    Ascorbate degradation: pathways, products and possibilities

    A role for l-ascorbate as the precursor of several plant compounds adds to its already broad metabolic utility. There are many examples of plant species in which oxalate and l-threonate are formed from l-ascorbate breakdown, and a number of roles have been proposed for this: structural, physiological, and biochemical. On the other hand, the synthesis of l-tartrate from l-ascorbate remains limited to a very few species, amongst which we must be grateful to count the domesticated grapevine Vitis vinifera and its relatives on which wine production is based. Pathways for the degradation of ascorbate were first proposed ~50 years ago and have formed the basis of more recent biochemical and molecular analyses. The present review seeks to summarize some of these findings and to propose opportunities for future research.</p

    Acute and chronic effects of multivitamin/mineral supplementation on objective and subjective energy measures

    Background: Vitamins and minerals play an essential role within many cellular processes including energy production and metabolism. Previously, supplementation with a multivitamin/mineral (MVM) for ≥28 days resulted in improvements to cognition and subjective state. We have also demonstrated shifts in metabolism during cognitively demanding tasks following MVM in females, both acutely and following 8-week supplementation. The current study aimed to assess these effects further in males and females using metabolically challenging exercise and cognitive tasks. Methods: The current randomised, placebo-controlled, parallel groups study investigated the effects of a MVM complex in 82 healthy young (18-35y) exercisers. Subjective ratings and substrate metabolism were assessed during 30 min each of increasingly effortful incremental exercise and demanding cognitive tasks. Assessments took place on acute study days following a single dose (Day 1) of MVM, containing 3 times recommended daily allowance of water-soluble vitamins plus CoQ10, and following 4-week supplementation (Day 28). Results: Energy expenditure (EE) was increased during cognitive tasks following MVM across Day 1 and Day 28, with greater effects in males. In males, MVM also increased carbohydrate oxidation and energy expenditure during exercise across Day 1 and Day 28. In females, mental tiredness was lower during exercise; increases in physical tiredness following 30 min of exercise were attenuated; and stress ratings following cognitive tasks were reduced following MVM. In males, MVM only lowered mental tiredness following 10 min of exercise. These effects were apparent irrespective of day, but effects on mental tiredness were greater on Day 28. Ferritin levels were also higher on Day 28 in those receiving MVM. Conclusion: These findings extend on existing knowledge, demonstrating increased carbohydrate oxidation and increased energy expenditure in males following MVM supplementation for the first time. Importantly, they show modulation of energy expenditure and subjective tiredness following a single dose, providing further evidence for acute effects of MVM. Differential effects in men and women suggest that sex may play an important role in the effects of MVM on energy metabolism and should be considered in future research.     Simulation of Daily and Monthly Stream Discharge from Small Watersheds Using the SWAT Model

    The Soil and Water Assessment Tool (SWAT) was evaluated and parameter sensitivities were determined while modeling daily streamflows in a small central Kentucky watershed over a two-year period. Streamflow data from 1996 were used to calibrate the model and streamflow data from 1995 were used for evaluation. The model adequately predicted the trends in daily streamflow during this period although Nash-Sutcliffe R2 values were –0.04 and 0.19 for 1995 and 1996, respectively. The model poorly predicted the timing of some peak flow values and recession rates during the last half of 1995. Excluding daily peak flow values from August to December improved the daily R2 to 0.15, which was similar to the 1996 daily R2 value. The Nash-Sutcliffe R2 for monthly total flows were 0.58 for 1995 and 0.89 for 1996 which were similar to values found in the literature. Since very little information was available on the sensitivity of the SWAT model to various inputs, a sensitivity analysis/calibration procedure was designed to evaluate parameters that were thought to influence stream discharge predictions. These parameters included, drainage area, slope length, channel length, saturated hydraulic conductivity, and available water capacity. Minimization of the average absolute deviation between observed and simulated streamflows identified optimum values/ranges for each parameter. Saturated hydraulic conductivity, alpha baseflow factor, drainage area, channel length, and channel width were the most sensitive parameters in modeling the karst influenced watershed. The sensitivity analysis process confirmed die trace studies in the karst watershed that a much larger area contributes to streamflow than can be described by the topographic boundaries. Overall, the results indicate that the SWAT model can be an effective tool for describing monthly runoff from small watersheds in central Kentucky that have developed on karst hydrology however calibration data are necessary to account for solution channels draining into or out of the topographic watershed

