    A requirement for CD45 distinguishes Ly49D-mediated cytokine and chemokine production from killing in primary natural killer cells

    Engagement of receptors on the surface of natural killer (NK) cells initiates a biochemical cascade ultimately triggering cytokine production and cytotoxicity, although the interrelationship between these two outcomes is currently unclear. In this study we investigate the role of the cell surface phosphatase CD45 in NK cell development and intracellular signaling from activating receptors. Stimulation via the major histocompatibility complex I–binding receptor, Ly49D on CD45−/− primary NK cells resulted in the activation of phosphoinositide-3-kinase and normal cytotoxicity but failed to elicit a range of cytokines and chemokines. This blockage is associated with impaired phosphorylation of Syk, Vav1, JNK, and p38, which mimics data obtained using inhibitors of the src-family kinases (SFK). These data, supported by analogous findings after CD16 and NKG2D stimulation of CD45−/− primary NK cells, place CD45 upstream of SFK in NK cells after stimulation via immunoreceptor tyrosine-based activation motif-containing receptors. Thus we identify CD45 as a pivotal enzyme in eliciting a precise subset of NK cell responses

    Loss of the Pro-Apoptotic BH3-only Bcl-2 Family Member Bim Inhibits BCR Stimulation–induced Apoptosis and Deletion of Autoreactive B Cells

    During development, the stochastic process assembling the genes encoding antigen receptors invariably generates B and T lymphocytes that can recognize self-antigens. Several mechanisms have evolved to prevent the activation of these cells and the concomitant development of autoimmune disease. One such mechanism is the induction of apoptosis in developing or mature B cells by engagement of the B cell antigen receptor (BCR) in the absence of T cell help. Here we report that B lymphocytes lacking the pro-apoptotic Bcl-2 family member Bim are refractory to apoptosis induced by BCR ligation in vitro. The loss of Bim also inhibited deletion of autoreactive B cells in vivo in two transgenic systems of B cell tolerance. Bim loss prevented deletion of autoreactive B cells induced by soluble self-antigen and promoted accumulation of self-reactive B cells developing in the presence of membrane-bound self-antigen, although their numbers were considerably lower compared with antigen-free mice. Mechanistically, we determined that BCR ligation promoted interaction of Bim with Bcl-2, inhibiting its survival function. These findings demonstrate that Bim is a critical player in BCR-mediated apoptosis and in B lymphocyte deletion

    Jailbreaking ChatGPT via Prompt Engineering: An Empirical Study

    Large Language Models (LLMs), like ChatGPT, have demonstrated vast potential but also introduce challenges related to content constraints and potential misuse. Our study investigates three key research questions: (1) the number of different prompt types that can jailbreak LLMs, (2) the effectiveness of jailbreak prompts in circumventing LLM constraints, and (3) the resilience of ChatGPT against these jailbreak prompts. Initially, we develop a classification model to analyze the distribution of existing prompts, identifying ten distinct patterns and three categories of jailbreak prompts. Subsequently, we assess the jailbreak capability of prompts with ChatGPT versions 3.5 and 4.0, utilizing a dataset of 3,120 jailbreak questions across eight prohibited scenarios. Finally, we evaluate the resistance of ChatGPT against jailbreak prompts, finding that the prompts can consistently evade the restrictions in 40 use-case scenarios.     PentestGPT: An LLM-empowered Automatic Penetration Testing Tool

    Penetration testing, a crucial industrial practice for ensuring system security, has traditionally resisted automation due to the extensive expertise required by human professionals. Large Language Models (LLMs) have shown significant advancements in various domains, and their emergent abilities suggest their potential to revolutionize industries. In this research, we evaluate the performance of LLMs on real-world penetration testing tasks using a robust benchmark created from test machines with platforms. Our findings reveal that while LLMs demonstrate proficiency in specific sub-tasks within the penetration testing process, such as using testing tools, interpreting outputs, and proposing subsequent actions, they also encounter difficulties maintaining an integrated understanding of the overall testing scenario. In response to these insights, we introduce PentestGPT, an LLM-empowered automatic penetration testing tool that leverages the abundant domain knowledge inherent in LLMs. PentestGPT is meticulously designed with three self-interacting modules, each addressing individual sub-tasks of penetration testing, to mitigate the challenges related to context loss. Our evaluation shows that PentestGPT not only outperforms LLMs with a task-completion increase of 228.6\% compared to the \gptthree model among the benchmark targets but also proves effective in tackling real-world penetration testing challenges. Having been open-sourced on GitHub, PentestGPT has garnered over 4,700 stars and fostered active community engagement, attesting to its value and impact in both the academic and industrial spheres

