The N=C bond length of 1.263 (6) Å is shorter than of the N—C bond [1.476 (5) Å], indicating a double bond. In the crystal, there is some pseudosymmetry. This occurs because most of the two mol­ecules are centrosymmetrically related. The mol­ecular structure is stabilized by intra­molecular O—H⋯N hydrogen bonds

    Objective: To explore the attack, prevalence, clinical features, diagnosis, therapy and prognosis of enterovirus 71 and coxsackie A16. Methods: Analyzing the general situation, main symptoms and signs, laboratory examination, virus effect analysis and the prognosis of 126 cases of hospital medical records. Results: Among the 126 cases, CA16 is in the majority, and EV71 virus is coexisting. Besides, the ordinary cases have mild symptoms without serious complications, and attention should be paid attention to the fever, blood glucose and myocardial enzyme spectrum and complications, etc. Severe cases need to be discovered early. Conclusion: Timely detection and treatment are the effective ways to reduce the deaths of seriously ill patients lie in full attention to the severe trend.目的  探讨肠道病毒71型(EV71)和柯萨奇A组16型(CoxA16)共感的发病、流行及临床特征、诊断、治疗及预后。方法  分析126例住院病历的一般情况,主要症状及体征,实验室检查,病毒分析情况及治疗预后效果。结果  126例病例中,以CA16为主,有EV71病毒同时存在,普通病例症状轻,无严重并发症,要注意发热、血糖、心肌酶谱及合并症等。尽早发现重症病例。结论  充分重视重症趋势,及时发现,及时治疗,是目前减少重症患者死亡的有效方法

    Protective Effect of Ethanol Extracts of Hericium erinaceus

    We investigated the effects of Hericium erinaceus (HEE) on alloxan induced diabetic neuropathic pain in laboratory rats. Alloxan induced diabetic rats were administered orally HEE. After 6 weeks of treatments, treatment with HEE 40 mg/kg in diabetic animals showed significant increase in pain threshold and paw withdrawal threshold and significant decrease in serum glucose and urine glucose. We also observed a significant increase in lactate dehydrogenase (LDH), Lipid peroxidation (LPO), glutathione peroxidase (GPx) activity, glutathione reductase (GR) activity, catalase (CAT) activity, Na+K+ATPase activity, and glutathione S transferase (GST) activity along with significant decreased levels of glutathione (GSH) content in diabetic rats. The total antioxidant status (TAOS) in the HEE-treated groups was significantly lower than that in the alloxan-treated group. HEE can offer pain relief in diabetic neuropathic pain. The improvement in diabetic state after HEE treatment along with the antioxidant activity could be the probable way by which it had alleviated diabetic neuropathy

    Frequent occurrence of unreduced gametes in Triticum turgidum-Aegilops tauschii hybrids.

    Abstract Spontaneous chromosome doubling via union of unreduced (2n) gametes has been thought to be the way that common wheat (Triticum aestivum L.) was originated from the hybridization of T. turgidum L. with Ae. tauschii Cosson. Previous works have observed unreduced gametes in F 1 hybrids of Ae. tauschii with six of the eight T. turgidum subspecies. It is not clear, however, whether the formation of these unreduced gametes is a norm in the F 1 hybrids. In the present study, we tried to answer this question by assessing the occurrence frequency of unreduced gametes in 115 T. turgidum-Ae. tauschii hybrid combinations, involving 76 genotypes of seven T. turgdium subspecies and 24 Ae. tauschii accessions. Our data show that these hybrid combinations differed significantly (P B 0.01, F = 11.40) in selfed seedset, an indicator for production of unreduced gametes. This study clearly showed that meiotic restitution genes are widely distributed within T. turgidum. However, significant differences were found between as well as within T. turgidum subspecies and in the interaction of the T. turgidum genotypes with those of Ae. taushii. The possible application of the meiotic restitution genes from T. turgidum in production of double haploids is also discussed

