    Population-Based Sexual Behavior Surveys in China: Liuzhou Compared with Other Prefectural Cities

    Sexual behaviors in China are rapidly changing; simultaneously, STI/HIV prevalence is increasing in the general population. To investigate these major shifts, we examined sexual behaviors and self-reported sexually transmitted infections (STI) in one prefectural city in southern China, Liuzhou, and compared it to other prefectural cities throughout China. We used adults age 18-39 from two sets of population-based surveys that paralleled each other in both content and method. The first set was the Liuzhou survey conducted in 2008 (n=398). The second set consisted of two national surveys collected in 2006 and 2010 (n=2186). Liuzhou respondents reported more active social and sexual behaviors than their national counterparts, including more socializing, dancing, drinking excessively, sexual activity among never married men and women, purchasing commercial sex among men, one-night stands among men, multiple sexual partnerships and self-reported STI among both men and women. Women in Liuzhou reported greater sexual risk behavior than their national counterparts, although overall they reported less than their male counterparts; they were also more likely to have had an abortion than women in other prefectural cities. Our findings provide a comprehensive overview of the sexual context of Liuzhou among the general population, which may help explain the greater STI/HIV prevalence in Liuzhou

    New insights into the germline genes and CDR3 repertoire of the TCRβ chain in Chiroptera

    IntroductionBats are recognized as natural reservoirs for many viruses, and their unique immune system enables them to coexist with these viruses without frequently exhibiting disease symptoms. However, the current understanding of the bat adaptive immune system is limited due to the lack of a database or tool capable of processing T-cell receptor (TCR) sequences for bats.MethodsWe performed germline gene annotation in three bat species using homologous genes and RSSs (Recombinational Signal Sequences) scanning method. Then we used the conserved C gene to construct the TCRβ chain receptor library of the Intermediate Horseshoe Bat. Bats' TCRβ data will be analyzed using MiXCR and constructed reference library.ResultsRegarding the annotation results, we found that the Pale Spear-nosed Bat has 37 members in the TRBV12 family, which is more than the total number of TRBV genes in the Greater Horseshoe Bat. The average number of unique TCRβ chain receptor sequences in each Intermediate Horseshoe Bat sample reached 24,904.DiscussionThe distinct variations in the distribution of TRBV genes among the three types of bats could have a direct impact on the diversity of the TCR repertoire, as evidenced by the presence of conserved amino acids that indicate the T-cell recognition of antigens in bats is MHC-restricted.     Recurrent Fusion Genes in Gastric Cancer: CLDN18-ARHGAP26 Induces Loss of Epithelial Integrity.

    Genome rearrangements, a hallmark of cancer, can result in gene fusions with oncogenic properties. Using DNA paired-end-tag (DNA-PET) whole-genome sequencing, we analyzed 15 gastric cancers (GCs) from Southeast Asians. Rearrangements were enriched in open chromatin and shaped by chromatin structure. We identified seven rearrangement hot spots and 136 gene fusions. In three out of 100 GC cases, we found recurrent fusions between CLDN18, a tight junction gene, and ARHGAP26, a gene encoding a RHOA inhibitor. Epithelial cell lines expressing CLDN18-ARHGAP26 displayed a dramatic loss of epithelial phenotype and long protrusions indicative of epithelial-mesenchymal transition (EMT). Fusion-positive cell lines showed impaired barrier properties, reduced cell-cell and cell-extracellular matrix adhesion, retarded wound healing, and inhibition of RHOA. Gain of invasion was seen in cancer cell lines expressing the fusion. Thus, CLDN18-ARHGAP26 mediates epithelial disintegration, possibly leading to stomach H(+) leakage, and the fusion might contribute to invasiveness once a cell is transformed.     Method of Feature Reduction in Short Text Classification Based on Feature Clustering

    One decisive problem of short text classification is the serious dimensional disaster when utilizing a statistics-based approach to construct vector spaces. Here, a feature reduction method is proposed that is based on two-stage feature clustering (TSFC), which is applied to short text classification. Features are semi-loosely clustered by combining spectral clustering with a graph traversal algorithm. Next, intra-cluster feature screening rules are designed to remove outlier feature words, which improves the effect of similar feature clusters. We classify short texts with corresponding similar feature clusters instead of original feature words. Similar feature clusters replace feature words, and the dimension of vector space is significantly reduced. Several classifiers are utilized to evaluate the effectiveness of this method. The results show that the method largely resolves the dimensional disaster and it can significantly improve the accuracy of short text classification

    Plant-mediated methane and nitrous oxide fluxes from a carex meadow in Poyang Lake during drawdown periods

