    Social isolation and energy metabolism in rat hippocampus

    El aislamiento social puede entenderse como una forma de deprivación sensorial. Se sabe que ambientes enriquecidos o complejos estimularmente afectan al SNC tanto a un nivel anatómico como fisiológico. En nuestro estudio, se usaron dos grupos de ratas, uno formado por animales aislados durante 30 días desde el final de la lactancia y un grupo control, que permaneció en grupos de tres animales durante el mismo período. Se analizó en los animales el metabolismo oxidativo cerebral del hipocampo (areas CA1, CA3 y giro dentado) mediante histoquímica para la citocromo c oxidasa (CO). Los resultados muestran un incremento significativo de la actividad CO en todas las regiones estudiadas en el grupo aislado, con diferencias entre las areas hipocampales en ambos grupos. Se discute la sensibilidad de la histoquímica de la CO en el estudio del posible papel activador del estrés en los animales aislado

    Lethal iron deprivation induced by non-neutralizing antibodies targeting transferrin receptor 1 in malignant B cells

    A number of antibodies have been developed that induce lethal iron deprivation (LID) by targeting the transferrin receptor 1 (TfR1/CD71) and either neutralizing transferrin (Tf) binding, blocking internalization of the receptor and/or inducing its degradation. We have developed recombinant antibodies targeting human TfR1 (ch128.1 and ch128.1Av), which induce receptor degradation and are cytotoxic to certain malignant B-cells. We now show that internalization of TfR1 bound to these antibodies can lead to its sequestration and degradation, as well as reduced Tf uptake, and the induction of a transcriptional response consistent with iron deprivation, which is mediated in part by downstream targets of p53. Cells resistant to these antibodies do not sequester and degrade TfR1 after internalization of the antibody/receptor complex, and accordingly maintain their ability to internalize Tf. These findings are expected to facilitate the rational design and clinical use of therapeutic agents targeting iron import via TfR1 in hematopoietic malignancies.Fil: Rodríguez, José A..     In this work we present a low-cost and flexible 2×2 MIMO testbed operating on the ISM band of 2.4 GHz with a total bandwith of 20 MHz. The transmitter and receiver baseband modules are PCs equipped with ADCs, DACs, memories, DSPs and FPGAs modules that allow a data transfer rate up to 200 MB/s per channel. Signals are generated at an IF of 15 MHz, upconverted to RF by two signal generators and downconverted to 15 MHz by means of two circuits specifically designed for this platform

    Spatiotemporal Dynamics of Grasslands Using Landsat Data in Livestock Micro-Watersheds in Amazonas (NW Peru)

    In Peru, grasslands monitoring is essential to support public policies related to the identification, recovery and management of livestock systems. In this study, therefore, we evaluated the spatial dynamics of grasslands in Pomacochas and Ventilla micro-watersheds (Amazonas, NW Peru). To do this, we used Landsat 5, 7 and 8 images and vegetation indices (normalized difference vegetation index (NDVI), enhanced vegetation index (EVI) and soil adjusted vegetation index (SAVI). The data were processed in Google Earth Engine (GEE) platform for 1990, 2000, 2010 and 2020 through random forest (RF) classification reaching accuracies above 85%. The application of RF in GEE allowed surface mapping of grasslands with pressures higher than 85%. Interestingly, our results reported the increase of grasslands in both Pomacochas (from 2457.03 ha to 3659.37 ha) and Ventilla (from 1932.38 ha to 4056.26 ha) micro-watersheds during 1990–2020. Effectively, this study aims to provide useful information for territorial planning with potential replicability for other cattle-raising regions of the country. It could further be used to improve grassland management and promote semi-extensive livestock farming

    La leucemia linfocítica crónica (LLC) es la leucemia crónica menos frecuente en México. En consideración a los avances recientes, a una mejor clasificación pronóstica y a la introducción de nuevas modalidades terapéuticas, la Agrupación Mexicana para el Estudio de la Hematología organizó el primer consenso mexicano en leucemia linfocítica crónica. Este consenso se llevó a cabo en Cancún, Quintana Roo, México, en Septiembre del 2007. Los objetivos de esta reunión fueron actualizar y compartir los conocimientos de la enfermedad entre los especialistas mexicanos, con el fin de mejorar el diagnóstico y el pronóstico de la LLC en México.     Machine learning and natural language processing (NLP) approach to predict early progression to first-line treatment in real-world hormone receptor-positive (HRþ)/HER2-negative advanced breast cancer patients.

Esta versión tiene Licencia Creative Commons CC-BY-NC-NDBackground: CDK4/6 inhibitors plus endocrine therapies are the current standard of care in the first-line treatment of HRþ/HER2-negative metastatic breast cancer, but there are no well-established clinical or molecular predictive factors for patient response. In the era of personalised oncology, new approaches for developing predictive models of response are needed. Materials and methods: Data derived from the electronic health records (EHRs) of real-world patients with HRþ/HER2-negative advanced breast cancer were used to develop predictive models for early and late progression to first-line treatment. Two machine learning approaches were used: a classic approach using a data set of manually extracted features from reviewed (EHR) patients, and a second approach using natural language processing (NLP) of freetext clinical notes recorded during medical visits. Results: Of the 610 patients included, there were 473 (77.5%) progressions to first-line treatment, of which 126 (20.6%) occurred within the first 6 months. There were 152 patients (24.9%) who showed no disease progression before 28 months from the onset of first-line treatment. The best predictive model for early progression using the manually extracted dataset achieved an area under the curve (AUC) of 0.734 (95% CI 0.687e0.782). Using the NLP free-text processing approach, the best model obtained an AUC of 0.758 (95% CI 0.714 e0.800). The best model to predict long responders using manually extracted data obtained an AUC of 0.669 (95% CI 0.608e0.730). With NLP free-text processing, the best model attained an AUC of 0.752 (95% CI 0.705e0.799). Conclusions: Using machine learning methods, we developed predictive models for early and late progression to first-line treatment of HRþ/HER2-negative metastatic breast cancer, also finding that NLP-based machine learning models are slightly better than predictive models based on manually obtained data

