    Konsep Pemikiran Pendidikan Islam Kontemporer di Indonesia

    Munculnya fenomena paham keislaman yang amat beragam, tentunya memilki latar belakang kemunculannya, batasan dan ciri-ciri dari masing-masing gerakan pemikiran Islam yang muncul di Indonesia. Masing-masing mempunyai corak dan wajah yang cukup beragama. Dari corak tersebut perlu diketahui dan dipelajari. Adapun jenis penelitian ini adalah kajian pustaka dengan menggunakan pendekatan sosiologi Agama dan sejarah yang akan mengungkap beberapa konsep pemikiran Islam kontemporer di Indonesia. Perkembangan pemikiran islam di Indonesia semakin berkembang dengan lahirnya beberapa tokoh-tokoh pemikirin islam diantaranya K. H. Ahmad Dahlan, Abdur Rahman Wahid (Gus Dur), Nur Khalid Majid, Fazlur Rahman, Muslim Hassan Hanafi, Adonis, Mahmud Muhammad Thaha, Nash Hamid Abu Zaid, Muhammad Syahrur, dengan berbagai pemikiran yang berbeda-beda. Adapun perkembangan dan konsep pemikiran Islam di Indonesia ada empat (4) yaitu: 1. Pemikiran Islam Fundamentali. 2. Neo Tradisionalis. 3. Neo Modernis. dan 4. Liberalis.Kata Kunci: Konsep Pemikiran Islam, Pendidikan Kontemporer The emergence of the phenomenon of Islamic understanding are very diverse, of course, have the background of its emergence, extent and characteristics of each movement of Islamic thought which emerged in Indonesia. The type of this research is literature review using a sociological approach to religion and history that will reveal some of the concepts of contemporary Islamic thought in Indonesia.     Metode yang digunakan dalam penelitian ini adalah metode deskriptif kualitatif. Penelitian deskriptif merupakan penelitian yang bertujuan menggambarkan kedudukan akal dalam islam. Sedangkan sumbernya diambil dari beberapa buku-buku ilmiah yang diterbitkan dengan maksud dijadikan sumber referensi Adapun Tekhnik pengumpulan data dalam penelitian ini ialah Studi Kepustakaan, Wawancara dan Observasi. Pembahasan di dalam ontologi di mulai tanpa asumsi dasar, melainkan mengandalkan kreativitas akal yaitu inspirasi, intuisi, dan ilham. Metode abstraksi digunakan ontologi untuk mencari kejelasaan tentang dunia fakta seluruhnya sampai pada pengertian fundamental. Pengetahuan fundamental yang dihasilkan oleh ontologi dapat dijadikan dasar untuk membahas kembali asumsi dasar yang oleh ilmu pengetahuan telah dianggap mapan kebenarannya.Kata Kunci: Ontologi dan Ilmu PengetahuanAbstract The method used in this study is a qualitative descriptive method. Descriptive research is a study that aims to describe the position of reason in Islam. While the source is taken from several scientific books published with the intention of being used as a source of reference. The data collection techniques in this study are Literature Studies, Interviews and Observations. The discussion in ontology starts without basic assumptions, but relies on resourceful creativity, namely inspiration, intuition, and inspiration. Abstraction methods are used ontologies to find clarity about the world of facts all arrive at a fundamental understanding. Fundamental knowledge produced by ontology can be used as a basis for revisiting the basic assumptions that science has assumed are established

    Jenis penelitian yang digunakan adalah deskriptif kualitatif. Populasi dalam penelitian ini adalah semua siswa dan guru PAI sebanyak 104. Adapun teknik yang digunakan adalah teknik puporsife sampling yaitu sampel diambil secara langsung atau sengaja yakni siswa kelas 2,4,5 sebanyak 53 orang . selanjutnya pengumpulan data melalui penelitian kepustakaan dan lapangan yang terdiri dari teknik observasi, wawancara, angket dan catatan dokumentasi. Adapun teknik analisis data yang digunakan adalah dengan cara deskriftif kualitatif. Dari hasil penelitian menunjukkan bahwa implementasi kurikulum 2013 dalam pembelajaran Pendidikan Agama Islam yang berkaitan dengan pemahaman guru dalam membuat perangkat pembelajaran di SDN.16 Tanrutedong Kabupaten Sidrap sudah berjalan dengan baik dilihat dari pemahaman guru sudah mampu membuat perangkat pembelajaran seperti silabus, rencana pelaksanaan pembelajaran(RPP) dan memahami rubrik-rubrik penilaian peserta didik. Adapun sikap peserta didik terhadap pembelajaran Pendidikan Agama Islam terbilang sangat baik dilihat dari perilaku sehari-hari siswa dan dapat dilihat dari hasil kusioner tentang sikap siswa dalam mengikuti pembelajaran Pendidikan Agama Islam.     Double-Stranded RNA Attenuates the Barrier Function of Human Pulmonary Artery Endothelial Cells

