, Kotter’s eight stages of change management and the 7Es performance measurement frameworks. The analysis shows that theprocess of municipal demarcation can be improved and provides proposals which should contribute to reducing boundary demarcation disputes.Keywords:Demarcation, dispute resolution, performance measurement, change management, good governanc

    Surface temperature trends from homogenized time series in South Africa : 1931-2015

    Previous assessments of historical trends of measured surface temperature in South Africa have all shown a general upward trend, in both mean and extreme values, over recent decades. In addition, some regional differences in trends have been identified. Most of these studies focused on the period from about 1961 up to the last year that could be included before publication, as only climate stations situated in the same position for the entire analysis period were analysed. A data homogenisation procedure enabled the combination of time series of stations from which trend analysis could be applied, extending the common analysis period for this study back to around 1931. The trend results, based on the WMO ETCCDI indices, continue to show the general warming trend shown in previous analyses, with a general increase in extreme warm events, and a general decrease in extreme cold events across South Africa. The analysis of seasonal trends show that, while there are noteworthy differences on a regional basis, austral summer shows on average the strongest warming, followed by autumn, winter and spring. The central interior, which exhibited significant cooling in previous analyses, now shows non-significant or similar trends when compared to the other parts of South Africa. There is no countrywide acceleration in the warming trends, but some regional consistencies in the temporal changes in trends could be determined, i.e. increases in trends in the central interior and decreases in trends along most of the coastal region.http://onlinelibrary.wiley.com/journal/10.1002/(ISSN)1097-00882018-04-30hb2017Geography, Geoinformatics and Meteorolog

    Evaluation of the anti-mycobacterial, anti-oxidative and anti-proliferative activities of Rubia cordifolia ethanolic leaf extract sub-fractions in human lymphocytes and neutrophils

    Background.  Rubia cordifolia has been used to treat various diseases and exhibits antimicrobial, anti-inflammatory, analgesic and antidiarrheal activities. The plant has not been investigated for its anti-mycobacterial activity against virulent tuberculosis strains as well as cytotoxic and anti-oxidant activities in mononuclear cells and neutrophils.Methods. Ethanolic and dichloromethane leaf extract fractions of Rubia cordifolia (0.2- 125ÎŒg/ml) were screened for anti-mycobacterial activity using a fluorescent microplate assay. The anti-oxidant activity of the fractions was tested using the radical scavenging assay, while their cytotoxicity to lymphocytes was measured using the WST-8 assay. Reactive oxygen species production by neutrophils exposed to fractions was tested using Cell Meterℱ Fluorimetric ROS Assay kit.Results. The ethanolic leaf fractions exhibited overall superior anti-mycobacterial activity than the dichloromethane group and were further screened for anti-oxidant and cytotoxic activities. Ethanolic sub-fractions K2F-3.2, K2F-3.3 and K2F-3.4 showed significant antioxidant activities at concentration of 50 ÎŒg/ml to 200 ÎŒg/ml and higher in both the cell free-based radical scavenging activity and total reactive oxygen species production assays. For lymphocytes, the test agents showed anti-proliferative activity at 25 ÎŒg/ml to 200 ÎŒg/ml for sub-fraction K2F-3.3B and 50 ÎŒg/ml to 200 ÎŒg/ml for agents K2F-3.2 and K2F-3.4.Conclusion. The current study is the first to record the in vitro anti-mycobacterial, anti-oxidant and cytotoxic activities of Rubia cordifolia plant leaf extract sub-fractions using the given experimental setups and further research activities to identify the bioactive components are to be pursued.Keywords: Rubia cordifolia, DPPH, Mycobacteria, cell proliferation, antioxidant activity, RO

    Extracellular secretion of a recombinant therapeutic peptide by Bacillus halodurans utilizing a modified flagellin type III secretion system

    <p>Abstract</p> <p>Background</p> <p>Through modification of the flagellin type III secretion pathway of <it>Bacillus halodurans </it>heterologous peptides could be secreted into the medium as flagellin fusion monomers. The stability of the secreted monomers was significantly enhanced through gene-targeted inactivation of host cell extracellular proteases. In evaluating the biotechnological potential of this extracellular secretion system an anti-viral therapeutic peptide, Enfuvirtide, was chosen. Currently, Enfuvirtide is synthesised utilizing 106 chemical steps. We used Enfuvirtide as a model system in an effort to develop a more cost-effective biological process for therapeutic peptide production.</p> <p>Results</p> <p>An attempt was made to increase the levels of the fusion peptide by two strategies, namely strain improvement through gene-targeted knock-outs, as well as vector and cassette optimization. Both approaches proved to be successful. Through chromosomal inactivation of the <it>spo0A, lytC </it>and <it>lytE </it>genes, giving rise to strain <it>B. halodurans </it>BhFDL05S, the secretion of recombinant peptide fusions was increased 10-fold. Cassette optimization, incorporating an expression vector pNW33N and the N- and C-terminal regions of the flagellin monomer as an in-frame peptide fusion, resulted in a further 3.5-fold increase in the secretion of recombinant peptide fusions.</p> <p>Conclusions</p> <p>The type III flagellar secretion system of <it>B. halodurans </it>has been shown to successfully secrete a therapeutic peptide as a heterologous flagellin fusion. Improvements to both the strain and expression cassette led to increased levels of recombinant peptide, showing promise for a biotechnological application.</p

