    Honors Advising for Large Programs

    This study was conducted within the Honors Program at the University of Iowa, which is the flagship public research university of the State of Iowa. Its Carnegie classification is Doctoral University with Highest Research Activity (R1), and it is a member of the Association of American Universities. Its current student population is about 21,600 undergraduates and about 9,600 graduate and professional students. Beyond using the rate of completion of the required 12 units of honors coursework within four semesters as a measure of the effectiveness of peer mentoring, we also surveyed students who had some type of peer mentoring experience as described above. To assess the effects of peer mentoring, we used nine Likert-scaled questions. (These questions are shown in Table 3.) We also asked two open-ended questions that required a written response: one question on how peer mentoring contributed to the mentee’s honors experience and another on ways to improve peer mentoring

    Hypervelocity Impact of Composite Overwrap Pressure Vessels

    There is a limited amount of hypervelocity impact (HVI) data on pressurized composite overwrapped pressure vessels (COPV). In recent years, NASA has performed HVI tests to characterize impact conditions resulting in either leak or burst of the COPVs representative of spacecraft hardware. This paper reports on the results of 40 tests that have been conducted on several types of COPV configurations, pressurized by inert gas to near the vessels rated maximum expected operating pressure (MEOP). These tests were used to better understand COPV response under HVI conditions and develop ballistic limit equations (BLE) related to these tests

    Record-breaking earthquake intervals in a global catalogue and an aftershock sequence

    For the purposes of this study, an interval is the elapsed time between two earthquakes in a designated region; the minimum magnitude for the earthquakes is prescribed. A record-breaking interval is one that is longer (or shorter) than preceding intervals; a starting time must be specified. We consider global earthquakes with magnitudes greater than 5.5 and show that the record-breaking intervals are well estimated by a Poissonian (random) theory. We also consider the aftershocks of the 2004 Parkfield earthquake and show that the record-breaking intervals are approximated by very different statistics. In both cases, we calculate the number of record-breaking intervals (<i>n</i><sub>rb</sub>) and the record-breaking interval durations Δ<i>t</i><sub>rb</sub> as a function of "natural time", the number of elapsed events. We also calculate the ratio of record-breaking long intervals to record-breaking short intervals as a function of time, <i>r(t)</i>, which is suggested to be sensitive to trends in noisy time series data. Our data indicate a possible precursory signal to large earthquakes that is consistent with accelerated moment release (AMR) theory

    A microbiological assay for host-specific fungal polyketide toxins

    Genetic analysis of biosynthetic pathways for fungal secondary metabolites depends on availability of efficient and dependable assays for the end products. Some fungal plant pathogens produce secondary metabolites called host-specific toxins. Until recently, all bioassays for these toxins required use of whole plants or plant parts (Yoder 1981 In: Toxins in Plant Disease, Durbin ed., pp. 45-78). Since host-specific toxins, by definition, affect only plants that are susceptible to the toxin-producing fungus, other plants, animals and microorganisms are not sensitive and therefore cannot be used in bioassays

    A Dimensional Model of Psychopathology Among Homeless Adolescents: Suicidality, Internalizing, and Externalizing Disorders

    The present study examined associations among dimensions of suicidality and psychopathology in a sample of 428 homeless adolescents (56.3% female). Confirmatory factor analysis results provided support for a three-factor model in which suicidality (measured with lifetime suicidal ideation and suicide attempts), internalizing disorders (assessed with lifetime diagnoses of major depressive episode and post-traumatic stress disorder), and externalizing disorders (indicated by lifetime diagnoses of conduct disorder, alcohol abuse, and drug abuse) were positively intercorrelated. The findings illustrate the utility of a dimensional approach that integrates suicidality and psychopathology into one model

    Split-Marker Recombination for Efficient Targeted Deletion of Fungal Genes

    A commonly used method for fungal gene deletion is introduction of linear DNA consisting of a selectable marker gene flanked on both sides by short stretches of DNA that target a gene of interest (Wirsel et al 1996 Curr. Genet 29:241-249). Gene deletion in Cochliobolus heterostrophus and Gibberella zeae occurs efficiently with this approach. To facilitate deletion construct synthesis, we have applied the split-marker” deletion strategy previously developed for Saccharomyces cerevisiae (Fairhead et al. 1996 Yeast 12:1439-57; Fairhead et al. 1998 Gene 223:33-46). Here, we describe both fusion PCR-based and plasmid-based deletion methods using this strategy with PEG-mediated protoplast transformation (Turgeon et al, 1985 Mol. Gen. Genet. 201:450-453). These methods are predicted to work well with any transformable fungus that undergoes homologous recombination between chromosomal and introduced DNA sequences

    A single codon insertion in PICALM is associated with development of familial subvalvular aortic stenosis in Newfoundland dogs

    Familial subvalvular aortic stenosis (SAS) is one of the most common congenital heart defects in dogs and is an inherited defect of Newfoundlands, golden retrievers and human children. Although SAS is known to be inherited, specific genes involved in Newfoundlands with SAS have not been defined. We hypothesized that SAS in Newfoundlands is inherited in an autosomal dominant pattern and caused by a single genetic variant. We studied 93 prospectively recruited Newfoundland dogs, and 180 control dogs of 30 breeds. By providing cardiac screening evaluations for Newfoundlands we conducted a pedigree evaluation, genome-wide association study and RNA sequence analysis to identify a proposed pattern of inheritance and genetic loci associated with the development of SAS. We identified a three-nucleotide exonic insertion in phosphatidylinositol-binding clathrin assembly protein (PICALM) that is associated with the development of SAS in Newfoundlands. Pedigree evaluation best supported an autosomal dominant pattern of inheritance and provided evidence that equivocally affected individuals may pass on SAS in their progeny. Immunohistochemistry demonstrated the presence of PICALM in the canine myocardium and area of the subvalvular ridge. Additionally, small molecule inhibition of clathrin-mediated endocytosis resulted in developmental abnormalities within the outflow tract (OFT) of Xenopus laevis embryos. The ability to test for presence of this PICALM insertion may impact dog-breeding decisions and facilitate reduction of SAS disease prevalence in Newfoundland dogs. Understanding the role of PICALM in OFT development may aid in future molecular and genetic investigations into other congenital heart defects of various species
