    Implication de MEK1 et MEK2 dans la morphogenĂšse du placenta de souris

    Le gĂ©nome des mammifĂšres contient deux gĂšnes ERK/MAP kinase kinase, soient Mek1 (Map2k1) et Mek2 (Map2k2), qui codent pour des enzymes responsables de l’activation de ERK1/2. Chez la souris, la perte de fonction de Mek1 engendre une mortalitĂ© embryonnaire, tandis que les mutants Mek2 survivent sans aucun phĂ©notype apparent. Afin d’élucider les fonctions potentielles associĂ©es Ă  MEK2 durant l’embryogenĂšse, la perte de Mek2 a Ă©tĂ© Ă©tudiĂ©e en prĂ©sence d’une haplo-insuffisance de Mek1. La majoritĂ© des embryons Mek1+/-Mek2+/- meurent durant la gestation due Ă  des dĂ©fauts placentaires affectant les tissus extraembryonnaires. Ainsi, bien que Mek1 joue un rĂŽle prĂ©dominant, ces rĂ©sultats mettre en lumiĂšre l’implication de Mek2 durant le dĂ©veloppement du placenta. La caractĂ©risation histologique des placentas Mek1+/-Mek2+/- a rĂ©vĂ©lĂ© une diminution de la vascularisation et la formation aberrante de cellules trophoblastiques gĂ©antes multinuclĂ©Ă©es (MTG). Des expĂ©riences gĂ©nĂ©tiques de traçage cellulaire in vivo ont dĂ©montrĂ© que les cellules MTG dĂ©rivent d’une diffĂ©renciation aberrante des SynT-II et que leur formation dĂ©coule en partie d’un effet cellule autonome. Un second objectif de cette thĂšse Ă©tait de mieux caractĂ©riser le ou les types cellulaires nĂ©cessitant une activation de la voie ERK/MAPK essentielle au dĂ©veloppement placentaire. Des analyses gĂ©nĂ©tiques et histologiques ont dĂ©montrĂ© que la formation des cellules MTG rĂ©sulte d’une fusion ectopique entre les deux couches de SynT qui participent normalement de façon indĂ©pendante Ă  la barriĂšre hĂ©matoplacentaire. La barriĂšre hĂ©matoplacentaire est constituĂ©e d’une double couche de SynT et de cellules dĂ©rivĂ©es de l’allantoĂŻs, soient les cellules endothĂ©liales et leurs pĂ©ricytes. La dĂ©lĂ©tion d’un allĂšle supplĂ©mentaire de Mek1 dans l’allantoĂŻs des mutants Mek1+/-Mek2+/- augmente la pĂ©nĂ©trance et l’expressivitĂ© du phĂ©notype placentaire. De plus, les expĂ©riences d’ablation cellulaire in vivo ont permis de dĂ©montrer que le dĂ©veloppement des SynT-I en une fine couche de cellules multinuclĂ©Ă©es dĂ©pend de la prĂ©sence de la deuxiĂšme couche de SynT. Finalement, par approche de gĂšnes candidats et par analyse de biopuces dans les placentas mutants Mek1Mek2, nous avons montrĂ© une expression dĂ©rĂ©gulĂ©e de cibles potentielles de ERK1/2 impliquĂ©es dans la dĂ©terination, la polaritĂ© et la fusion cellulaire, ce qui contribue Ă  la comprĂ©hension du phĂ©notype observĂ©.The mammalian genome contains two ERK/MAP kinase kinase genes, Mek1 and Mek2, which encode dual-specificity kinases responsible for ERK/MAP kinase activation. In the mouse, the loss of Mek1 function causes embryonic lethality, whereas Mek2 mutants survive with a normal lifespan, suggesting that Mek1 rescues the lack of Mek2 function. The first objective of my thesis was to clarify potential functions of Mek2 during mouse embryogenesis. To do, I have analyzed the loss of Mek2 function in the presence of Mek1 haploinsufficiency. Most Mek1+/-Mek2+/- embryos die during gestation from placenta defects affecting extra-embryonic tissues. Thus, even though Mek1 plays a predominant role, these results enlightened the function of Mek2 in placenta development. The histological characterization of Mek1+/-Mek2+/- placentas revealed a diminution of the vascularization and an aberrant formation of multinucleated trophoblast giant (MTG) cells. Genetic experiments on the SynT-II cellular lineage in vivo demonstrated that MTG cells derive from the aberrant SynT-II differentiation and that their formation results from a cell-autonomous effect. The second objective of my thesis was to determine in which cell types the ERK/MAPK activation is essential for placenta development. Genetic analyses combined with histological studies revealed that MTG formation resulted from the ectopic fusion between both layers of SynT, which normally participate in an independent way in the blood-placental barrier. The blood-placental barrier is constituted of a double layer of SynT and by the cells derived from the allantois, the endothelial cells and their perycites. The deletion of both Mek1 alleles in allantois-derived tissues in a Mek1+/-Mek2+/- placenta environment increases the penetrance and the expressivity of the MTG phenotype. These results demonstrate the role of the ERK/MAPK pathway in defined embryonic and extraembryonic cell populations for correct placenta formation. Using mouse genetics, we also demonstrated that the normal development of syncytiotrophoblasts type I into a thin layer of multinucleated cells depends on the presence of the syncytiotrophoblasts type II. Finally, the combined mutations of Mek1 and Mek2 genes alter the expression of several genes involved in cell fate specification, cell fusion and cell polarity that likely explain the underdeveloped placenta and the MTG phenotype seen in Mek1Mek2 mutants

