At least, 202 Questionnaires were collected, but only 187 questionnaires that can be processed

    Embodied perspectives on behavioral cognitive enhancement

    Recent debates about strategies to enhance human cognition concerned mostly pharmacological, environmental and genetic factors, as well as computerized cognitive training paradigms targeting healthy populations. We offer a new perspective on behavioral cognitive enhancement, arguing that embodied cognition represents a productive framework to explain results and to inform new studies aimed at enhancing cognition. Understanding cognitive mechanisms and their time-course through an embodied perspective contributes to our knowledge of brain functioning and its potential. We review two domains: a) physical exercise and b) embodied learning. For each domain, we summarize experimental evidence according to the level of embodiment of the knowledge representations targeted by interventions (i.e., situatedness, embodiment proper, grounding). Future research should integrate embodiment and cognitive enhancement in training paradigms focused on joint cognitive and physical tasks

    RNF168 cooperates with RNF8 to mediate FOXM1 ubiquitination and degradation in breast cancer epirubicin treatment

    The forkhead box M1 (FOXM1) transcription factor has a central role in genotoxic agent response in breast cancer. FOXM1 is regulated at the post-translational level upon DNA damage, but the key mechanism involved remained enigmatic. RNF168 is a ubiquitination E3-ligase involved in DNA damage response. Western blot and gene promoter-reporter analyses showed that the expression level and transcriptional activity of FOXM1 reduced upon RNF168 overexpression and increased with RNF168 depletion by siRNA, suggesting that RNF168 negatively regulates FOXM1 expression. Co-immunoprecipitation studies in MCF-7 cells revealed that RNF168 interacted with FOXM1 and that upon epirubicin treatment FOXM1 downregulation was associated with an increase in RNF168 binding and conjugation to the protein degradation-associated K48-linked polyubiquitin chains. Consistently, RNF168 overexpression resulted in an increase in turnover of FOXM1 in MCF-7 cells treated with the protein synthesis inhibitor cycloheximide. Conversely, RNF168, knockdown significantly enhanced the half-life of FOXM1 in both absence and presence of epirubicin. Using a SUMOylation-defective FOXM1-5x(K>R) mutant, we demonstrated that SUMOylation is required for the recruitment of RNF168 to mediate FOXM1 degradation. In addition, clonogenic assays also showed that RNF168 mediates epirubicin action through targeting FOXM1, as RNF168 could synergise with epirubicin to repress clonal formation in wild-type but not in FOXM1-deficient mouse embryo fibroblasts (MEFs). The physiological relevance of RNF168-mediated FOXM1 repression is further emphasized by the significant inverse correlation between FOXM1 and RNF168 expression in breast cancer patient samples. Moreover, we also obtained evidence that RNF8 recruits RNF168 to FOXM1 upon epirubicin treatment and cooperates with RNF168 to catalyse FOXM1 ubiquitination and degradation. Collectively, these data suggest that RNF168 cooperates with RNF8 to mediate the ubiquitination and degradation of SUMOylated FOXM1 in breast cancer genotoxic response

    Preliminary results in the automated detection of operational modal properties of the Portico Varano in the Camerino Ducal Palace

    Portico Varano in the Ducal Palace of Camerino (Italy) is a renaissance monumental quadriporticus that was damaged by the 2016 Central Italy seismic sequence. Within the field activities for saving cultural heritage foreseen within a recent European research project named ARCH, a long-term monitoring system, comprising different types of sensors, such as accelerometers, displacement transducers, environmental sensors, and a weather station, was set up to achieve comprehensive measures of its operational behaviour and the evolution of the damage. The monitoring system, installed in October 2020, is currently operating, providing valuable information on the experimentally observed dynamic behaviour, also considering changes in the environmental conditions. Starting from the results of the dynamic characterization of the structure and after the optimization of the position of the sensors, this paper shows and discusses the efforts made to track over time the modal characteristics of the Portico Varano in order to detect changes in its conditions. In addition, a procedure has been proposed and implemented combining information available from Italian National Institute of Geophysics and Volcanology (INGV) to identify recorded data related to seismic events relevant to operational conditions

    Telah dilakukan eksperimen untuk menentukan koefisien ekspansi linier batang kuningan dengan teknik ESPI (Electronic Speckle Pattern Interferometry). Objek uji yang digunakan terbuat dari kuningan dengan diameter 45 mm, panjang 44 mm, dan dikenai beban thermal dari temperatur 30°Csampai dengan 40°C menggunakan heater (pemanas) yang ditempelkan erat dibelakang objek. Data citra spekel selama pemanasan diamati dan setiap selang waktu 1 menit disimpan di memori komputer untuk diproses. Pemrosesannya dilakukan dengan cara mensubtraksikan (mengurangkan) ke dua citraspekel pixel per pixel dan hasilnya ditampilkan di layar monitor dalam bentuk pola frinji. Dari eksperimen diperoleh harga koefisien ekspansi linier kuningan α = (1,96 ± 0,02)10-5/°C dengan standard deviasi terhadap rata-rata sebesar 1,02%.An experiment to determine the linear coefficient of expansion for the brass rod have been done by using the ESPI (Electronic Speckle Pattern Interferometry) technique. The specimen we used were brass with diameter 45 mm, length 44 mm, and thermal loaded from temperature 30°C up to 40°C use heaters behind object. The image speckle data for each 1 menit during thermal were record by the computers. Its process done by subtraction to image speckle data pixel by pixel, and the result presented on monitor screen in the fringe pattern. From experiment obtained that the average value of the linear coefficient of expansion for the brass is, α = (1,96 ± 0 ,02)10-5/°C with the deviation from average value is 1.02 %

