    Production of functional protein hydrolysates from Egyptian breeds of soybean and lupin seeds

    Enzymatic hydrolysis is an agro-processing aid that can be utilized in order to improve nutritional quality of protein extracts from many sources. In this study, protein extracts from ungerminated and/or germinated local Egyptian soybean and lupin flours were hydrolyzed using the enzyme papain. The hydrolysis processes were carried out for 2 h and aliquots were withdrawn at different time intervals. We have analysed the protein hydrolysate after 30 min hydrolysis as an example of a partially hydrolyzed protein, and after 120 min as an example of greatly hydrolyzed protein. The hydrolysate (2 h treatment at 80°C and pH 7.4) from both soybean and lupin flour contained significantly decreased trypsin inhibitor activity and urease activity, and a reduced phytate content, which improved the overall protein quality. Hydrolysis caused almost complete inactivation of urease in all soybean and lupin samples regardless if the seeds were germinated or not. High protein content, nitrogen solubility and invitro protein digestibility was shown after hydrolysis. Total protein content (in g/100 g extract) increased in hydrolyzed samples from 48.1 to 51-60 for soybean (dependent on pre-treatment) and from36.8 to 39.9-48.6 for lupin. Total essential amino acid content was also increased in papain hydrolyzed samples, compared to that in raw and germinated legumes. More specifically, the amount of lysine,sulphur amino acids, histidine, and to a certain extent isoleucine and threonine increased in samples from both legume species. All soybean samples exhibited antioxidant activity while in lupin samples,only those subjected to hydrolysis showed activity.     Towards 100 % recycling of reclaimed asphalt in road surface courses: binder design methodology and case studies

    Reclaimed Asphalt (RA) has shown great potential to be reused in new asphalt mixtures, however its incorporation in top asphalt pavement layers is still very limited (10-30%). In fact, despite the advantages that its use implies, RA content in road pavement surface courses is still restricted in most countries due to mainly legislation limitations, but also some technical issues. This paper aims at being a step further to improve the latter by providing a methodology that allows producing fundamental inputs for confidently performing mix design of asphalt mixtures incorporating up to 100% RA. The methodology consists in an advanced preliminary binder’s blend design that can be used with any type of RA and also in presence of rejuvenators. This procedure includes in the production of blending charts and laws that considers the uncertainties on accounting the extent of final binder content, Degree of Blending and Replaced Virgin Binder. The description of the methodology is accompanied with results of two extreme case studies consisting in the preliminary design of binders for asphalt mixtures with high content of two types of RA corresponding to extreme cases: the short-term aged RA (STA-RA), having a very soft residual binder (Pen> 20dmm) and the long-term aged RA, having a much harder residual binder (Pen <10dmm). As a result, the proposed methodology allowed assessing the feasibility of using up to 90% of RA and determining whether the use of rejuvenating agents was needed

    Exploring the Universe with Metal-Poor Stars

    The early chemical evolution of the Galaxy and the Universe is vital to our understanding of a host of astrophysical phenomena. Since the most metal-poor Galactic stars (with metallicities down to [Fe/H]\sim-5.5) are relics from the high-redshift Universe, they probe the chemical and dynamical conditions of the Milky Way and the origin and evolution of the elements through nucleosynthesis. They also provide constraints on the nature of the first stars, their associated supernovae and initial mass function, and early star and galaxy formation. The Milky Way's dwarf satellites contain a large fraction (~30%) of the known most metal-poor stars that have chemical abundances that closely resemble those of equivalent halo stars. This suggests that chemical evolution may be universal, at least at early times, and that it is driven by massive, energetic SNe. Some of these surviving, ultra-faint systems may show the signature of just one such PopIII star; they may even be surviving first galaxies. Early analogs of the surviving dwarfs may thus have played an important role in the assembly of the old Galactic halo whose formation can now be studied with stellar chemistry. Following the cosmic evolution of small halos in simulations of structure formation enables tracing the cosmological origin of the most metal-poor stars in the halo and dwarf galaxies. Together with future observations and additional modeling, many of these issues, including the reionization history of the Milky Way, may be constrained this way. The chapter concludes with an outlook about upcoming observational challenges and ways forward is to use metal-poor stars to constrain theoretical studies.Comment: 34 pages, 11 figures.     Effects of fire-fighting on a fully developed compartment fire: temperatures and emissions

    This study evaluates the effects and consequences of fire-fighting operations on the main characteristics of a fully-developed compartment fire. It also presents data and evaluation of the conditions to which fire-fighters are exposed. A typical room enclosure was used with ventilation through a corridor to the front access door. The fire load was wooden pallets. Flashover was reached and the fire became fully developed before the involvement of the fire-fighting team. The progression of the fire-fighters through the corridor and the main-room suppression attack - in particular the effect of short, medium and long water pulses on either the hot gas layer or the fire seat - was charted against the compartment temperatures, heat release rates, oxygen levels and toxic species concentrations. The fire fighting team was exposed to extreme conditions, heat fluxes in excess of 35 kW/m2 and temperatures of the order of 250 oC even at crouching level. The fire equivalence ratio showed rich burning with high toxic emissions in particular of CO and unburnt hydrocarbons very early in the fire history and a stabilisation of the equivalence ratio at about 1.8. The fire fighting operations made the combustion temporarily richer and the emissions even higher

    Population Pharmacokinetic Modelling of FE 999049, a Recombinant Human Follicle-Stimulating Hormone, in Healthy Women After Single Ascending Doses

