    Response Generation by Context-aware Prototype Editing

    Open domain response generation has achieved remarkable progress in recent years, but sometimes yields short and uninformative responses. We propose a new paradigm for response generation, that is response generation by editing, which significantly increases the diversity and informativeness of the generation results. Our assumption is that a plausible response can be generated by slightly revising an existing response prototype. The prototype is retrieved from a pre-defined index and provides a good start-point for generation because it is grammatical and informative. We design a response editing model, where an edit vector is formed by considering differences between a prototype context and a current context, and then the edit vector is fed to a decoder to revise the prototype response for the current context. Experiment results on a large scale dataset demonstrate that the response editing model outperforms generative and retrieval-based models on various aspects

    Frequency-dependent orthotropic damping properties of Nomex honeycomb composites

    In this paper, the orthotropic damping behavior of Nomex honeycomb composites and its causes are investigated. The needed specimen sizes for the measurement of the frequency-dependent transverse shear moduli (TSM) and fundamental damping coefficients of the honeycomb cores were analyzed at first. Then, the effects of cell side length and beam orientation on the orthotropic damping properties were explored. The results reveal that relatively high TSM (GLT) and damping values (ηWT) can be obtained by decreasing the cell side length without adding any additional weight. Damping mechanism analysis indicates that the difference in damping contribution of the interfacial phase to honeycomb core in different directions leads to the orthotropic damping behavior of honeycomb core. This study is helpful to guide the TSM measurement and structure design of honeycomb composites

    PCB169 exposure aggravated the development of non-alcoholic fatty liver in high-fat diet-induced male C57BL/6 mice

    Polychlorinated biphenyls (PCBs) are lipophilic environmental toxicants. Epidemiological studies have established a link between PCBs and both metabolic syndrome and nonalcoholic fatty liver disease (NAFLD). Multiple studies have reported that exposure to both PCB156 and PCB126 among the 12 dioxin-like PCBs leads to the development of NAFLD. However, studies to elucidate whether PCB169 induces the development of NAFLD by constructing in vivo models have not been reported. Therefore, we evaluated the effects of exposure to PCB169 (5 mg/kg-bw) on hepatic lipid metabolism in C57BL/6 mice from control diet and high-fat diet cohorts. The results showed that PCB169 exposure reduced body weight and intraperitoneal fat mass in mice on the control diet, but the liver lipid levels were significantly increased, exacerbating NAFLD in mice on a high-fat diet. Through transcriptomics studies, it was found that PCB169 exposure induced significant up-regulation of Pparγ, Fasn, and Aacs genes involved in hepatic lipogenesis, as well as remarkable up-regulation of Hmgcr, Lss, and Sqle genes involved in cholesterol synthesis. Additionally, there was notable down-regulation of Pparα and Cpt1 genes involved in lipid β-oxidation, leading to abnormal lipid accumulation in the liver. In addition, we found that PCB169 exposure significantly activated the Arachidonic acid metabolism, PPAR signaling pathway, Metabolism of xenobiotics by cytochrome P450, and Retinol metabolism pathways, and so on. Our study suggests that PCB169 can modify gene expression related to lipid metabolism, augument lipid accumulation in the liver, and further contribute to the development of NAFLD, thereby revealing the detrimental effects associated with PCB exposure on animal growth and metabolism

However, the molecular mechanism of the action of genistein on cancer cell apoptosis is not well known. In this study, we investigated the effect of genistein on topoisomerase II-α (Topo IIα), an important protein involved in the processes of DNA replication and cell proliferation. The results revealed that inhibition of Topo IIα expression through the regulation of Specificity protein 1 and Specificity protein 3 may be one of the reasons for genistein’s induction of HeLa cell apoptosis

    Genome-Wide Characterization of Menin-Dependent H3K4me3 Reveals a Specific Role for Menin in the Regulation of Genes Implicated in MEN1-Like Tumors

