    Nephrectomy Delay of More than 10 Weeks from Diagnosis Is Associated with Decreased Overall Survival in pT3 RCC

    In this study, we aimed to evaluate the impact of surgical wait time (SWT) on outcomes of patients with renal cell carcinoma (RCC), and to investigate risk factors associated with prolonged SWT. Using the National Cancer Database, we retrospectively reviewed the records of patients with pT3 RCC treated with radical or partial nephrectomy between 2004 and 2014. The cohort was divided based on SWT. The primary outcome was 5-year overall survival (OS). Logistic regression analysis was used to investigate the risk factors associated with delayed surgery. Cox proportional hazards models were fitted to assess relations between SWT and 5-year OS after adjusting for confounding factors. A total of 22,653 patients were included in the analysis. Patients with SWT > 10 weeks had higher occurrence of upstaging. Using logistic regression, we found that female patients, African-American or Spanish origin patients, treatment in academic or integrated network cancer center, lack of insurance, median household income of <$38,000, and the Charlson–Deyo score of ≥1 were more likely to have prolonged SWT. SWT > 10 weeks was associated with decreased 5-year OS (hazard ratio [HR], 1.24; 95% confidence interval [CI], 1.15–1.33). This risk was not markedly attenuated after adjusting for confounding variables, including age, gender, race, insurance status, Charlson–Deyo score, tumor size, and surgical margin status (adjusted HR, 1.13; 95% CI, 1.04–1.24). In conclusion, the vast majority of patients underwent surgery within 10 weeks. There is a statistically significant trend of increasing SWT over the study period.     Reconstruction of daily chlorophyll-a concentrations in the transit of severe tropical cyclone Hudhud using the ExDINEOF method

    Tropical regions experience a diverse range of dense clouds, posing challenges for the daily reconstruction of chlorophyll-a concentration data. This underscores the pressing need for a practical method to reconstruct daily-scale chlorophyll-a concentrations in such regions. While traditional data reconstruction methods focus on single variables and rely on specific factors to infer missing data at specific locations, these single-variable methods may falter when applied to tropical oceans due to the scarcity of available data. Fortunately, all oceanographic variables undergo similar atmospheric and marine dynamic processes, creating internal relationships between them. This allows for the reconstruction of missing data through correlations between variables. Thus, this study introduces a multivariate reconstruction approach using the extended data interpolating empirical orthogonal function (ExDINEOF) method to reconstruct missing daily-scale chlorophyll-a concentration data. The ExDINEOF method considers the simultaneous relationships among multiple variables for data reconstruction in tropical oceans. To verify the method’s robustness, missing data were reconstructed during the formation and passage of severe tropical cyclone Hudhud through the Bay of Bengal. The results demonstrate that ExDINEOF outperforms traditional data reconstruction methods, exhibiting favorable spatial distribution and enhanced accuracy within the dynamic tropical marine environment. Furthermore, an assessment of marine physical environmental factors associated with chlorophyll-a concentration data provides additional evidence for the ExDINEOF method’s accuracy. Notably, the ExDINEOF method offers comprehensive spatial distribution aligned with underlying physical mechanisms governing phytoplankton distribution patterns, detailed phytoplankton growth, bloom, extinction variations in time series, satisfactory accuracy, and comprehensive local-level details

    The sustainable mining Internet of Things deals with the applications of IoT technology to the coupled needs of sustainable recovery of metals and a healthy environment for a thriving planet. In this chapter, the IoT architecture and technology is presented to support development of a digital mining platform emphasizing the exploration of rock–fluid–environment interactions to develop extraction methods with maximum economic benefit, while maintaining and preserving both water quantity and quality, soil, and, ultimately, human health. New perspectives are provided for IoT applications in developing new mineral resources, improved management of tailings, monitoring and mitigating contamination from mining. Moreover, tools to assess the environmental and social impacts of mining including the demands on dwindling freshwater resources. The cutting-edge technologies that could be leveraged to develop the state-of-the-art sustainable mining IoT paradigm are also discussed

    Understanding the Interaction between Regulatory Focus and Message Framing in Determining Chinese Consumers’ Attitudes toward Artificial Meat

    While production and consumption of meat cast a shadow over the prospects for sustainable development, artificial meat may be the solution. However, consumer acceptability of artificial meat is a major impediment to its use as a suitable alternative. This study analyzed the relationship between regulatory focus and consumer acceptance of artificial meat using randomized controlled trial data. Results showed that promotion focus results in a higher acceptance of artificial meat products due to a higher perceived benefit and lower perceived risk, whereas prevention focus results in a lower acceptance of artificial meat products due to perceived benefit being lower and perceived risk being higher. The moderating effect of the message framing was investigated employing structural equation modeling (SEM). It was discovered that a gain-oriented message framing could greatly strengthen the association between promotion focus and perceived benefit, whereas an avoidance-oriented message framing could significantly diminish the relationship between prevention focus and perceived risk. This study has crucial implications for how policymakers and industries communicate with consumers about artificial meat

    Reptin is required for the transcription of telomerase reverse transcriptase and over-expressed in gastric cancer

    <p>Abstract</p> <p>Background</p> <p>Telomerase is activated in oncogenesis, which confers an immortal phenotype to cancer cells. The AAA + ATPase Reptin is required for telomerase biogenesis by maintaining telomerase RNA (hTER) stability and is aberrantly expressed in certain cancers. Given its role in chromatin remodeling and transcription regulation, we determined the effect of Reptin on the transcription of the <it>telomerase reverse transcriptase </it>(<it>hTERT</it>) gene, a key component of the telomerase complex and its expression in gastric cancer.</p> <p>Results</p> <p>Knocking down Reptin or its partner Pontin using small interfering RNA in gastric and cervical cancer cells led to significant decreases in <it>hTERT </it>mRNA, but <it>hTERT </it>promoter activity was inhibited in only Reptin-depleted cells. Reptin interacted with the c-MYC oncoprotein and its stimulatory effect on the <it>hTERT</it>promoter was significantly dependent on functional E-boxes in the promoter. Moreover, Reptin bound to the <it>hTERT </it>proximal promoter and the loss of the Reptin occupancy led to dissociation of c-MYC from the <it>hTERT </it>promoter in Reptin-depleted cells. Reptin inhibition dramatically impaired clonogenic potential of gastric cancer cells by inducing cell growtharrest and over-expression of Reptin was observed in primary gastric cancer specimens.</p> <p>Conclusions</p> <p>The <it>hTERT </it>gene is a direct target of Reptin, and <it>hTERT </it>transcription requires constitutive expression of Reptin and its cooperation with c-MYC. Thus, Reptin regulates telomerase at two different levels. This finding, together with the requirementof Reptin for the clonogenic potential of cancer cells and its over-expression in gastriccancer and other solid tumors, suggests that Reptin may be a putative therapeutic target.</p