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Postoperative subacute static progressive stretch does not increase the risk of distal lower limb venous thromboembolism

    Purpose: Static progressive stretch (SPS) can be applied to treat chronic joint stiffness. However, the impacts of subacute application of SPS to the distal lower limbs, where deep vein thrombosis (DVT) is common, on venous thromboembolism remain unclear. This study aims to explore the risk of venous thromboembolism events following subacute application of SPS. Methods: A retrospective cohort study was conducted on patients diagnosed with DVT following a lower extremity orthopedic surgery before being transferred to the rehabilitation ward from May 2017 to May 2022. Patients with unilateral lower limb comminuted para-articular fractures, transferred to rehabilitation ward for further treatment within 3 weeks after operation, followed up more than 12 weeks since initial manual physiotherapy, and diagnosed DVT by ultrasound before rehabilitation course were included in the study. Patients with polytrauma, without evidence of previous peripheral vascular disease or incompetence, had medication for thrombosis treatment or prophylaxis before the operation, detected with paralysis due to nervous system impairment, infected after operation during the regime, or with acute progression of DVT were excluded. The included patients were randomized to the standard physiotherapy and the SPS integrated groups for observation. Associated DVT and pulmonary embolism data were collected during the physiotherapy course to compare the groups. SSPS 28.0 and GraphPad Prism 9 were used for data processing. A p < 0.05 was set significant difference. Results: In total of 154 patients with DVT participating in this study, 75 of them were treated with additional SPS for postoperative rehabilitation. The participants in the SPS group showed improved range of motion (12.3° ± 6.7°). However, in the SPS group, there was no difference in thrombosis volume between the start and termination (p = 0.106, p = 0.787, respectively), although difference was seen intra-therapy (p < 0.001). Contingency analysis revealed the pulmonary embolism incidence (OR = 0.703) in the SPS group compared to the mean physiotherapy. Conclusion: The SPS technique is a safe and reliable option to prevent potential joint stiffness without aggravating the risk of distal DVT for postoperative patients suffering from relevant trauma

    Cistanche tubulosa Protects Dopaminergic Neurons through Regulation of Appotosis and Glial Cell-derived Neurotrophic Factor: in Vivo and in Vitro

    Parkinson’s disease (PD) is a neurodegenerative disease with the pathological hallmark of reduced nigrostriatal dopamine. In traditional Chinese medicine (TCM) clinical practice, the nanopowder of Cistanche tubulosa has therapeutic effects on PD. To identify the therapeutic mechanism, this study tested the protective effect of different doses of MPP+-induced toxicity in MES23.5 cells using the MTT assay and in the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) -induced PD mice (vehicles). Immunohistochemistry was used to assess cytomorphology and tyrosine hydroxylase (TH) expression. Behavioural tests in vehicles, high performance liquid chromatography (HPLC) tests in dopamine, immunohistochemistry and western blot analysis were used to detect the expression of TH, glial cell line-derived neurotrophic factor (GDNF) and its receptors. Our results demonstrated that the C. tubulosa nanopowder improved the viability of MPP+-treated cells, increased TH expression and reduced the number of apoptotic cells. It also increased Bcl2 protein expression and suppressed Bax protein expression in MPP+-treated cells in a dose-dependent manner. In addition, C. tubulosa nanopowder improved the behavioural deficits in vehicle mice, reduced the stationary duration of swimming, enhanced the ability for spontaneous activity, and increased the expression of GDNF, the GDNF family receptor alpha (GFRα1) and Ret in cells of the substantia nigra (SN). Furthermore, the protein expression of GDNF, GFRα1 and Ret increased after treatment with different doses of C. tubulosa nanopowder, with a significant difference between the high-dose and vehicle groups. The protein expression of Bcl2 and Bax were similar in the in vivo and in vitro, which suggested that C. tubulosa nanopowder has anti-apoptotic effects in neurons
