    Variations in protein concentration and nitrogen sources in different positions of grain in wheat

    The distribution patterns of total protein and protein components in different layers of wheat grain were investigated using the pearling technique, and the sources of different protein components and pearling fractions were identified using (15)N isotope tracing methods. It was found that N absorbed from jointing to anthesis (JA) and remobilized to the grain after anthesis was the principal source of grain N, especially in the outer layer. For albumin and globulin, the amount of N absorbed during different stages all showed a decreasing trend from the surface layer to the center part. Whereas, for globulin and glutenin, the N absorbed after anthesis accounted for the main part indicating that for storage protein, the utilization of N assimilated after anthesis is greater than that of the stored N assimilated before anthesis. It is concluded that manipulation of the N application rate during different growth stages could be an effective approach to modulate the distribution of protein fractions in pearled grains for specific end-uses

    Mechano-stimulated modifications in the chloroplast antioxidant system and proteome changes are associated with cold response in wheat

    BACKGROUND: Mechanical wounding can cause morphological and developmental changes in plants, which may affect the responses to abiotic stresses. However, the mechano-stimulation triggered regulation network remains elusive. Here, the mechano-stimulation was applied at two different times during the growth period of wheat before exposing the plants to cold stress (5.6 °C lower temperature than the ambient temperature, viz., 5.0 °C) at the jointing stage. RESULTS: Results showed that mechano-stimulation at the Zadoks growth stage 26 activated the antioxidant system, and substantially, maintained the homeostasis of reactive oxygen species. In turn, the stimulation improved the electron transport and photosynthetic rate of wheat plants exposed to cold stress at the jointing stage. Proteomic and transcriptional analyses revealed that the oxidative stress defense, ATP synthesis, and photosynthesis-related proteins and genes were similarly modulated by mechano-stimulation and the cold stress. CONCLUSIONS: It was concluded that mechano-stimulated modifications of the chloroplast antioxidant system and proteome changes are related to cold tolerance in wheat. The findings might provide deeper insights into roles of reactive oxygen species in mechano-stimulated cold tolerance of photosynthetic apparatus, and be helpful to explore novel approaches to mitigate the impacts of low temperature occurring at critical developmental stages.     Molecular basis of ligand recognition and activation of human V2 vasopressin receptor.

    Vasopressin type 2 receptor (V2R) belongs to the vasopressin (VP)/oxytocin (OT) receptor subfamily of G protein-coupled receptors (GPCRs), which comprises at least four closely related receptor subtypes: V1aR, V1bR, V2R, and OTR. These receptors are activated by arginine vasopressin (AVP) and OT, two endogenous nine-amino acid neurohypophysial hormones, which are thought to mediate a biologically conserved role in social behavior and sexual reproduction. V2R is mainly expressed in the renal collecting duct principal cells and mediates the antidiuretic action of AVP by accelerating water reabsorption, thereby playing a vital role in controlling water homeostasis. Moreover, numerous gain-of-function and loss-of-function mutations of V2R have been identified and are closely associated with human diseases, including nephrogenic syndrome of inappropriate diuresis (NSIAD) and X-linked congenital nephrogenic diabetes insipidus (NDI). Thus, V2R has attracted intense interest as a drug target. However, due to a lack of structural information, how AVP recognizes and activates V2R remains elusive, which hampers the V2R-targeted drug design. Here, we determined a 2.6 Å resolution cryo-EM structure of the full-length, G s -coupled human V2R bound to AVP (Fig. 1a; Supplementary information, Table S1). The G s protein was engineered based on mini-G s that was used in the crystal structure determination of the G s -coupled adenosine A 2A receptor (A 2A R) to stabilize the V2R–G s protein complex (Supplementary information, Data S1). The final structure of the AVP–V2R–G s complex contains all residues of AVP (residues 1–9), the Gα s Ras-like domain, Gβγ subunits, Nb35, scFv16, and the V2R residues from T31 to L339 8.57 (superscripts refer to Ballesteros–Weinstein numbering). The majority of amino acid side chains, including AVP, transmembrane domain (TMD), all flexible intracellular loops (ICLs) and extracellular loops (ECLs) except for ICL3 and G185–G188 in ECL2, were well resolved in the model, refined against the EM density map (Fig. 1a; Supplementary information, Figs.     Cholecystokinin A receptor (CCKAR) belongs to family A G-protein-coupled receptors and regulates nutrient homeostasis upon stimulation by cholecystokinin (CCK). It is an attractive drug target for gastrointestinal and metabolic diseases. One distinguishing feature of CCKAR is its ability to interact with a sulfated ligand and to couple with divergent G-protein subtypes, including Gs, Gi and Gq. However, the basis for G-protein coupling promiscuity and ligand recognition by CCKAR remains unknown. Here, we present three cryo-electron microscopy structures of sulfated CCK-8-activated CCKAR in complex with Gs, Gi and Gq heterotrimers, respectively. CCKAR presents a similar conformation in the three structures, whereas conformational differences in the 'wavy hook' of the Gα subunits and ICL3 of the receptor serve as determinants in G-protein coupling selectivity. Our findings provide a framework for understanding G-protein coupling promiscuity by CCKAR and uncover the mechanism of receptor recognition by sulfated CCK-8

    Improved PSO_AdaBoost Ensemble Algorithm for Imbalanced Data

    The Adaptive Boosting (AdaBoost) algorithm is a widely used ensemble learning framework, and it can get good classification results on general datasets. However, it is challenging to apply the AdaBoost algorithm directly to imbalanced data since it is designed mainly for processing misclassified samples rather than samples of minority classes. To better process imbalanced data, this paper introduces the indicator Area Under Curve (AUC) which can reflect the comprehensive performance of the model, and proposes an improved AdaBoost algorithm based on AUC (AdaBoost-A) which improves the error calculation performance of the AdaBoost algorithm by comprehensively considering the effects of misclassification probability and AUC. To prevent redundant or useless weak classifiers the traditional AdaBoost algorithm generated from consuming too much system resources, this paper proposes an ensemble algorithm, PSOPD-AdaBoost-A, which can re-initialize parameters to avoid falling into local optimum, and optimize the coefficients of AdaBoost weak classifiers. Experiment results show that the proposed algorithm is effective for processing imbalanced data, especially the data with relatively high imbalances