    Aims: Plants have been suggested to have significant effects on methane (CH4) and nitrous oxide (N2O) fluxes from littoral wetlands, but it remains unclear in subtropical lakes. Methods: We conducted in situ measurement of CH4 and N2O fluxes for two years. To distinguish between the effects of shoots and roots, three treatments (i.e., intact plants as control, shoot clipping, and root exclusion) were used. Effects of plant biomass, temperature, and soil moisture on CH4 and N2O fluxes were analyzed. Results: The mean ecosystem CH4 emission rate was 36 μg CH4 m−2 h−1 for drying periods, but 8219 μg CH4 m−2 h−1 for drying-wetting transition periods. CH4 fluxes were positively correlated with below-ground and total biomass, but not with above-ground biomass. Clipping did not significantly alter CH4 flux rate, but root exclusion decreased the CH4 flux by 116 % as compared to the control. N2O emissions were similar for both the drying and drying-wetting transition periods, with a mean rate of 20 μg N2O m−2 h−1. Both clipping and root exclusion significantly increased N2O fluxes as compared to the control. Conclusions: There was no significant correlation between CH4 and N2O fluxes. Roots dominated plant-mediated enhancement in CH4 fluxes, but played almost an equal role as shoots in plant-regulated suppression on N2O fluxes in this Carex meadow during drawdown periods

    Sh3rf2 Haploinsufficiency Leads to Unilateral Neuronal Development Deficits and Autistic-Like Behaviors in Mice

    Summary: Autism spectrum disorders (ASDs) include a variety of developmental brain disorders with clinical findings implicating the dysfunction of the left hemisphere. Here, we generate mice lacking one copy of Sh3rf2, which was detected in ASD patients, to determine whether Sh3rf2 is involved in brain development and whether mutation of SH3RF2 is causative for ASD and the mechanisms linking it to ASD traits. We find that mice with Sh3rf2 haploinsufficiency display significant deficits in social interaction and communication, as well as stereotyped or repetitive behaviors and hyperactivity and seizures. Disturbances in hippocampal dendritic spine development, aberrant composition of glutamatergic receptor subunits, and abnormal excitatory synaptic transmission were detected in heterozygous mutants. Remarkably, these defects are selectively unilateral. Our results support a notion that Sh3rf2 haploinsufficiency is a highly penetrant risk factor for ASD, with disease pathogenesis most likely resulting from deficits in synaptic function in the left hemisphere of the brain. : Wang et al. find that Sh3rf2 is important for neuron development and that Sh3rf2 haploinsufficiency in mice results in typical ASD-like behaviors, as well as selective unilateral disturbances in hippocampal dendritic spine development, composition of glutamatergic receptor subunits, and excitatory synaptic transmission.     Improving the production of AHL lactonase AiiO-AIO6 from Ochrobactrum sp. M231 in intracellular protease-deficient Bacillus subtilis

    Quorum quenching (QQ) blocks bacterial cell-to-cell communication (i.e., quorum sensing), and is a promising antipathogenic strategy to control bacterial infection via inhibition of virulence factor expression and biofilm formation. QQ enzyme AiiO-AIO6 from Ochrobactrum sp. M231 has several excellent properties and shows biotherapeutic potential against important bacterial pathogens of aquatic species. AiiO-AIO6 can be secretory expressed in Bacillus subtilis via a non-classical secretion pathway. To improve AiiO-AIO6 production, four intracellular protease-deletion mutants of B. subtilis 1A751 were constructed by individually knocking out the intracellular protease-encoding genes (tepA, ymfH, yrrN and ywpE). The AiiO-AIO6 expression plasmid pWB-AIO6BS was transformed into the B. subtilis 1A751 and its four intracellular protease-deletion derivatives. Results showed that all recombinant intracellular protease-deletion derivatives (BSΔtepA, BSΔymfH, BSΔyrrN and BSΔywpE) had a positive impact on AiiO-AIO6 production. The highest amount of AiiO-AIO6 extracellular production of BSΔywpE in shake flask reached 1416.47 U/mL/OD600, which was about 121% higher than that of the wild-type strain. Furthermore, LC–MS/MS analysis of the degrading products of 3-oxo-C8-HSL by purification of AiiO-AIO6 indicated that AiiO-AIO6 was an AHL-lactonase which hydrolyzes the lactone ring of AHLs. Phylogenetic analysis showed that AiiO-AIO6 was classified as a member of the α/β hydrolase family with a conserved “nucleophile-acid-histidine” catalytic triad. In summary, this study showed that intracellular proteases were responsible for the reduced yields of heterologous proteins and provided an efficient strategy to enhance the extracellular production of AHL lactonase AiiO-AIO6.publishedVersio
