    Efficacy of Lycium barbarum polysaccharide and synergism with paclitaxel/cisplatin in ovarian cancer in mice

    Purpose: To determine the efficacy of Lycium barbarum polysaccharide (LBP) in ovarian cancer, and the synergistic effect when used in combination with paclitaxel/cisplatin (PXT∕DDP).Methods: ID-8 cells were injected subcutaneously into 5 groups of female C57BL/6 mice (5 mice/group): control (Con), chemotherapy (CH, PTX/DDP), combination therapy 1 (co-CH1, PTX/DDP + 50 mg/kg LBP), combination therapy 2 (co-CH2, 100 mg/kg LBP) and combination therapy 3 (co-CH3, 150 mg/kg LBP). Tissue morphological changes were monitored by hematoxylin and eosin (H&E) staining. Protein and mRNA levels of Keap1, Nrf2) and heme oxygenase-1 (HemO-1) were analyzed by western blotting and real-time polymerase chain reaction (PCR), as applicable.Results: Growth rate and volume of tumors were significantly reduced in the chemotherapy and combination therapy groups, while organ index increased significantly in co-CH group. Morphological structure of tumor, liver and kidney became normal after combination therapy. Levels of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), uric acid (UA), creatinine (Cr) and blood urea nitrogen (BUN) were significantly decreased in co-CH group relative to CH group. Lymphocytes, monocytes (MNC), neutrophils, basophils and eosnophils were significantly regulated by combination therapy. In CH and co-CH1-3 groups, the mRNA and protein levels of Keap1, HO-1 and Nrf2 were significantly increased relative to those of control mice.Conclusion: LBP in combination with PXT∕DDP enhances the efficacy of the latter, and reduced its toxicity when used for the treatment of ovarian malignant tumor in mice, by activating Keap1/Nrf2 pathway and promoting immunity.Keywords: Lycium barbarum polysaccharide (LBP), Paclitaxel/Cisplatin, Keap1/Nrf2 pathway, Cytotoxicit

    Theoretical and Computational Analysis of Credit and Liquidity Risk with Multiple Defaults

    Since the 2008 global financial crisis, regulators have been paying considerable attention to the credit and liquidity risks. Two such concepts (related to credit and liquidity risks) that have been repeatedly mentioned in the regulatory announcements are the credit value adjustment (CVA) and the Incremental Risk Charge (IRC). The CVA is an adjustment to the previous trade price when the counterparty risk has been added. The IRC is a new type of risk charge defined in Basel II which covers the major exposures of the counterparty and liquidity risk in the trading book. The current models on CVA and IRC have specific shortcomings. The CVA is currently calculated on a one-period model with restriction on the number of defaults. The IRC is computed using the time consuming Monte Carlo simulations. In this dissertation, we have made significant contributions to risk analysis by solving CVA in both two-default and full model without the restriction on the number of defaults as well as providing an analytical method for calculating IRC. Our research can be considered as a major step forward in expanding the current credit and liquidity risks models. Compared to the current one-default CVA calculations, our two-default and full calculations offer the distinct advantages of more accurate and practical CVA results. On top of that, our PDE method provides the speed and accuracy which allows us to finish a thorough risk exposure analysis and identify the conditions when the first default CVA overestimates or underestimates the counterparty risk. As opposed to the current numerical approach of calculating IRC, we offer an analytical method which provides an approximation of VaR on the two-period model and exact value of VaR on the infinite-default model. This is the first analytic solution in the literature on the multi-period capital model and may impact the view of current measure of risk controls in the banks. Thus credit risk control can be greatly improved if this new analytic solution can be applied in financial industry. Combined together, the work in this dissertation makes significant improvements in credit risk analysis in the multi-period credit and liquidity risks models

    Twist angle effects on the dynamic response of in-plane-switching liquid crystal displays

    Twist angle effect on the response time of in-plane-switching liquid crystal displays are analyzed. We propose a device configuration whose top and bottom boundary liquid crystal layers are symmetric to each other with respect to the electric field direction. The analytical results of this device configuration indicate that the response time is improved at least 4X faster than that of a conventional in-plane-switching twisted-nematic mode and normal in-plane-switching mode.Comment: 12 Pages, 3 Figures. will be appeared in Appl.     On a new class of Finsler metrics

    In this paper, the geometric meaning of (alpha,beta)-norms is made clear. On this basis, we introduce a new class of Finsler metrics called general (alpha,beta)-metrics, which are defined by a Riemannian metric and an 1-form. These metrics not only generalize original (alpha,beta)-metrics naturally, but also include some metrics structured by R. Bryant. The notion of general (alpha,beta)-metrics is one of the original ideas belongs to the first author(another one is beta-deformations intruduced in the paper "Deformations and Hilbert's Fourth Problem"). We believe that the researches on general (alpha,beta)-metrics will enrich Finsler geometry and the approaches offer references for further study. But it seems that the classical methods suitable for (alpha,beta)-metrics may not be suitable for them, the idea used in this paper, which is closely related to beta-deformations, is non-classical.
