    Five lessons for researchers who want to collaborate with governments and development organisations but avoid the common pitfalls

    The appeal of collaborating with a government agency, or an organisation funded by one, seems obvious. It provides researchers with much needed resources and information, while also offering practitioners and policymakers a way of generating the evidence needed to design better programmes. In practice, however, it's not always easy to make collaborative research work well. Susan Dodsworth and Nic Cheeseman outline some simple lessons for those looking to collaborate while avoiding the common pitfalls.     Isolation, characterization, and genome sequence of the first representative of a novel class within the Chloroflexi that is abundant in some U.S. Great Basin hot springs and may play important roles in N and C cycling

    A thermophilic, facultatively microaerophilic, heterotrophic bacterium, designated strain JAD2, was isolated from sediments of Great Boiling Spring (GBS), an ~80oC, circumneutral hot spring in the Great Basin GB). The strain grew anaerobically on yeast extract or peptone with an optimal growth temperature of 70-75oC. Growth was stimulated by addition of 0.01 atm O2 to the culture vessel headspace, but was inhibited by higher concentrations (0.2 atm). Cells of JAD2 formed non-motile filaments ranging from 10 to \u3e300 μm in length, which typically decreased in length during stationary phase. 16S rRNA gene-targeted pyrotag sequencing and clone library data suggest that close relatives of this isolate are prominent members of the sediment communities in GBS. Shotgun sequencing of the JAD2 genome produced an assembly consisting of ~3.2 Mbp with an average G+C content of 67.3%. Phylogenies inferred from the 16S rRNA gene and predicted amino acid sequences of various conserved proteins indicate that JAD2 is the first cultivated representative of the GAL35 group, a new class within the Chloroflexi. Predicted genes in the draft genome encoding a putative carbon monoxide dehydrogenase (coxMSL), nitrite reductase (nrfHA) and nitrous oxide reductase (nosZ) suggest that this isolate may play important roles in N and C cycling in GBS sediments

    How to hold elections safely and democratically during the COVID-19 pandemic

    The COVID-19 pandemic poses major challenges for those charged with overseeing electoral processes, but the innovative ways in which practitioners are addressing these challenges indicate that elections can be safely held even under pandemic conditions. These innovations also represent opportunities for strengthening electoral practices and making them more resilient to a variety of other risks. This briefing draws on existing experience of elections held during the COVID-19 pandemic and previous health crises to address five areas of vulnerability: inclusive and accountable electoral management, poll worker safeguarding, interinstitutional collaboration, feasible and effective election observation, and the risk of electoral violence. The analyses indicate that there are a large number of things that electoral practitioners can do to hold elections safely under pandemic conditions. Most of these are techniques that have been employed previously in some form, and we caution against the introduction of entirely new and untested approaches at the current time. The most useful innovations are those such as widening poll-worker recruitment, inter-institutional coordination and hybrid election observation that build on existing practice. The analyses also highlight the importance of not losing track of the need to bolster the transparency, accountability and security of electoral practices. Far from there being a trade-off between making elections safe in pandemic conditions and achieving these other aims, we argue that efforts to maximise electoral integrity and to preserve electoral peace will also help to ensure that democratic elections can be safeguarded from the risks associated with COVID-19. Our principal recommendations fall into three categories: those targeted at electoral administrators, those aimed at election observation organisations, and general recommendations that are relevant to administrators, observers and electoral assistance providers

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements

    How does the objective of aid affect its impact on accountability?: evidence from two aid programmes in Uganda

    Recent research indicates that the political impact of aid, including its impact on accountability institutions, is contingent on its objective. This article explains how this occurs. It relies on evidence from two aid programmes in Uganda, one targeted at poverty reduction and one at democratic governance. I argue that the stated objective of aid programmes masks a deeper cause; individual aid managers’ views of what development entails and how it should be pursued.