    Autoantibodies against the immunodominant sCha epitope discriminate the risk of sudden death in chronic Chagas cardiomyopathy

    In Chagas disease (ChD) caused by Trypanosoma cruzi, new biomarkers to predict chronic cardiac pathology are urgently needed. Previous studies in chagasic patients with mild symptomatology showed that antibodies against the immunodominant R3 epitope of sCha, a fragment of the human basic helix-loop-helix transcription factor like 5, correlated with cardiac pathology. To validate sCha as a biomarker and to understand the origin of anti-sCha antibodies, we conducted a multicenter study with several cohorts of chagasic patients with severe cardiac symptomatology. We found that levels of antibodies against sCha discriminated the high risk of sudden death, indicating they could be useful for ChD prognosis. We investigated the origin of the antibodies and performed an alanine scan of the R3 epitope. We identified a minimal epitope MRQLD, and a BLAST search retrieved several T. cruzi antigens. Five of the hits had known or putative functions, of which phosphonopyruvate decarboxylase showed the highest cross-reactivity with sCha, confirming the role of molecular mimicry in the development of anti-sCha antibodies.     Repeated pancreatic resection for pancreatic metastases from renal cell Carcinoma: A Spanish multicenter study (PANMEKID)

    Background and objectives: Recurrent isolated pancreatic metastasis from Renal Cell Carcinoma (RCC) after pancreatic resection is rare. The purpose of our study is to describe a series of cases of relapse of pancreatic metastasis from renal cancer in the pancreatic remnant and its surgical treatment with a repeated pancreatic resection, and to analyse the results of both overall and disease -free survival. Methods: Multicenter retrospective study of patients undergoing pancreatic resection for RCC pancreatic metastases, from January 2010 to May 2020. Patients were grouped into two groups depending on whether they received a single pancreatic resection (SPS) or iterative pancreatic resection. Data on short and long-term outcome after pancreatic resection were collected. Results: The study included 131 pancreatic resections performed in 116 patients. Thus, iterative pancreatic surgery (IPS) was performed in 15 patients. The mean length of time between the first pancreatic surgery and the second was 48.9 months (95 % CI: 22.2-56.9). There were no differences in the rate of postoperative complications. The DFS rates at 1, 3 and 5 years were 86 %, 78 % and 78 % vs 75 %, 50 % and 37 % in the IPS and SPS group respectively (p = 0.179). OS rates at 1, 3, 5 and 7 years were 100 %, 100 %, 100 % and 75 % in the IPS group vs 95 %, 85 %, 80 % and 68 % in the SPS group (p = 0.895). Conclusion: Repeated pancreatic resection in case of relapse of pancreatic metastasis of RCC in the pancreatic remnant is justified, since it achieves OS results similar to those obtained after the first resection

    Pancreatic metastases from renal cell carcinoma. Postoperative outcome after surgical treatment in a Spanish multicenter study (PANMEKID)

    Background: Renal Cell Carcinoma (RCC) occasionally spreads to the pancreas. The purpose of our study is to evaluate the short and long-term results of a multicenter series in order to determine the effect of surgical treatment on the prognosis of these patients. Methods: Multicenter retrospective study of patients undergoing surgery for RCC pancreatic metastases, from January 2010 to May 2020. Variables related to the primary tumor, demographics, clinical characteristics of metastasis, location in the pancreas, type of pancreatic resection performed and data on short and long-term evolution after pancreatic resection were collected. Results: The study included 116 patients. The mean time between nephrectomy and pancreatic metastases' resection was 87.35 months (ICR: 1.51-332.55). Distal pancreatectomy was the most performed technique employed (50 %). Postoperative morbidity was observed in 60.9 % of cases (Clavien-Dindo greater than IIIa in 14 %). The median follow-up time was 43 months (13-78). Overall survival (OS) rates at 1, 3, and 5 years were 96 %, 88 %, and 83 %, respectively. The disease-free survival (DFS) rate at 1, 3, and 5 years was 73 %, 49 %, and 35 %, respectively. Significant prognostic factors of relapse were a disease free interval of less than 10 years (2.05 [1.13-3.72], p 0.02) and a history of previous extrapancreatic metastasis (2.44 [1.22-4.86], p 0.01). Conclusions: Pancreatic resection if metastatic RCC is found in the pancreas is warranted to achieve higher overall survival and disease-free survival, even if extrapancreatic metastases were previously removed. The existence of intrapancreatic multifocal compromise does not always warrant the performance of a total pancreatectomy in order to improve survival. (C) 2021 The Authors. Published by Elsevier Ltd