    Historical rainfall trends in South Africa : 1921–2015

    The aim of this study is to update the analysis of historical rainfall trends with reference to the work from previous studies, through optimizing the highest spatial resolution with the longest possible period of analysis, i.e., 1921–2015. Two interlinked datasets, namely the district rainfall and individual rainfall stations datasets were used for the trend analyses, namely, daily time series of 60 individual rainfall stations and the daily district rainfall of 88 of 94 rainfall districts in South Africa. The extreme precipitation indices defined by the World Meteorological Organization Expert Team on Climate Change Detection and Indices were applied. In general, the results show an increase in rainfall for most rainfall stations in the southern interior of South Africa, and indications of decreases in rainfall in the far northern and north-eastern parts. The increase in the annual rainfall in the south is reflected in the seasonal trends, where summer rainfall shows a similar increase, but also extends into the central interior. For other seasons, most of the country shows no significant historical trends in annual total rainfall. From the extreme rainfall analyses, an increase in daily rainfall extremes in the southern to western interior is apparent. Also, most of the country experienced increases in the intensity of daily rainfall, which confirms global results in general. Decreases in rainfall from wet spells were noted in most places over the east and north-east, while the southern and eastern parts along the escarpment experienced shorter annual dry spells. This study improves on previous studies in the region by more than doubling the analysis period, largely eliminating the influence that decadal-scale cycles might have on analyses over shorter periods. However, some differences in the trend results compared to previous studies are apparent, e.g., less pronounced drying in the east and the previously observed increase in rainfall in the western and southern interior not extending as far as the south-western Cape.     Identifying biologically meaningful hot-weather events using threshold temperatures that affect life-history

    Increases in the frequency, duration and intensity of heat waves are frequently evoked in climate change predictions. However, there is no universal definition of a heat wave. Recent, intense hot weather events have caused mass mortalities of birds, bats and even humans, making the definition and prediction of heat wave events that have the potential to impact populations of different species an urgent priority. One possible technique for defining biologically meaningful heat waves is to use threshold temperatures (T thresh ) above which known fitness costs are incurred by species of interest. We set out to test the utility of this technique using T thresh values that, when exceeded, affect aspects of the fitness of two focal southern African bird species: the southern pied babbler Turdiodes bicolor (T thresh = 35.5°C) and the common fiscal Lanius collaris (T thresh = 33°C). We used these T thresh values to analyse trends in the frequency, duration and intensity of heat waves of magnitude relevant to the focal species, as well as the annual number of hot days (maximum air temperature > T thresh ), in north-western South Africa between 1961 and 2010. Using this technique, we were able to show that, while all heat wave indices increased during the study period, most rapid increases for both species were in the annual number of hot days and in the maximum intensity (and therefore intensity variance) of biologically meaningful heat waves. Importantly, we also showed that warming trends were not uniform across the study area and that geographical patterns in warming allowed both areas of high risk and potential climate refugia to be identified. We discuss the implications of the trends we found for our focal species, and the utility of the T thresh technique as a conservation tool

    Examining the Involvement of School Governing Bodies in Promoting Educators in South Africa

    The place of school governing bodies in the promotion of educators in South African public schools is crucial. This study focuses on the perspective of the school governing body as seen as the contributors to challenges that are experienced by educators during promotion processes and ineffectiveness of the school system. A quantitative method was employed to collect data from ten schools in ILembe District, South Africa. Random sampling was used to select 180 respondents. The collected data was analysed using SPSS, version 25.The findings of the study show that although there are educational policy guidelines that are made available to guide schools on how the processes of promotion are conducted, schools continue to experience challenges with some stakeholders who do not keep within their roles. Several factors such as incompetence of the school governing body, lack of training of the SGBs, illiteracy of the SGBs, political influence, and interference of the union members were found to be stumbling blocks in pursuing fair processes when dealing with promotions. The research recommends that the Department of Education (DoE) take a leading role in appointing independent bodies to deal with promotions in order to mitigate corruption and abuse of power by stakeholders.Business Managemen

    Cyclodextrin-Based Nanofibers and Membranes: Fabrication, Properties and Applications

    Cyclodextrin (CD)-based electrospun nanofibers have become critical role players in the water treatment arena due to their high porosities, small diameters, high surface area-to-volume ratio and other unique properties they exhibit. Investigations demonstrate that nanofibers containing CD molecules can be facially blended with other polymeric species and/or photocatalytic and magnetic nanoparticles to enhance their rates of adsorption, inclusion complexation and selective photodegradation. These properties make them excellent candidates for the removal of water pollutants. On the other hand, the electrospinning process has become the method of choice in the fabrication of various types of CD nanofibrous mats due to its versatility, cost-effectiveness and its potential for the mass production of uniform nanofibers. CDs and CD-derivatives have also found application in membrane technologies, particularly in mixed matrix and thin film composite membranes. CD-blended membranes display improved performances in terms of selectivity, rejection, permeation and flux with reduced fouling propensities and can be used for drinking water purification and removal of emerging micropollutants. This chapter critically reviews CD-based electrospun nanofibers looking at their production, characterization methods and various applications. The use of CDs as membrane materials and how they can be fully explored in water treatment are also investigated