    Ce mĂ©moire de maĂźtrise a pour but d'explorer le mĂ©tier de youtubeur, tout en s'intĂ©ressant au rapport au travail de ces crĂ©ateurs. Au fil des annĂ©es, l'engouement envers la plateforme YouTube a Ă©tĂ© fulgurant, allant mĂȘme jusqu'Ă  la mise sur pied d'un programme partenaire, afin de permettre aux crĂ©ateurs de commencer Ă  ĂȘtre rĂ©munĂ©rĂ©s pour les vidĂ©os qu'ils mettaient en ligne. ParallĂšlement, les parcours professionnels sont de plus en plus atypiques et plusieurs jeunes semblent attirĂ©s par le travail autonome et mĂȘme, par l'entrepreneuriat (Pennel, 2013; Noiseux, 2012; Longo et al., 2019). Comme la plateforme YouTube est accessible dans plusieurs pays, des chercheurs de partout dans le monde se sont intĂ©ressĂ©s Ă  ce mĂ©dium, surtout sous l'angle de son utilisation et de sa consommation par les individus. Toutefois, peu d'entre eux se sont penchĂ©s sur ces travailleurs, d'un point de vue orientation, perspective de carriĂšre et rapport au travail. Pour cette recherche qualitative, une recherche documentaire sur la plateforme YouTube a Ă©tĂ© menĂ©e. Ensuite, douze youtubeurs ont Ă©tĂ© rencontrĂ©s lors d'entrevues individuelles, conduites de façon semi-dirigĂ©e. L'analyse de ces donnĂ©es a permis de rĂ©diger une description du travail des youtubeurs, puis de dĂ©gager quatre profils types, soit 1) le youtubeur de carriĂšre, 2) de youtubeur Ă  entrepreneur, 3) le youtubeur et ses conciliations et 4) l'aspirant youtubeur. Ces quatre profils types ont pu ĂȘtre comparĂ©s en regard des six ethos du travail songĂ©s par Mercure et Vultur (2010). Dans la discussion, il sera possible d'affirmer qu'ĂȘtre youtubeur peut reprĂ©senter une activitĂ© professionnalisante, un travail pouvant apporter des revenus aux crĂ©ateurs. La notion d'indĂ©pendance et de dĂ©pendance envers le travail sur plateforme sera aussi explicitĂ©e. Finalement, des rĂ©flexions seront abordĂ©es sur le mĂ©tier de youtubeur, exercĂ© dans le contexte singulier de la pandĂ©mie de COVID-19.This master's thesis aims to explore the profession of youtubers, while focusing on the relationship between these creators and their work. Over the years, the popularity of the YouTube platform has been soaring, even going so far as to set up a partner program to allow creators to start earning money for the videos they put online. At the same time, career paths are increasingly atypical and many young people seem attracted to self-employment and even entrepreneurship (Pennel, 2013; Noiseux, 2012; Longo et al., 2019). As the YouTube platform is accessible in several countries, researchers from around the world have been interested in this medium, especially in terms of its use and consumption by individuals. However, few of them have studied these workers from the point of view of guidance sciences, career perspective and relationship to work. For this qualitative research, a literature search on the YouTube platform was conducted. Then, twelve youtubers were met during individual interviews, in a semi-structured manner. The analysis of these data allowed us to write a description of the youtubers' work, and then to identify four typical profiles : 1) the career youtuber, 2) from youtuber to entrepreneur, 3) the youtuber and his conciliations, and 4) the aspiring youtuber. These four typical profiles could be compared against the six work ethos thought by Mercure and Vultur (2010). In the discussion, it will be possible to affirm that being a youtuber can represent a professionalizing activity, a job that can bring income to creators. The notion of independence and dependence towards platform work will also be explored. Finally, reflections will be addressed on the profession of youtuber, exercised in the singular context of the COVID-19 pandemic