    Group Psychological Treatment Preferences of Individuals Living With Chronic Disease: Brief Report of a Saskatchewan-Based Cross-Sectional Survey

    Given that individuals with chronic diseases comorbid with psychological distress experience worse clinical outcomes than those without psychological distress, treatment of the psychological sequalae that accompanies chronic diseases is of utmost importance. Thus, the present study aimed to examine group treatment preferences among adults living with chronic disease in Saskatchewan, Canada. An online survey regarding group treatment preferences was administered to 207 participants living with chronic disease comorbid with psychological distress. The most often reported treatment scenario was virtual sessions (45%) lasting 1 h (51%) and occurring every other week (45%) in the evening (63%) for 3 to4 months (40%). Preferences included a medium group (48%), a relatively closed group nature (ie, only occasional new members; 44%), and group leadership including at least 1 professional living with chronic disease (54%). Future-oriented (81%), supportive (83%), skill-based (95%), and group discussions (78%) were desired treatment characteristics among participants. Survey results showed clear preferences on treatment content and session logistics. Slight variations exist by gender and age, but a consensus can be identified and act as a preliminary treatment plan. This study contributes to the body of literature on psychological treatment preferences for individuals living with chronic disease by outlining the preferred format and composition of groups according to those with lived experience. Group-based psychological treatment for chronic disease patients should account for these preferences to improve its acceptability and usefulness among patients

    Altered Functionality, Morphology, and Vesicular Glutamate Transporter Expression of Cortical Motor Neurons from a Presymptomatic Mouse Model of Amyotrophic Lateral Sclerosis

    Amyotrophic lateral sclerosis (ALS) is a lethal disorder characterized by the gradual degeneration of motor neurons in the cerebrospinal axis. Whether upper motor neuron hyperexcitability, which is a feature of ALS, provokes dysfunction of glutamate metabolism and degeneration of lower motor neurons via an anterograde process is undetermined. To examine whether early changes in upper motor neuron activity occur in association with glutamatergic alterations, we performed whole-cell patch-clamp recordings to analyze excitatory properties of Layer V cortical motor neurons and excitatory postsynaptic currents (EPSCs) in presymptomatic G93A mice modeling familial ALS (fALS). We found that G93A Layer V pyramidal neurons exhibited altered EPSC frequency and rheobase values indicative of their hyperexcitability status. Biocytin loading of these hyperexcitable neurons revealed an expansion of their basal dendrite arborization. Moreover, we detected increased expression levels of the vesicular glutamate transporter 2 in cortical Layer V of G93A mice. Altogether our data show that functional and structural neuronal alterations associate with abnormal glutamatergic activity in motor cortex of presymptomatic G93A mice. These abnormalities, expected to enhance glutamate release and to favor its accumulation in the motor cortex, provide strong support for the view that upper motor neurons are involved early on in the pathogenesis of ALS

    Characterizing permafrost active layer dynamics and sensitivity to landscape spatial heterogeneity in Alaska

    An important feature of the Arctic is large spatial heterogeneity in active layer conditions, which is generally poorly represented by global models and can lead to large uncertainties in predicting regional ecosystem responses and climate feedbacks. In this study, we developed a spatially integrated modeling and analysis framework combining field observations, local-scale ( ∌ 50 m resolution) active layer thickness (ALT) and soil moisture maps derived from low-frequency (L + P-band) airborne radar measurements, and global satellite environmental observations to investigate the ALT sensitivity to recent climate trends and landscape heterogeneity in Alaska. Modeled ALT results show good correspondence with in situ measurements in higherpermafrost-probability (PP ≄ 70 %) areas (n = 33; R = 0.60; mean bias = 1.58 cm; RMSE = 20.32 cm), but with larger uncertainty in sporadic and discontinuous permafrost areas. The model results also reveal widespread ALT deepening since 2001, with smaller ALT increases in northern Alaska (mean trend = 0.32 ± 1.18 cm yr−1 ) and much larger increases (> 3 cm yr−1 ) across interior and southern Alaska. The positive ALT trend coincides with regional warming and a longer snow-free season (R = 0.60 ± 0.32). A spatially integrated analysis of the radar retrievals and model sensitivity simulations demonstrated that uncertainty in the spatial and vertical distribution of soil organic carbon (SOC) was the largest factor affecting modeled ALT accuracy, while soil moisture played a secondary role. Potential improvements in characterizing SOC heterogeneity, including better spatial sampling of soil conditions and advances in remote sensing of SOC and soil moisture, will enable more accurate predictions of active layer conditions and refinement of the modeling framework across a larger domain
