    A low-cost airborne platform for ecological monitoring

    This work documents the development of an aerial environmental monitoring platform based on a paramotor, dubbed robofoil. Significant advantages are achieved in safety, durability, ease of use and flexibility by employing an inflated wing. The aircraft is easy to fly, has near vertical ascent into wind and an intrinsic fail-safe. The ability to control the wing angle of attack and interchange wings according to weather or mission requirements makes this platform truly flexible. With an onboard autopilot and manual override, the vehicle is intuitive to fly and has a short learning curve for the user. With flight speeds ranging from 0 to 40 knots, the vehicle is well-suited to targeted surveillance as well as being resilient to gusty conditions. With a high payload capability, the platform can carry fuel for flights in excess of an hour in the current version. We have established that it is possible to use genetic programming, a machine learning technique, to evolve application-specific systems purely through training.     Severe loss-of-function mutations in the adrenocorticotropin receptor (ACTHR, MC2R) can be found in patients diagnosed with salt-losing adrenal hypoplasia

    Objective: Familial glucocorticoid deficiency type I (FGD1) is a rare form of primary adrenal insufficiency resulting from recessive mutations in the ACTH receptor (MC2R, MC2R). Individuals with this condition typically present in infancy or childhood with signs and symptoms of cortisol insufficiency, but disturbances in the renin-angiotensin system, aldosterone synthesis or sodium homeostasis are not a well-documented association of FGD1. As ACTH stimulation has been shown to stimulate aldosterone release in normal controls, and other causes of hyponatraemia can occur in children with cortisol deficiency, we investigated whether MC2R changes might be identified in children with primary adrenal failure who were being treated for mineralocorticoid insufficiency. Design: Mutational analysis of MC2R by direct sequencing. Patients: Children (n = 22) who had been diagnosed with salt-losing forms of adrenal hypoplasia (19 isolated cases, 3 familial), and who were negative for mutations in DAX1 (NR0B1) and SF1 (NR5A1). Results: MC2R mutations were found in three individuals or kindred (I: homozygous S74I; II: novel compound heterozygous R146H/560delT; III: novel homozygous 579-581delTGT). These changes represent severely disruptive loss-of-function mutations in this G-protein coupled receptor, including the first reported homozygous frameshift mutation. The apparent disturbances in sodium homeostasis were mild, manifest at times of stress (e.g. infection, salt-restriction, heat), and likely resolved with time. Conclusions: MC2R mutations should be considered in children who have primary adrenal failure with apparent mild disturbances in renin-sodium homeostasis. These children may have been misdiagnosed as having salt-losing adrenal hypoplasia. Making this diagnosis has important implications for treatment, counselling and long-term prognosi

    A catchment-scale irrigation systems model for sugarcane Part 1: Model development

    In South Africa, the demand for water exceeds available supplies in many catchments. In order to justify existing water requirements and to budget and plan in the context of growing uncertainty regarding water availability, a model to assist in the assessment and management of catchment water supply and demand interactions, and the associated impacts on theprofitability of irrigated sugarcane, has been developed. The model, ACRUCane, operates as a submodel within the ACRU agrohydrological model and simulates the water budget of a field of irrigated sugarcane. The water budget is based on the integration of several widely accepted algorithms and concepts, accounts for different irrigation system types performing at different levels of uniformity and different water management strategies. Furthermore, it can simulate a wide variety ofwater availability scenarios and constraints through its link with ACRU simulated hydrology. The crop yield algorithms used in the model were verified using data from three different irrigation trials with widely varying irrigation treatments, where the model was shown to adequately distinguish the impacts of different watering strategies on crop yields.     Monitoring of offshore geological carbon storage integrity: Implications of natural variability in the marine system and the assessment of anomaly detection criteria

    The design of efficient monitoring programmes required for the assurance of offshore geological storage requires an understanding of the variability and heterogeneity of marine carbonate chemistry. In the absence of sufficient observational data and for extrapolation both spatially and seasonally, models have a significant role to play. In this study a previously evaluated hydrodynamic-biogeochemical model is used to characterise carbonate chemistry, in particular pH heterogeneity in the vicinity of the sea floor. Using three contrasting regions, the seasonal and short term variability are analysed and criteria that could be considered as indicators of anomalous carbonate chemistry identified. These criteria are then tested by imposing a number of randomised DIC perturbations on the model data, representing a comprehensive range of leakage scenarios. In conclusion optimal criteria and general rules for developing monitoring strategies are identified. Detection criteria will be site specific and vary seasonally and monitoring may be more efficient at periods of low dynamics. Analysis suggests that by using high frequency, sub-hourly monitoring anomalies as small as 0.01 of a pH unit or less may be successfully discriminated from natural variability – thereby allowing detection of small leaks or at distance from a leakage source. Conversely assurance of no leakage would be profound. Detection at deeper sites is likely to be more efficient than at shallow sites where the near bed system is closely coupled to surface processes. Although this study is based on North Sea target sites for geological storage, the model and the general conclusions are relevant to the majority of offshore storage sites lying on the continental shelf

