    Mestrado em Estudos Alem√£esO presente trabalho tem como tema o ensino do Alem√£o nos liceus portugueses, num per√≠odo extremamente conturbado e controverso da hist√≥ria da Alemanha e de Portugal, nomeadamente o tempo do Terceiro Reich e dos primeiros anos do Estado Novo. Este estudo, que partiu da pesquisa, consulta, sele√ß√£o e posterior an√°lise cr√≠tica de legisla√ß√£o e de publica√ß√Ķes peri√≥dicas da √©poca, debru√ßa-se sobre as reformas do sistema educativo portugu√™s, ocorridas de 1926 a 1948, sobre a influ√™ncia que exerceram na elabora√ß√£o dos programas curriculares e sobre posi√ß√£o ocupada pelo Alem√£o no √Ęmbito das l√≠nguas estrangeiras. √Č tamb√©m analisada a evolu√ß√£o dos conte√ļdos tem√°ticos que gradualmente se foram distanciando do estudo da literatura como meio de aquisi√ß√£o de conhecimentos culturais de um povo. Este trabalho pretende tamb√©m p√īr em evid√™ncia o papel desempenhado pelas teorias de ensino, do final do s√©culo XIX e do in√≠cio do s√©culo XX, na escolha das metodologias did√°tico-pedag√≥gicas para o ensino do Alem√£o e na elabora√ß√£o de manuais. Na √ļltima parte deste trabalho, √© dado enfoque √†s atividades desenvolvidas nos liceus para a promo√ß√£o das rela√ß√Ķes luso-alem√£s, no √Ęmbito de uma pol√≠tica de aproxima√ß√£o cultural e de difus√£o ideol√≥gica.The present work aims to study the teaching of German in Portuguese secondary schools during an extremely turbulent and controversial period of German and Portuguese History, namely the time of the Third Reich and the early years of Salazar‚Äôs New State. This dissertation, which was based on the survey, selection and critical analysis of legislation and periodical publications of that time, focuses on the reforms of the Portuguese educational system from 1926 to 1948, as well as and their influence on the development of curricula and on the importance of German as a foreign language in Portuguese schools. The evolution of the topics, which gradually withdrew from the idea of studying literature as a means of acquiring cultural knowledge, is analysed as well. This work also aims to highlight the role played by education theories at the end of the 19th century and early 20th in the choice of methodologies for the teaching of German and the construction of course books. In the last part of this work, focus is given to the activities carried out in secondary schools to promote Portuguese-German relations in the context of a policy of cultural rapprochement and ideological diffusion

    Nutritional and phytochemical composition of Vaccinium padifolium Sm wild berries and radical scavenging activity

    Blueberries have a well-deserved reputation as a potential functional food, supported by studies which have identified and quantified various nutrients and bioactive phytochemicals with known benefits for human diet and health. Wild blueberries have attracted particular attention due to the levels and concentrations of those phytonutrients. This study aims to evaluate for the first time the chemical composition of Madeira Island's endemic Vaccinium padifolium Sm wild berry. Results show that this fruit contains high values of total soluble phenolic content (around 4 g GAE kg-1 FW), as well as significant values of total monomeric anthocyanin content (around 3 g eq. cyanidin kg-1 FW) and DPPH scavenging activity (around 86.72%). Additionally, results reveal that this fruit has water content of about 88% as well as low sugar content (17.98 and 29.73 g kg-1 for glucose and fructose, respectively). Results also confirm that this wild blueberry is a good source of dietary fiber, fat and minerals. The high level of terpenoid compounds stands out in the aroma profile analysis.Ana C.     Caloric restriction rescues yeast cells from alpha-synuclein toxicity through autophagic control of proteostasis

    őĪ-Synuclein (SNCA) is a presynaptic protein that is associated with the pathophysiology of synucleinopathies, including Parkinson's disease. SNCA is a naturally aggregation-prone protein, which may be degraded by the ubiquitin-proteasome system (UPS) and by lysosomal degradation pathways. Besides being a target of the proteolytic systems, SNCA can also alter the function of these pathways further, contributing to the progression of neurodegeneration. Deterioration of UPS and autophagy activities with aging further aggravates this toxic cycle. Caloric restriction (CR) is still the most effective non-genetic intervention promoting lifespan extension. It is known that CR-mediated lifespan extension is linked to the regulation of proteolytic systems, but the mechanisms underlying CR rescue of SNCA toxicity remain poorly understood. This study shows that CR balances UPS and autophagy activities during aging. CR enhances UPS activity, reversing the decline of the UPS activity promoted by SNCA, and keeps autophagy at homeostatic levels.     Establishment of a reliable protocol for gDNA extraction from olive oil.

    With no possibility to reach in quantity the production of countries with large areas of olive orchards, as a small country, Portugal needs to define a strategy for valuing the high quality and specificities of its olive oil. No doubt the focus must be on the valorization of the Portuguese cultivars, the key factor in determining the singularity of the produced olive oils. Fraud detection, as the use of non-Portuguese varieties, is the main aim of varietal and DOP olive oil producers. In this sense, it is mandatory to have tools allowing the control of the varietie(s) giving rise to the olive oil, both in quality and in quantity. One objective of the project Por3O running at University of √Čvora is the establishment of a molecular tool that identifies the varieties used to produce a given olive oil. Ideally, this tool could be further proposed for screening for frauds and to support olive oil certification. However, as PCR-based tool, it requires the availability of genomic DNA (gDNA) with quality enough to be used on fragment amplification. The step related with extraction of high quality gDNA from olive oil samples, which considers gDNA integrity and absence of inhibitory compounds, is often a limiting step in the development of a PCR-based genotyping tools. The establishment of a robust and efficient extraction protocol is thus crucial. In theory, the use of DNA commercial kits has the advantage of a higher reproductivity greatly removing technician expertise biases. Several DNA commercial kits were tested on its capacity to extract gDNA from a commercial blend olive oil and they were compared with an in-house method based on CTAB-based protocol previously published [1]. The different methods were compared in terms of starting volume of oil sample required for extraction, average gDNA concentration, total gDNA extraction yield and efficiency in Single-Sequence Repeats (SSRs) markers amplification. Considering the results achieved, we propose a workflow regarding gDNA extraction to further molecular analysis