    Preliminary Understanding of Complexities in Swimming Performance of Common Minnow (Cyprinidae) Taxa

    Understanding swimming performance of native freshwater fishes has implications for ecology, conservation, and management. In particular, this type of information has practical importance for improving the understanding of fish dispersal, occurrence, migration, and invasive potential. The objective of this study was to characterize swimming performance of 2 taxa from the comparatively understudied minnow family (Cyprinidae) and test for potential drivers as a function of total length, sex, habitat, morphology, or some combination. The study assessed Spotfin Shiner (Cyprinella spiloptera; n = 66) and Bluntnose Minnow (Pimephales notatus; n = 24) populations from an ontogenic range of male and female individuals from lentic and lotic habitats in Indiana and Ohio. Akaike information criterion (AIC) model selection identified the most parsimonious linear regression model to predict swimming performance of Spotfin Shiner and Bluntnose Minnow independently. Overall, larger Spotfin Shiners were superior swimmers compared with smaller individuals. In both species, individuals having more streamlined heads and elongated caudal regions were better swimmers. Additionally, Spotfin Shiners that were collected from lotic environments were generally better swimmers than individuals from lentic environments. Models did not recover sex-specific effects in either species—or meaningful total length, or habitat effects, in Bluntnose Minnows. Overall, this study provides evidence of a complex series of swimming performance covariates when assessing or understanding performance. This has implications for aquatic population, assemblage, and community ecology as well as management and conservation efforts

    Assessment of Cloud Screening with Apparent Surface Reflectance in Support of the ICESat-2 Mission

    The separation of cloud and clear scenes is usually one of the first steps in satellite data analysis. Before deriving a geophysical product, almost every satellite mission requires a cloud mask to label a scene as either clear or cloudy through a cloud detection procedure. For clear scenes, products such as surface properties may be retrieved; for cloudy scenes, scientist can focus on studying the cloud properties. Hence the quality of cloud detection directly affects the quality of most satellite operational and research products. This is certainly true for the Ice, Cloud, and land Elevation Satellite-2 (lCESat-2), which is the successor to the ICESat-l. As a top priority mission, ICESat-2 will continue to provide measurements of ice sheets and sea ice elevation on a global scale. Studies have shown that clouds can significantly affect the accuracy of the retrieved results. For example, some of the photons (a photon is a basic unit of light) in the laser beam will be scattered by cloud particles on its way. So instead of traveling in a straight line, these photons are scattered sideways and have traveled a longer path. This will result in biases in ice sheet elevation measurements. Hence cloud screening must be done and be done accurately before the retrievals

    AbstractIn humans, chikungunya virus (CHIKV) infection causes fever, rash, and acute and persisting polyarthralgia/arthritis associated with joint swelling. We report a new CHIKV disease model in adult mice that distinguishes the wild-type CHIKV-LR strain from the live-attenuated vaccine strain (CHIKV-181/25). Although eight-week old normal mice inoculated in the hind footpad developed no hind limb swelling with either virus, CHIKV-LR replicated in musculoskeletal tissues and caused detectable inflammation. In mice deficient in STAT1-dependent interferon (IFN) responses, CHIKV-LR caused significant swelling of the inoculated and contralateral limbs and dramatic inflammatory lesions, while CHIKV-181/25 vaccine and another arthritogenic alphavirus, Sindbis, failed to induce swelling. IFN responses suppressed CHIKV-LR and CHIKV-181/25 replication equally in dendritic cells in vitro whereas macrophages were refractory to infection independently of STAT1-mediated IFN responses. Glycosaminoglycan (GAG) binding may be a CHIKV vaccine attenuation mechanism as CHIKV-LR infectivity was not dependent upon GAG, while CHIKV-181/25 was highly dependent

    Video killed the radio star? The influence of presentation modality on detecting high-stakes, emotional lies

