    Quick and sensitive determination of gene expression of fatty acid synthase in vitro by using real-time polymerase chain reaction amplification (PCR)

    Obesity results from an imbalance between energy intake and energy expenditure, which leads to a pathological accumulation of adipose tissue, but the underlying mechanism at gene level, is far from being elucidated. The objective of this study was to investigate the correlation between mRNA express from fatty acid synthase (FAS) with a different glucose level in primary adipocytes by real-time polymerase chain reaction amplification (PCR), which can aid in the understanding of the mechanism of obesity in vitro. By using the following formula, this study was able to quantify the mRNA expression of FAS of unknown samples: Y = -3.156X + 41.21 (Y = threshold cycle, X = log starting quantity). The high concentrations of glucose group significantly improved the mRNA expression of FAS (P < 0.01) rather than 0.25 and 0% concentrations of glucose. These results provide significant data that confirm an association between different glucose level and FAS expression in preadipocytes. The glucose concentration of the high group substantially augmented the mRNA expression of FAS.Key words: Expression, fatty acid synthase, lipid deposition, real-time polymerase chain reaction amplification (PCR)

    Thickness dependence of microstructures in La0.8Ca0.2MnO3 thin films

    The thickness dependence of microstructures of La0.8Ca0.2MnO3 (LCMO)/SrTiO3 (STO) thin films was investigated by high-resolution x-ray diffraction, small angle x-ray reflection, grazing incidence x-ray diffraction, scanning electron microscopy, and atomic force microscopy. The results show that all the LCMO films are well oriented in (00l) direction perpendicular to the substrate surface. Self-organized crystalline grains with a tetragonal shape are uniformly distributed on the film surface, indicating the deposition condition being of benefit to the formation of the crystalline grains. With increasing the film thickness, the crystalline quality of the LCMO film is improved, while the surface becomes rougher. There exists a nondesigned cap layer on the upper surface of the LCMO layer for all the samples. The mechanism is discussed briefly.published_or_final_versio

    Microstructures and resistivity of cuprate/manganite bilayer deposited on SrTiO3 substrate

    Thin Yba[SUB2]Cu[SUB3]O[SUB7-δ/La[SUB0.67]Ca[SUB0.33]MnO[SUB3] (YBCO/LCMO) films were grown on SrTiO[SUB3](STO)substrates by magnetron sputtering technique. The microstructures of the bilayers were characterized and a standard four-probe technique was applied to measure the resistivity of the samples. The interdiffusions at the YBCO/LCMO and LCMO/STO interfaces formed two transient layers with the thickness of about 3 and 2 nm, respectively. All the bilayers were well textured along the c axis. At low temperature, the superconductivity can only be observed when the thickness of YBCO is more than 25 nm. When the thickness of YBCO is less than 8 nm, the bilayers show only ferromagnetism. The superconductivity and ferromagnetism perhaps coexist in the bilayer with the YBCO thickness of 12.5 nm. These interesting properties are related to the interaction between spin polarized electrons in the manganites and the cooper pairs in the cuprates. © 2003 American Institute of Physics.published_or_final_versio

    Improving the Energy-Conversion Efficiency of a PV-TE System with an Intelligent Power-Track Switching Technique and Efficient Thermal-Management Scheme

    A photovoltaic-thermoelectric (PV-TE) hybrid system can be used for efficient thermal energy utilization from the generated waste heat in PV devices. In this article, an efficient PV-TE hybrid system with intelligent power-track switching technique and thermal management based on energy conversion is proposed. To make the output power of PV-TE system stable and normalized, an incorporated stable voltage circuit is designed based on energy conversion. In addition, a control-and-monitoring strategy is launched in the system to realize the normal collecting for the output power of PV-TE system. Finally, a battery protection circuit is performed to ensure that the energy converted by the entire system is effectively stored. The experimental results show that more electrical energy about 84 034 J was obtained with our energy harvesting system than that of a single photovoltaic (PV) cell. Besides, the thermal gradient of PV cells is indirectly reduced the operation of the whole system, which is automatically monitored due to the proposed intelligent power-track switching technique

    Hepatotoxicity or Hepatoprotection? Pattern Recognition for the Paradoxical Effect of the Chinese Herb Rheum palmatum L. in Treating Rat Liver Injury