    Extremely Halophilic Archaeal Communities are Resilient to Short‐Term Entombment in Halite

    Some haloarchaea avoid the harsh conditions present in evaporating brines by entombment in brine inclusions within forming halite crystals, where a subset of haloarchaea survives over geological time. However, shifts in the community structure of halite‐entombed archaeal communities remain poorly understood. Therefore, we analysed archaeal communities from in situ hypersaline brines collected from Trapani saltern (Sicily) and their successional changes in brines versus laboratory‐grown halite over 21 weeks, using high‐throughput sequencing. Haloarchaea were dominant, comprising >95% of the archaeal community. Unexpectedly, the OTU richness of the communities after 21 weeks was indistinguishable from the parent brine and overall archaeal abundance in halite showed no clear temporal trends. Furthermore, the duration of entombment was less important than the parent brine from which the halite derived in determining the community composition and relative abundances of most genera in halite‐entombed communities. These results show that halite‐entombed archaeal communities are resilient to entombment durations of up to 21 weeks, and that entombment in halite may be an effective survival strategy for near complete communities of haloarchaea. Additionally, the dominance of ‘halite specialists’ observed in ancient halite must occur over periods of years, rather than months, hinting at long‐term successional dynamics in this environment

    Basal protein phosphatase 2A activity restrains cytokine expression: Role for MAPKs and tristetraprolin

    PP2A is a master controller of multiple inflammatory signaling pathways. It is a target in asthma; however the molecular mechanisms by which PP2A controls inflammation warrant further investigation. In A549 lung epithelial cells in vitro we show that inhibition of basal PP2A activity by okadaic acid (OA) releases restraint on MAPKs and thereby increases MAPK-mediated pro-asthmatic cytokines, including IL-6 and IL-8. Notably, PP2A inhibition also impacts on the anti-inflammatory protein - tristetraprolin (TTP), a destabilizing RNA binding protein regulated at multiple levels by p38 MAPK. Although PP2A inhibition increases TTP mRNA expression, resultant TTP protein builds up in the hyperphosphorylated inactive form. Thus, when PP2A activity is repressed, pro-inflammatory cytokines increase and anti-inflammatory proteins are rendered inactive. Importantly, these effects can be reversed by the PP2A activators FTY720 and AAL(s), or more specifically by overexpression of the PP2A catalytic subunit (PP2A-C). Moreover, PP2A plays an important role in cytokine expression in cells stimulated with TNFα; as inhibition of PP2A with OA or PP2A-C siRNA results in significant increases in cytokine production. Collectively, these data reveal the molecular mechanisms of PP2A regulation and highlight the potential of boosting the power of endogenous phosphatases as novel anti-inflammatory strategies to combat asthmatic inflammation

    Tree migration-rates : narrowing the gap between inferred post-glacial rates and projected rates

    Faster-than-expected post-glacial migration rates of trees have puzzled ecologists for a long time. In Europe, post-glacial migration is assumed to have started from the three southern European peninsulas (southern refugia), where large areas remained free of permafrost and ice at the peak of the last glaciation. However, increasing palaeobotanical evidence for the presence of isolated tree populations in more northerly microrefugia has started to change this perception. Here we use the Northern Eurasian Plant Macrofossil Database and palaeoecological literature to show that post-glacial migration rates for trees may have been substantially lower (60–260 m yr–1) than those estimated by assuming migration from southern refugia only (115–550 m yr–1), and that early-successional trees migrated faster than mid- and late-successional trees. Post-glacial migration rates are in good agreement with those recently projected for the future with a population dynamical forest succession and dispersal model, mainly for early-successional trees and under optimal conditions. Although migration estimates presented here may be conservative because of our assumption of uniform dispersal, tree migration-rates clearly need reconsideration. We suggest that small outlier populations may be a key factor in understanding past migration rates and in predicting potential future range-shifts. The importance of outlier populations in the past may have an analogy in the future, as many tree species have been planted beyond their natural ranges, with a more beneficial microclimate than their regional surroundings. Therefore, climate-change-induced range-shifts in the future might well be influenced by such microrefugia

    We describe a lattice simulation of the masses and decay constants of the lowest-lying vector and axial resonances, and the electroweak S parameter, in an SU(3) gauge theory with Nf=2N_f = 2 and 6 fermions in the fundamental representation. The spectrum becomes more parity doubled and the S parameter per electroweak doublet decreases when NfN_f is increased from 2 to 6, motivating study of these trends as NfN_f is increased further, toward the critical value for transition from confinement to infrared conformality.Comment: 4 pages, 5 figures; to be submitted to PR
