    Mash Test No. 3-10 of a Non-Proprietary, High-Tension Cable Median Barrier for Use in 6H:1V V-Ditch (Test No. MWP-8)

    The Midwest States Pooled Fund Program has been developing a prototype design for a non-proprietary, high-tension cable median barrier for use in a 6H:1V V-ditch. This system incorporates four evenly spaced cables, Midwest Weak Posts (MWP) spaced at 8 to 16 ft (2.4-4.9 m) intervals, and a bolted, tabbed bracket to attach three or four cables to the sides of each post. According to the Manual for Assessing Safety Hardware (MASH) testing matrix for cable barriers installed within a 6H:1V median V-ditch, a series of eight full-scale tests are required to evaluate the safety performance of a system. A ninth test is required to establish the working width for the systems with variable post spacing. Several previous tests have failed due to posts penetrating the occupant compartment. In order to mitigate this behavior, a modified MWP was designed. Test no. MWP-8 was conducted on the modified barrier system, consisting of MWPs with rounded top edges and a Âľ-in. (19-mm) diameter weakening hole at the groundline. This test was conducted according to MASH test designation no. 3-10 and utilized an 1100C passenger car impacting the barrier on a level terrain. The vehicle was contained by the system. Through the initial two vehicle crossover events across the barrier and posts, no floorpan tearing was observed. During the third impact series with the posts, post penetration into the occupant compartment and floorpan tearing was observed. Therefore, test no. MWP-8 was deemed unacceptable

    Bridging of double-stranded breaks by the nonhomologous end-joining ligation complex is modulated by DNA end chemistry

    The nonhomologous end-joining (NHEJ) pathway is the primary repair pathway for DNA double strand breaks (DSBs) in humans. Repair is mediated by a core complex of NHEJ factors that includes a ligase (DNA Ligase IV; L4) that relies on juxtaposition of 3΄ hydroxyl and 5΄ phosphate termini of the strand breaks for catalysis. However, chromosome breaks arising from biological sources often have different end chemistries, and how these different end chemistries impact the way in which the core complex directs the necessary transitions from end pairing to ligation is not known. Here, using single-molecule FRET (smFRET), we show that prior to ligation, differences in end chemistry strongly modulate the bridging of broken ends by the NHEJ core complex. In particular, the 5΄ phosphate group is a recognition element for L4 and is critical for the ability of NHEJ factors to promote stable pairing of ends. Moreover, other chemical incompatibilities, including products of aborted ligation, are sufficient to disrupt end pairing. Based on these observations, we propose a mechanism for iterative repair of DSBs by NHEJ

    DNA Ligase IV Guides End-Processing Choice during Nonhomologous End Joining

    Nonhomologous end joining (NHEJ) must adapt to diverse end structures during repair of chromosome breaks. Here, we investigate the mechanistic basis for this flexibility. DNA ends are aligned in a paired-end complex (PEC) by Ku, XLF, XRCC4, and DNA ligase IV (LIG4); we show by single-molecule analysis how terminal mispairs lead to mobilization of ends within PECs and consequent sampling of more end-alignment configurations. This remodeling is essential for direct ligation of damaged and mispaired ends during cellular NHEJ, since remodeling and ligation of such ends both require a LIG4-specific structural motif, insert1. Insert1 is also required for PEC remodeling that enables nucleolytic processing when end structures block direct ligation. Accordingly, cells expressing LIG4 lacking insert1 are sensitive to ionizing radiation. Cellular NHEJ of diverse ends thus identifies the steps necessary for repair through LIG4-mediated sensing of differences in end structure and consequent dynamic remodeling of aligned ends

    The Effects of Close Companions (and Rotation) on the Magnetic Activity of M Dwarfs

    We present a study of close white dwarf and M dwarf (WD+dM) binary systems and examine the effect that a close companion has on the magnetic field generation in M dwarfs. We use a base sample of 1602 white dwarf -- main sequence binaries from Rebassa et al. to develop a set of color cuts in GALEX, SDSS, UKIDSS, and 2MASS color space to construct a sample of 1756 WD+dM high-quality pairs from the SDSS DR8 spectroscopic database. We separate the individual WD and dM from each spectrum using an iterative technique that compares the WD and dM components to best-fit templates. Using the absolute height above the Galactic plane as a proxy for age, and the H{\alpha} emission line as an indicator for magnetic activity, we investigate the age-activity relation for our sample for spectral types \leqM7. Our results show that early-type M dwarfs (\leqM4) in close binary systems are more likely to be active and have longer activity lifetimes compared to their field counterparts. However, at a spectral type of M5 (just past the onset of full convection in M dwarfs), the activity fraction and lifetimes of WD+dM binary systems becomes more comparable to that of the field M dwarfs. One of the implications of having a close binary companion is presumed to be increased stellar rotation through disk-disruption, tidal effects, or angular momentum exchange. Thus, we interpret the similarity in activity behavior between late-type dMs in WD+dM pairs and late-type field dMs to be due to a decrease in sensitivity in close binary companions (or stellar rotation), which has implications for the nature of magnetic activity in fully-convective stars.     2-Hour Accelerated Diagnostic Protocol to Assess Patients With Chest Pain Symptoms Using Contemporary Troponins as the Only Biomarker