    The hepatotoxicity of some Chinese herbs has been a cause for concern in recent years. However, some herbs, such as rhubarb, have been documented as having both therapeutic and toxic effects on the liver, leading to the complex problem of distinguishing the benefits from the risks of using this herb. To comparatively analyze the dose-response relationship between rhubarb and hepatic health, we administrated total rhubarb extract(RE) to normal and carbon tetrachloride(CCl4)-treated rats for 12 weeks at 4 dosage levels(2.00, 5.40, 14.69 and 40.00 g·kg−1, measured as the quantity of crude material), followed by biochemical and histopathological tests of the rats' livers. A composite pattern was extracted by factor analysis, using all the biochemical indices as variables, into a visual representation of two mathematically obtained factors, which could be interpreted as the fibrosis factor and the cellular injury factor, according to the values of the variable loadings. The curative effect of administering the two lowest dosages of RE to CCl4-treated rats was mainly expressed as a decrease in the extent of cellular injury. The hepatoprotective mechanism of RE might be related to its antioxidant effect, the antagonism of the free radical damage to hepatocytes caused by CCl4. By contrast, the RE-induced liver damage was mainly expressed as a significant increase in the amount of fibrosis in both normal rats at all dosage levels and CCl4-treated rats at the two highest dosage levels. Therefore, the hepatotoxic potential of RE could be attributable to the liver cell fibrosis induced by high doses of the herb. This study illustrates the bidirectional potential of rhubarb and demonstrates the feasibility of using factor analysis to study the dose-response relationships between herbal medicines and hepatotoxicity or the healing effects of these herbs by extracting the underlying interrelationships among a number of functional bio-indices in a holistic manner

    The prevalence of hyperuricemia in China: a meta-analysis

    <p>Abstract</p> <p>Background</p> <p>The prevalence of hyperuricemia varied in different populations and it appeared to be increasing in the past decades. Recent studies suggest that hyperuricemia is an independent risk factor for cardiovascular disease. However, there has not yet been a systematic analysis of the prevalence of hyperuricemia in China.</p> <p>Methods</p> <p>Epidemiological investigations on hyperuricemia in China published in journals were identified manually and on-line by using CBMDISC, Chongqing VIP database and CNKI database. Those Reported in English journals were identified using MEDLINE database. Selected studies had to describe an original study defined by strict screening and diagnostic criteria. The fixed effects model or random effects model was employed according to statistical test for homogeneity.</p> <p>Results</p> <p>Fifty-nine studies were selected, the statistical information of which was collected for systematic analysis. The results showed that the pooled prevalence of hyperuricemia in male was 21.6% (95%CI: 18.9%-24.6%), but it was only 8.6% (95%CI: 8.2%-10.2%) in female. It was found that thirty years was the risk point age in male and it was fifty years in female.</p> <p>Conclusions</p> <p>The prevalence of hyperuricemia is different as the period of age and it increases after 30 years in male and 50 in female. Interventions are necessary to change the risk factors before the key age which is 30 years in male and 50 in female.</p

    Modelling of inquiry diagnosis for coronary heart disease in traditional Chinese medicine by using multi-label learning

    <p>Abstract</p> <p>Background</p> <p>Coronary heart disease (CHD) is a common cardiovascular disease that is extremely harmful to humans. In Traditional Chinese Medicine (TCM), the diagnosis and treatment of CHD have a long history and ample experience. However, the non-standard inquiry information influences the diagnosis and treatment in TCM to a certain extent. In this paper, we study the standardization of inquiry information in the diagnosis of CHD and design a diagnostic model to provide methodological reference for the construction of quantization diagnosis for syndromes of CHD. In the diagnosis of CHD in TCM, there could be several patterns of syndromes for one patient, while the conventional single label data mining techniques could only build one model at a time. Here a novel multi-label learning (MLL) technique is explored to solve this problem.</p> <p>Methods</p> <p>Standardization scale on inquiry diagnosis for CHD in TCM is designed, and the inquiry diagnostic model is constructed based on collected data by the MLL techniques. In this study, one popular MLL algorithm, ML-kNN, is compared with other two MLL algorithms RankSVM and BPMLL as well as one commonly used single learning algorithm, k-nearest neighbour (kNN) algorithm. Furthermore the influence of symptom selection to the diagnostic model is investigated. After the symptoms are removed by their frequency from low to high; the diagnostic models are constructed on the remained symptom subsets.</p> <p>Results</p> <p>A total of 555 cases are collected for the modelling of inquiry diagnosis of CHD. The patients are diagnosed clinically by fusing inspection, pulse feeling, palpation and the standardized inquiry information. Models of six syndromes are constructed by ML-kNN, RankSVM, BPMLL and kNN, whose mean results of accuracy of diagnosis reach 77%, 71%, 75% and 74% respectively. After removing symptoms of low frequencies, the mean accuracy results of modelling by ML-kNN, RankSVM, BPMLL and kNN reach 78%, 73%, 75% and 76% when 52 symptoms are remained.</p> <p>Conclusions</p> <p>The novel MLL techniques facilitate building standardized inquiry models in CHD diagnosis and show a practical approach to solve the problem of labelling multi-syndromes simultaneously.</p
