    Effect of organic manures and microbial inoculants on yield, root colonization and total bacterial population in turmeric (Curcuma longa L.) intercropped in arecanut (Areca catechu L.) garden

    The effect of combination of organic manures namely, compost, vermicompost, phosphocompost and mustard cake and microbial inoculants namely, Azospirillum brasilense and arbuscular mycorrhiza (Glomus fasciculatum) were evaluated for the organic production of turmeric (Curcuma longa) (cv. Suguna) grown as intercrop in arecanut (Areca catechu) (cv. Mohitnagar) plantation at Mondouri (Nadia, West Bengal) A significant difference in rhizome yield was noticed when organic manure-microbial inoculant combination was applied when compared with recommended dose of fertilizers (inorganic). Among different treatment combinations tried, the most effective treatment was vermicompost + Azospirillum sp. + Glomus sp. (28.94 t ha-1), followed by compost + Azospirillum sp. + Glomus sp. (26.93 t ha-1), as compared to recommended inorganic NPK (24.11 t ha-1). Maximum root colonisation (74%) with microbial inoculants at 180 days after planting was observed with vermicompost + Azospirillum sp. + Glomus sp. Maximum bacterial population (105.25 x 105 CFU g-1 soil) at harvest was noticed in compost + Azospirillum sp. + Glomus sp., as compared to lowest population with recommended NPK (56.35 x 105 CFU g-1 of soil). &nbsp

    Intra-specific morphological and morphometric variability of Radopholus similis (Cobb, 1893) Thorne, 1949

    All the root inhabiting migratory endoparasitic nematode populations of Radopholus procured from banana crop of Vellayani, Thiruvananthapuram, Kerala were identified as Radopholus similis. Heat killed females were straight to slightly ventrally curved posteriorly. Female’s head was low, rounded, continuous or slightly setoff with the body contour. Females were 500-660 ”m long and were comparatively longer than males. Males had button shaped head set off by a constriction; female with three to five lip annuli, four crenate and areolated lateral incisures, stylet 14-18 ”m long with rounded knobs, vulva post-equatorial (58%), sometimes with slight protuberant lips, ovary paired and equally developed, oesophageal gland overlapped the intestine dorsally, tail elongate-conoid with narrowly rounded terminus. The stylet length (”m), width of stylet knob (”m), distance of excretory pore from anterior end (”m), distance from head to basal bulb (”m), lateral field structure, shape of stylet knob, head region, position of phasmid, tail shape with its terminus, morphometric values like m%, o% and v% and a, c and cÂŽ ratios of females were stable (CV<12%) features. There is an existence of intra-specific variability in the morphological and morphometric features of R. similis. The main morphological diversity was observed with P% of male and female, b ratio of female and stylet length, distance of DEGO from stylet base, o% and T% of male. A high degree of intra-specific morphometric variability was observed with regard to the total body length (”m), body width (”m), stylet length (”m), distance of dosrsal oesophageal gland orifice (DEGO) from stylet base (”m), number of lip annuli, lip height (”m), distance from head to basal bulb (”m), distance of anus from anterior end (”m), tail length (”m), anal body width (”m), distance of phasmid from tail terminus (”m), number of lateral lines, width of lateral field (”m), b ratio and P % among females of R. similis. Morphometric features like m%, o% and v% of females of R. similis showed least variability. These can be considered as the stable morphometric characters for discrimination of females of R. similis. Ratios like ‘a’ and ‘c’ of females of R. similis were found moderately variable. The morphometric feature and of male i.e. distance from head to basal bulb (”m) was found least variable; while number of lip annuli and spicule length (”m) were moderately variable. &nbsp

    Review of Pesticide Urinary Biomarker Measurements from Selected US EPA Children’s Observational Exposure Studies

    Children are exposed to a wide variety of pesticides originating from both outdoor and indoor sources. Several studies were conducted or funded by the EPA over the past decade to investigate children’s exposure to organophosphate and pyrethroid pesticides and the factors that impact their exposures. Urinary metabolite concentration measurements from these studies are consolidated here to identify trends, spatial and temporal patterns, and areas where further research is required. Namely, concentrations of the metabolites of chlorpyrifos (3,5,6-trichloro-2-pyridinol or TCPy), diazinon (2-isopropyl-6-methyl-4-pyrimidinol or IMP), and permethrin (3-phenoxybenzoic acid or 3-PBA) are presented. Information on the kinetic parameters describing absorption and elimination in humans is also presented to aid in interpretation. Metabolite concentrations varied more dramatically across studies for 3-PBA and IMP than for TCPy, with TCPy concentrations about an order of magnitude higher than the 3-PBA concentrations. Temporal variability was high for all metabolites with urinary 3-PBA concentrations slightly more consistent over time than the TCPy concentrations. Urinary biomarker levels provided only limited evidence of applications. The observed relationships between urinary metabolite levels and estimates of pesticide intake may be affected by differences in the contribution of each exposure route to total intake, which may vary with exposure intensity and across individuals

    Genome-Wide Association Study Identifies Chromosome 10q24.32 Variants Associated with Arsenic Metabolism and Toxicity Phenotypes in Bangladesh

    Arsenic contamination of drinking water is a major public health issue in many countries, increasing risk for a wide array of diseases, including cancer. There is inter-individual variation in arsenic metabolism efficiency and susceptibility to arsenic toxicity; however, the basis of this variation is not well understood. Here, we have performed the first genome-wide association study (GWAS) of arsenic-related metabolism and toxicity phenotypes to improve our understanding of the mechanisms by which arsenic affects health. Using data on urinary arsenic metabolite concentrations and approximately 300,000 genome-wide single nucleotide polymorphisms (SNPs) for 1,313 arsenic-exposed Bangladeshi individuals, we identified genome-wide significant association signals (P<5×10−8) for percentages of both monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA) near the AS3MT gene (arsenite methyltransferase; 10q24.32), with five genetic variants showing independent associations. In a follow-up analysis of 1,085 individuals with arsenic-induced premalignant skin lesions (the classical sign of arsenic toxicity) and 1,794 controls, we show that one of these five variants (rs9527) is also associated with skin lesion risk (P = 0.0005). Using a subset of individuals with prospectively measured arsenic (n = 769), we show that rs9527 interacts with arsenic to influence incident skin lesion risk (P = 0.01). Expression quantitative trait locus (eQTL) analyses of genome-wide expression data from 950 individual's lymphocyte RNA suggest that several of our lead SNPs represent cis-eQTLs for AS3MT (P = 10−12) and neighboring gene C10orf32 (P = 10−44), which are involved in C10orf32-AS3MT read-through transcription. This is the largest and most comprehensive genomic investigation of arsenic metabolism and toxicity to date, the only GWAS of any arsenic-related trait, and the first study to implicate 10q24.32 variants in both arsenic metabolism and arsenical skin lesion risk. The observed patterns of associations suggest that MMA% and DMA% have distinct genetic determinants and support the hypothesis that DMA is the less toxic of these two methylated arsenic species. These results have potential translational implications for the prevention and treatment of arsenic-associated toxicities worldwide