    This study examines teachers’ perceptions of gifted students in their class, in a Quebec school service centre, and the influence of personal and contextual variables. The sample consists of 21 primary school teachers (18 women and 3 men). After the lexical analysis carried out with the Alceste software, four specific classes of statements emerge from the teachers’ discourse. In addition, age, teaching cycle, type of class, number of years of experience, involvement in an intervention plan and experience with giftedness predict the heterogeneity of the perceptions of teachers towards giftedness. The results suggest more traditional and stereotypical perceptions of giftedness by teachers.     Compromised cerebrovascular regulation and cerebral oxygenation in pulmonary arterial hypertension

    Background : Functional cerebrovascular regulatory mechanisms are important for maintaining constant cerebral blood flow and oxygen supply in heathy individuals and are altered in heart failure. We aim to examine whether pulmonary arterial hypertension (PAH) is associated with abnormal cerebrovascular regulation and lower cerebral oxygenation and their physiological and clinical consequences. Methods and Results : Resting mean flow velocity in the middle cerebral artery mean flow velocity in the middle cerebral artery (MCAvmean); transcranial Doppler), cerebral pressure‐flow relationship (assessed at rest and during squat‐stand maneuvers; analyzed using transfer function analysis), cerebrovascular reactivity to CO2, and central chemoreflex were assessed in 11 patients with PAH and 11 matched healthy controls. Both groups also completed an incremental ramp exercise protocol until exhaustion, during which MCAvmean, mean arterial pressure, cardiac output (photoplethysmography), end‐tidal partial pressure of CO2, and cerebral oxygenation (near‐infrared spectroscopy) were measured. Patients were characterized by a significant decrease in resting MCAvmean (P<0.01) and higher transfer function gain at rest and during squat‐stand maneuvers (both P<0.05). Cerebrovascular reactivity to CO2 was reduced (P=0.03), whereas central chemoreceptor sensitivity was increased in PAH (P<0.01), the latter correlating with increased resting ventilation (R2=0.47; P<0.05) and the exercise ventilation/CO2 production slope (Embedded Image slope; R2=0.62; P<0.05) during exercise for patients. Exercise‐induced increases in MCAvmean were limited in PAH (P<0.05). Reduced MCAvmean contributed to impaired cerebral oxygen delivery and oxygenation (both P<0.05), the latter correlating with exercise capacity in patients with PAH (R2=0.52; P=0.01). Conclusions : These findings provide comprehensive evidence for physiologically and clinically relevant impairments in cerebral hemodynamic regulation and oxygenation in PAH

    17ÎČ-Estradiol and estrogen receptor α protect right ventricular function in pulmonary hypertension via BMPR2 and apelin