    Frost Multidimensional Perfectionism Scale: the portuguese version

    CONTEXTO: A Escala Multidimensional de Perfeccionismo de Frost (FMPS) √© uma das escalas mais usadas em todo o mundo para avaliar o perfeccionismo. OBJETIVO: Analisar as caracter√≠sticas psicom√©tricas da vers√£o portuguesa da FMPS. M√ČTODOS: A amostra foi constitu√≠da por 217 estudantes do ensino superior (178 mulheres). Um subgrupo (n = 166) completou o reteste ap√≥s quatro semanas. RESULTADOS: A consist√™ncia interna da escala mostrou ser elevada (alfa de Cronbach = ,857). As correla√ß√Ķes item-total corrigido variaram entre ,019 e ,548. Os resultados tamb√©m sugeriram uma boa estabilidade temporal da escala, sendo a correla√ß√£o teste-reteste de ,765. Foi realizada a an√°lise das componentes principais com rota√ß√£o Varimax e com base no Scree plot foram extra√≠das duas solu√ß√Ķes fatoriais robustas (quatro e seis fatores). A an√°lise paralela (Monte Carlo PCA) confirmou a solu√ß√£o de seis fatores. A validade concorrente com a escala MPS de Hewitt e Flett foi elevada, assim como a sua capacidade discriminante dos afetos positivos e negativos (Perfil de Estados de Humor - POMS). CONCLUS√ÉO: As duas estruturas fatoriais (quatro e seis fatores) encontradas na vers√£o portuguesa da Escala Multidimensional de Perfeccionismo de Frost replicam os resultados obtidos por diferentes autores, em diferentes amostras e culturas. Esse fato sugere que essa escala √© um instrumento robusto para a avalia√ß√£o do perfeccionismo em v√°rios contextos, cl√≠nicos e de investiga√ß√£o, bem como em estudos transculturais.BACKGROUND: The Frost Multidimensional Perfectionism Scale is one of the most world widely used measures of perfectionism. OBJECTIVE: To analyze the psychometric properties of the Portuguese version of the Frost Multidimensional Perfectionism Scale. METHODS: Two hundred and seventeen (178 females) students from two Portuguese Universities filled in the scale, and a subgroup (n = 166) completed a retest with a four weeks interval. RESULTS: The scale reliability was good (Cronbach alpha = .857). Corrected item-total correlations ranged from .019 to .548. The scale test-retest reliability suggested a good temporal stability with a test-retest correlation of .765. A principal component analysis with Varimax rotation was performed and based on the Scree plot, two robust factorial structures were found (four and six factors). The principal component analyses, using Monte Carlo PCA for parallel analyses confirmed the six factor solution. The concurrent validity with Hewitt and Flett MPS was high, as well as the discriminant validity of positive and negative affect (Profile of Mood Stats-POMS). DISCUSSION: The two factorial structures (of four and six dimensions) of the Portuguese version of Frost Multidimensional Perfectionism Scale replicate the results from different authors, with different samples and cultures. This suggests this scale is a robust instrument to assess perfectionism, in several clinical and research settings as well as in transcultural studies

    Analysis of the importance of applying the concept of experience in tourism products and services as a way to add value to tourism. Its objective is to verify if the dimensions of the tourist experience are applied to tourism products and services marketed, in the city of São Luís, Maranhão, with the object of analysis being the Historical Soiree Project of the Municipal Tourism Secretariat of São Luís, Maranhão, Brazil. Strategy of promotion of tourism in the region based on playful work and interaction with the viewer. The research was characterized as exploratory-descriptive, carried out by means of bibliographic and documentary research and field research, having as a data collection instrument interviews with tourists and visitors from January to February 2019. It discusses the concept of experience in its multiple meanings and how tourist experiences can provide memorable moments for tourists. It highlights the concept of experience tourism or experiential tourism when listed in tourism products and services. It characterizes the Historical Soiree, showing how this action is performed to provide interaction with visitors, aligning the discussions of the dimensions of the experience with the reports of the participants

    Multi-organ NMR metabolomics to assess in vivo overall metabolic impact of cisplatin in mice

    This work describes, to our knowledge, the first NMR metabolomics analysis of mice kidney, liver, and breast tissue in response to cisplatin exposure, in search of early metabolic signatures of cisplatin biotoxicity. Balb/c mice were exposed to a single 3.5 mg/kg dose of cisplatin and then euthanized; organs (kidney, liver, breast tissue) were collected at 1, 12, and 48 h. Polar tissue extracts were analyzed by NMR spectroscopy, and the resulting spectra were studied by multivariate and univariate analyses. The results enabled the identification of the most significant deviant metabolite levels at each time point, and for each tissue type, and showed that the largest metabolic impact occurs for kidney, as early as 1 h post-injection. Kidney tissue showed a marked depletion in several amino acids, comprised in an overall 13-metabolites signature. The highest number of changes in all tissues was noted at 12 h, although many of those recovered to control levels at 48 h, with the exception of some persistently deviant tissue-specific metabolites, thus enabling the identification of relatively longer-term effects of cDDP. This work reports, for the first time, early (1-48 h) concomitant effects of cDDP in kidney, liver, and breast tissue metabolism, thus contributing to the understanding of multi-organ cDDP biotoxicity.publishe