    OBJECTIVE: The purpose of this analysis was to develop a population pharmacokinetic model for a novel recombinant human follicle-stimulating hormone (FSH) (FE 999049) expressed from a human cell line of foetal retinal origin (PER.C6(®)) developed for controlled ovarian stimulation prior to assisted reproductive technologies.METHODS: Serum FSH levels were measured following a single subcutaneous FE 999049 injection of 37.5, 75, 150, 225 or 450 IU in 27 pituitary-suppressed healthy female subjects participating in this first-in-human single ascending dose trial. Data was analysed by nonlinear mixed effects population pharmacokinetic modelling in NONMEM 7.2.0.RESULTS: A one-compartment model with first-order absorption and elimination rates was found to best describe the data. A transit model was introduced to describe a delay in the absorption process. The apparent clearance (CL/F) and apparent volume of distribution (V/F) estimates were found to increase with body weight. Body weight was included as an allometrically scaled covariate with a power exponent of 0.75 for CL/F and 1 for V/F.CONCLUSIONS: The single-dose pharmacokinetics of FE 999049 were adequately described by a population pharmacokinetic model. The average drug concentration at steady state is expected to be reduced with increasing body weight

    Pipamperone Population Pharmacokinetics Related to Effectiveness and Side Effects in Children and Adolescents

    Background: Pipamperone is a frequently prescribed antipsychotic in children and adolescents in the Netherlands, Belgium, and Germany. However, pediatric pharmacokinetics and the relationship with side effects and efficacy are unknown. Currently, divergent pediatric dosing recommendations exist. Objectives: The objective of this study was to describe the population pharmacokinetics of pipamperone in children and adolescents; to correlate measured and predicted pipamperone trough concentrations and predicted 24-h area under the curves with effectiveness, extrapyramidal symptoms, and sedation; and to propose dose recommendations based on simulations. Methods: Pipamperone concentrations were collected from Dutch pediatric patients in a prospective naturalistic trial (n = 8), and German pediatric patients in a therapeutic drug monitoring service (n = 22). A total of 70 pipamperone concentrations were used to develop a population pharmacokinetic model with non-linear mixed-effects modeling (NONMEM®). Additionally, an additional random sample of 21 German patients with 33 pipamperone concentrations from the same therapeutic drug monitoring service was used for external validation. Pharmacokinetic parameters were related to clinical improvement, sedation, and extrapyramidal symptoms. Simulations were performed to determine optimal dosages. Results: In a one-compartment model, the apparent volume of distribution was 416 L/70 kg and the apparent clearance was 22.1 L/h/70 kg. Allometric scaling was used to correct for differences in bodyweight. The model was successfully externally validated. The median [25th–75th percentile] measured pipamperone trough concentrations were numerically higher in responders (98.0 µg/L [56.0–180.5 µg/L]) than in non-responders (58.0 µg/L [14.9–105.5 µg/L]), although non-significant (p = 0.14). A twice-daily 0.6-mg/kg dosage was better than a fixed dosage to attain the concentration range observed in responders. Conclusions: Our findings suggest that pipamperone therapeutic reference ranges may be lower for children with behavioral problems than recommended for adults with psychotic symptoms (100–400 µg/L).     Flexural behaviour of hot-finished high strength steel square and rectangular hollow sections

    High strength steels, considered in the context of the structural Eurocodes, as steels with a yield strength over 460 MPa, are gaining increasing attention from structural engineers and researchers owing to their potential to enable lighter and more economic structures. This paper focuses on the bending strength of hot-finished high strength steel (HSS) square and rectangular hollow sections; the results of detailed experimental and numerical studies are presented and structural design rules for HSS cross-sections are proposed. A total of 22 in-plane bending tests, in three-point bending and four-point bending configurations, on HSS sections in grades S460 and S690 were conducted. The experimental results were replicated by means of non-linear finite element modelling. Upon validation of the finite element models, parametric studies were performed to assess the structural response of HSS sections over a wider range of cross-section slenderness, cross-section aspect ratio and moment gradient. The experimental results combined with the obtained numerical results were used to assess the suitability of the current European (EN 1993-1-1 and EN 1993-1-12) cross-section classification limits for HSS structural components.     A data mining approach for classifying DNA repair genes into ageing-related or non-ageing-related

    <p>Abstract</p> <p>Background</p> <p>The ageing of the worldwide population means there is a growing need for research on the biology of ageing. DNA damage is likely a key contributor to the ageing process and elucidating the role of different DNA repair systems in ageing is of great interest. In this paper we propose a data mining approach, based on classification methods (decision trees and Naive Bayes), for analysing data about human DNA repair genes. The goal is to build classification models that allow us to discriminate between ageing-related and non-ageing-related DNA repair genes, in order to better understand their different properties.</p> <p>Results</p> <p>The main patterns discovered by the classification methods are as follows: (a) the number of protein-protein interactions was a predictor of DNA repair proteins being ageing-related; (b) the use of predictor attributes based on protein-protein interactions considerably increased predictive accuracy of attributes based on Gene Ontology (GO) annotations; (c) GO terms related to "response to stimulus" seem reasonably good predictors of ageing-relatedness for DNA repair genes; (d) interaction with the XRCC5 (Ku80) protein is a strong predictor of ageing-relatedness for DNA repair genes; and (e) DNA repair genes with a high expression in T lymphocytes are more likely to be ageing-related.</p> <p>Conclusions</p> <p>The above patterns are broadly integrated in an analysis discussing relations between Ku, the non-homologous end joining DNA repair pathway, ageing and lymphocyte development. These patterns and their analysis support non-homologous end joining double strand break repair as central to the ageing-relatedness of DNA repair genes. Our work also showcases the use of protein interaction partners to improve accuracy in data mining methods and our approach could be applied to other ageing-related pathways.</p