    Inactivating mutations in the MEN1 gene predisposing to the multiple endocrine neoplasia type 1 (MEN1) syndrome can also cause sporadic pancreatic endocrine tumors. MEN1 encodes menin, a subunit of MLL1/MLL2-containing histone methyltransferase complexes that trimethylate histone H3 at lysine 4 (H3K4me3). The importance of menin-dependent H3K4me3 in normal and transformed pancreatic endocrine cells is unclear. To study the role of menin-dependent H3K4me3, we performed in vitro differentiation of wild-type as well as menin-null mouse embryonic stem cells (mESCs) into pancreatic islet-like endocrine cells (PILECs). Gene expression analysis and genome-wide H3K4me3 ChIP-Seq profiling in wild-type and menin-null mESCs and PILECs revealed menin-dependent H3K4me3 at the imprinted Dlk1-Meg3 locus in mESCs, and all four Hox loci in differentiated PILECs. Specific and significant loss of H3K4me3 and gene expression was observed for genes within the imprinted Dlk1-Meg3 locus in menin-null mESCs and the Hox loci in menin-null PILECs. Given that the reduced expression of genes within the DLK1-MEG3 locus and the HOX loci is associated with MEN1-like sporadic tumors, our data suggests a possible role for menin-dependent H3K4me3 at these genes in the initiation and progression of sporadic pancreatic endocrine tumors. Furthermore, our investigation also demonstrates that menin-null mESCs can be differentiated in vitro into islet-like endocrine cells, underscoring the utility of menin-null mESC-derived specialized cell types for genome-wide high-throughput studies

    Colorectal adenomas contain multiple somatic mutations that do not coincide with synchronous adenocarcinoma specimens

    We have performed a comparative ultrasequencing study of multiple colorectal lesions obtained simultaneously from four patients. Our data show that benign lesions (adenomatous or hyperplastic polyps) contain a high mutational load. Additionally multiple synchronous colorectal lesions show non overlapping mutational signatures highlighting the degree of heterogeneity between multiple specimens in the same patient. Observations in these cases imply that considering not only the number of mutations but an effective oncogenic combination of mutations can determine the malignant progression of colorectal lesions

    Determination of the Influence of Fuel Switching Regulation on the Sulfur Dioxide Content of Air in a Port Area Using DID Model

    Since January 1, 2018, ships berthed at all ports of the three designated emission control areas (ECAs) in China are required to use fuel with sulfur content not exceeding 0.5% (m/m), excluding one hour postarrival and one hour predeparture. To understand changes in SO2 due to this policy, two observation stations were established on Waigaoqiao Dock in the Yangtze River estuary. Three data types were collected from March 2018 to May 2018, namely, wind speed and direction, SO2 concentration, and ships’ arrival and departure times. The statistics indicate that the wind direction changed little during the observation period and SO2 concentration was below 5 µg/m3 77.47% of the time. Meanwhile, ships’ arrival and departure at the dock had a distinct influence on overall SO2 distribution, including occurrence of concentrations ≥5 µg/m3. The three types of data were divided into six groups and a difference-in-difference model was used for analysis. The result shows that SO2 concentration increases due to the use of high-sulfur fuel and is especially significant when the wind is southwesterly. Furthermore, there was a positive correlation between increases in SO2 concentrations over 5 µg/m3 and the number of ships arriving or departing from the port. This study reports the positive impact of fuel switching on air quality and can be used to evaluate adherence to the ECA policy

    A Real-Time Measurement-Modeling System for Ship Air Pollution Emission Factors

    The lack of techniques for monitoring ship emissions all day and in all weather conditions to obtain real-time emission factor values is the main problem in understanding the characteristics of ship emissions, and there is still no perfect solution. In this study, a real-time measurement-modeling system was designed and implemented. The system was divided into three parts: (1) a portable exhaust monitoring device, which could be mounted on a drone, aircraft, patrol boat, dock, and bridge crane, as well as on the shore, to conduct all-weather and real-time online monitoring of ship emissions; (2) a monitoring information platform for ship emissions, based on a Spring + Spring MVC + MyBatis (SSM) framework and Vue front-end technology; and (3) a cloud server that received real-time ship emission measurement data and stored it after verification and analysis to calculate the pollutant gas and particulate matter emission factors. Following development, this system was used to monitor the emissions of ocean-going and inland river ships. Analysis of the acquired data showed that the system could effectively measure the emission factors of ship exhausts full-time in a variety of weather scenarios. This system can improve the efficiency of maritime law enforcement and provide technical support for promoting the construction of ship emission control areas. It can also help researchers obtain ship emission data, as well as an improved understanding of the emission characteristics of ships
