Molekulare Kapseln und Protein - Rezeptoren auf Calixaren - Basis
Im Rahmen dieser Dissertation wurden molekulare
Kapseln durch Selbstorganisation zweier ionischer
Calix[4]arenhalbschalen in polarer Lösung hergestellt und
mittels verschiedener Methoden wie NMR - Titration, Job, NOESY
- Experimenten detailliert untersucht. Weiterhin wurden die
kapseln durch ESI-MS genau charakterisiert. Zum erstenmal
gelang uns die Bestimmung der relativen StabilitÀt der
selbstorganisierten Kapseln in Lösung. Unsere Ergebnisse
belegen eine relativ gute Ăbereinstimmung zwischen
Bindungskonstanten aus der Lösung (NMR-Titration) und absoluten
PeakintensitĂ€ten in den ESI-MS Spektren. Ăber MS/MS-Experimente
gelang darĂŒber hinaus die Bestimmung der StabilitĂ€t der
molekularen Kapseln in der Gasphase. Wegen des amphiphilen
Charakters des Calix[4]arentetraphosphonats wurde es in eine
StearinsÀure - Monoschicht an der Luft/Wasser-GrenzflÀche
eingelagert. Dadurch wurde ein biomimetisches Membrane - Modell
mit darin befindlichen mobilem Rezeptoren aufgebaut. Mit Hilfe
dessen können Proteine je nach IEP in bis zu nanomolaren
Konzentrationen in Wasser, Puffer - Lösung bzw. Salz - Lösung
nachgewiesen werden

Protein surface mimetics: understanding how ruthenium tris(bipyridines) interact with proteins.
Protein surface mimetics achieve high affinity binding by exploiting a scaffold to project binding groups over a large area of solvent exposed protein surface to make multiple co-operative non-covalent interactions. Such recognition is a pre-requisite for competitive/ orthosteric inhibition of protein-protein interactions (PPIs). This paper describes biophysical and structural studies on ruthenium(II) tris(bipyridine) surface mimetics that recognize cytochrome (cyt) c and inhibit the cyt c/ cyt c peroxidase (CCP) PPI. Binding is electrostatically driven, with enhanced affinity achieved through enthalpic contributions thought to arise from the ability of the surface mimetics to make a greater number of non-covalent interactions with surface exposed basic residues on cyt c in comparison to CCP. High field natural abundance 1H-15N HSQC NMR experiments are consistent with surface mimetics binding to cyt c in similar manner to CCP. This provides a framework for understanding recognition of proteins by supramolecular receptors and informing the design of ligands superior to the protein partners upon which they are inspired
A Simple Tetraminocalix[4]arene as a Highly Efficient Catalyst under "On-Water" Conditions through Hydrophobic Amplification of Weak Hydrogen Bonds
8The simple tetraminocalix[4]arene 1, which contains weak H-bond-donor NH2 groups, is reported to be a highly efficient organocatalyst for the Vinylogous Mukaiyama Aldol Reaction (VMAR) of 2-(trimethylsilyloxy)furan 5 with α-ketoesters 6âa-l under "on-water" conditions owing to the hydrophobic amplification of weak H-bond interactions. The catalytic efficiency of calixarene catalyst 1 was shown to be closely related to its recognition abilities towards the reactants 5 and 6 through a multipoint recognition model. Gas-phase chemistry of molecular containers
The remarkable technical advances in mass spectrometry during the last decades, including soft ionisation techniques, the coupling of electrospray ionisation to flow reactors, and the broad scope of tandem mass spectrometric experiments applicable to mass-selected ions allow investigating the chemistry of molecular capsules in solution as well as in the absence of any environment. With these methods, mass spectrometry is capable of answering many questions starting from providing analytical characterisation data (elemental composition, stoichiometry, etc.) to structural aspects (connectivities, positions of building blocks in supramolecular complexes) and to the examination of solution and gas-phase reactivity including reactions inside molecular containers. The present article reviews this work with a focus rather on the chemical questions that can be answered than on the technical specialities of (tandem) mass spectrometry