    Purpose. In many contexts in which high-stakes lies occur (such as security settings or the courtroom), observers must evaluate whether the stories they hear are credible. However, little research has evaluated the ability of observers to detect high-stakes lies, nor the influence of the manner in which the deception is presented on judgment accuracy. This study investigated whether the presentation modality of high-stakes lies influences both explicit and implicit deception detection accuracy. Methods. Participants (N = 231) were randomly assigned to one of four presentation modalities: audiovisual, video-only, audio-only, or transcript-only and asked to evaluate the honesty of targets -half of whom were sincere and half deceptive killers -making a plea for the return of a missing relative both explicitly (direct lie/truth decision) and implicitly (via emotional reactions). Results. Overall, explicit deception detection accuracy was slightly above chance (M = 52.5%), and honest pleas were accurately identified at a higher rate than deceptive pleas. Although there were no differences in overall accuracy across modality, observers reading transcripts exhibited a truth bias, which resulted in them detecting truthful pleas at a higher rate than with the other groups. Although explicit accuracy was at the level of chance, implicit reactions indicated that observers were able to unconsciously discern liars from truth-tellers. Conclusions. Despite the high-stakes nature of the lies presented here, they were difficult to detect. Lies presented via written language were missed at a higher rate when assessed using explicit but not implicit judgments

    Decreased Births Among Black Female Adolescents Following School Desegregation

    Although the socioeconomic impact of school desegregation in the U.S. has been well documented, little is known about the health consequences of this policy. The purpose of this study was to quantify the associations between school desegregation and adolescent births among black and white females. We compared the change in prevalence of adolescent births in areas that implemented school desegregation plans in the 1970s with areas that implemented school desegregation plans in other decades, using difference-in-difference methods with 1970 and 1980 Census microdata. School desegregation policy in the U.S. in the 1970s was associated with a significant reduction of 3.2 percentage points in the prevalence of births among black female adolescents between 1970 and 1980. This association was specific to black female adolescents and was not observed among white adolescents

    The association between blood pressure and years of schooling versus educational credentials: Test of the sheepskin effect

    Purpose Attaining a degree may offer greater opportunities for health than years of schooling alone. This study examines whether there is a degree, or “sheepskin”, effect on the association between education and blood pressure. Methods Multivariable-adjusted ordinal and linear regression models assessed associations of years of schooling and degree attainment with systolic and diastolic blood pressure in a sample of 552 adults aged 38–47 years. Results Years of schooling was inversely associated with systolic blood pressure adjusting for age, gender and race (β=−0.4, 95% CL:−0.7,−0.1 mmHg systolic blood pressure/year of schooling). Additional adjustment for mother’s education, childhood verbal intelligence quotient, childhood health and childhood socioeconomic status had minimal impact on effect size (β= −0.3, 95% CI=−0.7,0.0). However, years of schooling was no longer associated with blood pressure in the fully adjusted model which included additional adjustment for degree attained (β=0.0, 95% CL:−0.5, 0.4). In the fully adjusted model (including adjustment for years of schooling), individuals with a graduate degree still had significantly lower systolic blood pressure than HS degree-holders (e.g. β=−9.2, 95% CL:−15.2,−3.2 for graduate vs. high school degree). Findings were similar for diastolic blood pressure. Conclusion The association of years of schooling with blood pressure may be largely due to degree attainment rather than simply the knowledge and skills accumulated due to years of schooling alone

    Within-Session Analysis of Amphetamine-Elicited Rotation Behavior Reveals Differences between Young Adult and Middle-Aged F344/BN Rats with Partial Unilateral Striatal Dopamine Depletion

    Preclinical modeling of Parkinson's disease using 6-hydroxydopamine (6-OHDA) has been valuable in developing and testing therapeutic strategies. Recent efforts have focused on modeling early stages of disease by infusing 6-OHDA into the striatum. The partial DA depletion that follows intrastriatal 6-OHDA is more variable than the near complete depletion following medial forebrain bundle infusion, and behavioral screening assays are not as well characterized in the partial lesion model. We compared relationships between amphetamine-elicited rotation behavior and DA depletion following intrastriatal 6-OHDA (12.5 μg) in 6 month vs. 18 month F344/BN rats, at 2-weeks and 6-weeks post-lesion. We compared the total number of rotations with within-session (bin-by-bin) parameters of rotation behavior as indicators of DA depletion. Striatal DA depletion was greater in the young adult than in the middle-aged rats at 2 weeks but not at 6 weeks post-lesion. The total number of rotations for the whole session and striatal DA depletion did not differ between the two age groups. Regression analysis revealed a greater relationship between within-session parameters of rotation behavior and DA depletion in the middle-aged group than in the young adult group. These results have implications for estimating DA depletion in preclinical studies using rats of different ages