    Objectives The purpose of this study was to determine whether a new accelerated diagnostic protocol (ADP) for possible cardiac chest pain could identify low-risk patients suitable for early discharge (with follow-up shortly after discharge). Background Patients presenting with possible acute coronary syndrome (ACS), who have a low short-term risk of adverse cardiac events may be suitable for early discharge and shorter hospital stays. Methods This prospective observational study tested an ADP that included pre-test probability scoring by the Thrombolysis In Myocardial Infarction (TIMI) score, electrocardiography, and 0 + 2 h values of laboratory troponin I as the sole biomarker. Patients presenting with chest pain due to suspected ACS were included. The primary endpoint was major adverse cardiac event (MACE) within 30 days. Results Of 1,975 patients, 302 (15.3%) had a MACE. The ADP classified 392 patients (20%) as low risk. One (0.25%) of these patients had a MACE, giving the ADP a sensitivity of 99.7% (95% confidence interval [CI]: 98.1% to 99.9%), negative predictive value of 99.7% (95% CI: 98.6% to 100.0%), specificity of 23.4% (95% CI: 21.4% to 25.4%), and positive predictive value of 19.0% (95% CI: 17.2% to 21.0%). Many ADP negative patients had further investigations (74.1%), and therapeutic (18.3%) or procedural (2.0%) interventions during the initial hospital attendance and/or 30-day follow-up. Conclusions Using the ADP, a large group of patients was successfully identified as at low short-term risk of a MACE and therefore suitable for rapid discharge from the emergency department with early follow-up. This approach could decrease the observation period required for some patients with chest pain.     Nonhomologous end-joining (NHEJ) is the main pathway for repair of DNA double-strand breaks (DSBs), the most cytotoxic form of DNA damage resulting from ionizing radiation, chemotherapeutics, and normal cellular processes. The mechanisms that control NHEJ play key roles in development, in immunity, and in response to cancer therapy; however, the current state of knowledge regarding the physical nature of the NHEJ repair process is limited. Here we used super-resolution microscopy to define the organization of NHEJ complexes in cells, showing that long filaments form at either side of the break.     Does Regional Development Influence Sedimentary Blue Carbon Stocks? A Case Study From Three Australian Estuaries

    Mitigating climate change through the reduction of atmospheric CO2 levels is of interest, particularly through maintaining and re-establishing natural ecosystems that act as carbon sinks, such as coastal vegetated habitats or “blue carbon” systems. Here we compare sedimentary blue carbon (C) stocks from 37 sediment cores collected in pristine (n = 13), agricultural (n = 11), and urban (n = 13) estuaries within the same geomorphological region, located on the eastern coast of Australia. The mean estimated C stocks for each carbon system (seagrass, mangrove, and saltmarshes) were 402 ± 78, 830 ± 109, and 723 ± 100 Mg C ha-1, respectively, conservatively estimated up to 3 m depths. Analysis of variance revealed no significant difference between C stocks per area (C ha-1) considering each habitat type and between specific estuaries. However, the total estuarine C stocks were found to be greater with increasing levels of conservation, based on larger areas of blue carbon vegetation. The potential loss of C to the atmosphere from these small regional estuaries are 500,574 ± 118,635 tons of CO2 equivalent (CO2e), based on specific assumptions. The implication of these results are that there are large C stocks in small regional estuaries which supports the protection of blue C systems in developing coastal areas and highlights the uncertainties of the CO2 emissions from potential blue C habitat degradation

    A consensus statement for trauma surgery capacity building in Latin America

    Background Trauma is a significant public health problem in Latin America (LA), contributing to substantial death and disability in the region. Several LA countries have implemented trauma registries and injury surveillance systems. However, the region lacks an integrated trauma system. The consensus conference’s goal was to integrate existing LA trauma data collection efforts into a regional trauma program and encourage the use of the data to inform health policy. Methods We created a consensus group of 25 experts in trauma and emergency care with previous data collection and injury surveillance experience in the LA. region. Experts participated in a consensus conference to discuss the state of trauma data collection in LA. We utilized the Delphi method to build consensus around strategic steps for trauma data management in the region. Consensus was defined as the agreement of ≥ 70% among the expert panel. Results The consensus conference determined that action was necessary from academic bodies, scientific societies, and ministries of health to encourage a culture of collection and use of health data in trauma. The panel developed a set of recommendations for these groups to encourage the development and use of robust trauma information systems in LA. Consensus was achieved in one Delphi round. Conclusions The expert group successfully reached a consensus on recommendations to key stakeholders in trauma information systems in LA. These recommendations may be used to encourage capacity building in trauma research and trauma health policy in the region