    Women with pulmonary arterial hypertension (PAH) exhibit better right ventricular (RV) function and survival than men; however, the underlying mechanisms are unknown. We hypothesized that 17ÎČ-estradiol (E2), through estrogen receptor α (ER-α), attenuates PAH-induced RV failure (RVF) by upregulating the procontractile and prosurvival peptide apelin via a BMPR2-dependent mechanism. We found that ER-α and apelin expression were decreased in RV homogenates from patients with RVF and from rats with maladaptive (but not adaptive) RV remodeling. RV cardiomyocyte apelin abundance increased in vivo or in vitro after treatment with E2 or ER-α agonist. Studies employing ER-α–null or ER-ÎČ–null mice, ER-α loss-of-function mutant rats, or siRNA demonstrated that ER-α is necessary for E2 to upregulate RV apelin. E2 and ER-α increased BMPR2 in pulmonary hypertension RVs and in isolated RV cardiomyocytes, associated with ER-α binding to the Bmpr2 promoter. BMPR2 is required for E2-mediated increases in apelin abundance, and both BMPR2 and apelin are necessary for E2 to exert RV-protective effects. E2 or ER-α agonist rescued monocrotaline pulmonary hypertension and restored RV apelin and BMPR2. We identified what we believe to be a novel cardioprotective E2/ER-α/BMPR2/apelin axis in the RV. Harnessing this axis may lead to novel RV-targeted therapies for PAH patients of either sex

    Asthma, rhinitis, and atopic dermatitis (AD) are interrelated clinical phenotypes that partly overlap in the human interactome. The concept of “one-airway-one-disease,” coined over 20 years ago, is a simplistic approach of the links between upper- and lower-airway allergic diseases. With new data, it is time to reassess the concept. This article reviews (i) the clinical observations that led to Allergic Rhinitis and its Impact on Asthma (ARIA), (ii) new insights into polysensitization and multimorbidity, (iii) advances in mHealth for novel phenotype definitions, (iv) confirmation in canonical epidemiologic studies, (v) genomic findings, (vi) treatment approaches, and (vii) novel concepts on the onset of rhinitis and multimorbidity. One recent concept, bringing together upper- and lower-airway allergic diseases with skin, gut, and neuropsychiatric multimorbidities, is the “Epithelial Barrier Hypothesis.” This review determined that the “one-airway-one-disease” concept does not always hold true and that several phenotypes of disease can be defined.     La voie de signalisation cellulaire ERK/MAPK (extracellular signal-regulated kinase/mitogen-activated protein kinase) est impliquĂ©e dans plusieurs fonctions cellulaires. Chez la souris, l’invalidation de gĂšnes codant pour des membres de cette voie est souvent associĂ©e Ă  une mort embryonnaire consĂ©cutive Ă  un dĂ©veloppement anormal du placenta. Ce dernier est essentiel aux Ă©changes nutritionnels et gazeux entre les circulations maternelle et embryonnaire, ainsi qu’à l’évacuation des dĂ©chets mĂ©taboliques. Ces Ă©changes s’effectuent sans contact direct entre les deux circulations. Chez la souris, la barriĂšre hĂ©matoplacentaire est constituĂ©e d’une triple couche de cellules trophoblastiques et de cellules endothĂ©liales de l’embryon. En effet, Ă  qui attribuer un crime lorsque la structure engage des actions collectives (bataillons, foules, organisations rĂ©sistantes) ou que les actes commis ont Ă©tĂ© ordonnĂ©s par des supĂ©rieurs hiĂ©rarchiques ? Comment prendre en considĂ©ration Ie fait que le contexte de guerre peut-ĂȘtre perturbant ou traumatisant pour les soldats, sans pour autant en venir Ă  excuser les actes commis ? Et pourtant, il faut arriver Ă  proposer des solutions, car ces questions revĂȘtent une importance capitale, non seulement au sein des procĂšs internationaux, mais aussi dans le contexte gĂ©nĂ©ral d'aprĂšs-guerre. L'exactitude des inculpations, la visibilitĂ© des mĂ©thodes juridiques et des procĂ©dures sont autant d'Ă©lĂ©ments qui acquiĂšrent une portĂ©e particuliĂšre dans les efforts pour installer une paix aussi durable que possible.