    Circulating RNA may result from excessive cell damage or acute viral infection and can interact with vascular endothelial cells. Despite the obvious clinical implications associated with the presence of circulating RNA, its pathological effects on endothelial cells and the governing molecular mechanisms are still not fully elucidated. We analyzed the effects of double stranded RNA on primary human pulmonary artery endothelial cells (hPAECs). The effect of natural and synthetic double-stranded RNA (dsRNA) on hPAECs was investigated using trans-endothelial electric resistance, molecule trafficking, calcium (Ca2+) homeostasis, gene expression and proliferation studies. Furthermore, the morphology and mechanical changes of the cells caused by synthetic dsRNA was followed by in-situ atomic force microscopy, by vascular-endothelial cadherin and F-actin staining. Our results indicated that exposure of hPAECs to synthetic dsRNA led to functional deficits. This was reflected by morphological and mechanical changes and an increase in the permeability of the endothelial monolayer. hPAECs treated with synthetic dsRNA accumulated in the G1 phase of the cell cycle. Additionally, the proliferation rate of the cells in the presence of synthetic dsRNA was significantly decreased. Furthermore, we found that natural and synthetic dsRNA modulated Ca2+ signaling in hPAECs by inhibiting the sarco-endoplasmic Ca2+-ATPase (SERCA) which is involved in the regulation of the intracellular Ca2+ homeostasis and thus cell growth. Even upon synthetic dsRNA stimulation silencing of SERCA3 preserved the endothelial monolayer integrity. Our data identify novel mechanisms by which dsRNA can disrupt endothelial barrier function and these may be relevant in inflammatory processes

    Mutation in NSUN2, which Encodes an RNA Methyltransferase, Causes Autosomal-Recessive Intellectual Disability

    Causes of autosomal-recessive intellectual disability (ID) have, until very recently, been under researched because of the high degree of genetic heterogeneity. However, now that genome-wide approaches can be applied to single multiplex consanguineous families, the identification of genes harboring disease-causing mutations by autozygosity mapping is expanding rapidly. Here, we have mapped a disease locus in a consanguineous Pakistani family affected by ID and distal myopathy. We genotyped family members on genome-wide SNP microarrays and used the data to determine a single 2.5 Mb homozygosity-by-descent (HBD) locus in region 5p15.32–p15.31; we identified the missense change c.2035G>A (p.Gly679Arg) at a conserved residue within NSUN2. This gene encodes a methyltransferase that catalyzes formation of 5-methylcytosine at C34 of tRNA-leu(CAA) and plays a role in spindle assembly during mitosis as well as chromosome segregation. In mouse brains, we show that NSUN2 localizes to the nucleolus of Purkinje cells in the cerebellum. The effects of the mutation were confirmed by the transfection of wild-type and mutant constructs into cells and subsequent immunohistochemistry. We show that mutation to arginine at this residue causes NSUN2 to fail to localize within the nucleolus. The ID combined with a unique profile of comorbid features presented here makes this an important genetic discovery, and the involvement of NSUN2 highlights the role of RNA methyltransferase in human neurocognitive development

    ALG-2 and peflin regulate COPII targeting and secretion in response to calcium signaling

    ER-to-Golgi transport is the first step in the constitutive secretory pathway, which, unlike regulated secretion, is believed to proceed nonstop independent of Ca2+ flux. However, here we demonstrate that penta-EF hand (PEF) proteins ALG-2 and peflin constitute a hetero-bifunctional COPII regulator that responds to Ca2+ signaling by adopting one of several distinct activity states. Functionally, these states can adjust the rate of ER export of COPII-sorted cargos up or down by ∼50%. We found that at steady-state Ca2+, ALG-2/peflin hetero-complexes bind to ER exit sites (ERES) through the ALG-2 subunit to confer a low, buffered secretion rate, while peflin-lacking ALG-2 complexes markedly stimulate secretion. Upon Ca2+ signaling, ALG-2 complexes lacking peflin can either increase or decrease the secretion rate depending on signaling intensity and duration—phenomena that could contribute to cellular growth and intercellular communication following secretory increases or protection from excitotoxicity and infection following decreases. In epithelial normal rat kidney (NRK) cells, the Ca2+-mobilizing agonist ATP causes ALG-2 to depress ER export, while in neuroendocrine PC12 cells, Ca2+ mobilization by ATP results in ALG-2-dependent enhancement of secretion. Furthermore, distinct Ca2+ signaling patterns in NRK cells produce opposing ALG-2-dependent effects on secretion. Mechanistically, ALG-2-dependent depression of secretion involves decreased levels of the COPII outer shell and increased peflin targeting to ERES, while ALG-2-dependent enhancement of secretion involves increased COPII outer shell and decreased peflin at ERES. These data provide insights into how PEF protein dynamics affect secretion of important physiological cargoes such as collagen I and significantly impact ER stress