    Fenomena yang terjadi selama ini pengusaha agribisnis/petani produsen belum menggunakan jasa sistem informasi komunikasi pasar tepat waktu. Sehingga pengusaha agribisnis tidak mengetahui harga pasar yang sebenarnya sehingga menjual produk dengan harga murah, produsen akan selalu dirugikan karena menjual produk dengan harga murah, pendapatan petani produsen tetap selalu rendah dan hidup di bawah garis kemiskinan. Penelitian ini menggunakan pendekatan kualitatif dengan metode riset partisipatoris yaitu kegiatan yang dilakukan oleh peneliti mengikuti dan memahami serta membahas masalah-masalah yang dihadapi masyarakat produsen/pedagang agribisnis dan juga melakukan pola pembinaannya. Untuk itu, peneliti bertemu langsung dengan masyarakat produsen/pedagang agribisnis dan menjalin persahabatan dengan sejumlah anggota masyarakat dalam menyelesaikan permasalahan penelitian. Metode ini berdampingan dengan menggunakan pendekatan Descriptive analysis untuk menjawab masalah yang memerlukan keterangan, gambaran dan sejenisnya secara faktual dan aktual. Populasi dalam penelitian ini adalah populasi terhingga. Sampel Penelitian ada di wilayah sentra produksi agribisnis Kecamatan Kayu Aro Kabupaten Kerinci. Hasil penelitian, pengusaha agribisnis terutama petani agribisnis sebagian belum menggunakan komunikasi pasar tepat waktu dan sebagian sudah menggunakan komunikasi pasar tepat waktu. Bagi pedagang perantara sudah melakukan sistem informasi komunikasi pasar tepat waktu baik untuk pemasaran lokal, domestik dan ekspor dan dapat menggunakannya dengan baik dan benar. Pengusaha agribisnis sudah dibina untuk dapat mengetahui harga pasar dengan menggunakan media komunikasi telepon rumah dan handphone. Oleh karena itu petani produsen dan pedagang agribisnis agar membentuk jejaring informasi komunikasi pasar pada semua pedagang agen di luar provinsi dan semua pedagang agen di luar pulau dan membentuk wadah persatuan informasi harga serta bersatu dalam menentukan harga pasar dan tidak saling menjatuhkan harga

Such a potent receptor, however, needs to be further balanced by positive and negative regulators to prevent harmful effects that may arise from inappropriate stimulation. Src family protein tyrosine kinase Lyn is involved in both positive and negative regulation, since the both gain-of-function Lyn and loss-of-function Lyn mutations caused autoimmunity in mice. The exact signalling pathway(s) regulated by Lyn in B cells, however, are still not clear.     Study on the Effect of an Intermittent Ventilation Strategy on Controlling Formaldehyde Concentrations in Office Rooms

    Material emission and ventilation are two aspects influencing indoor air quality. In this study, a model predictive control (MPC) strategy is proposed for intermittent ventilation system in office buildings, to achieve a healthy indoor environment. The strategy is based on a dynamic model for predicting emissions of volatile organic compounds (VOCs) from materials. The key parameters of formaldehyde from panel furniture in the model are obtained by an improved C-history method and large-scale chamber experiments. The effectiveness of the determined key parameters is validated, which are then used to predict the formaldehyde concentration variation and the pre-ventilation time in a typical office room. In addition, the influence of some main factors (i.e., vacant time, loading ratio, air change rate) on the pre-ventilation time is analyzed. Results indicate that the pre-ventilation time of the intermittent ventilation system ranges from several minutes to several hours. The pre-ventilation time decreases exponentially with the increase in the vacant time, the air change rate, and with the decrease in the loading ratio. When the loading ratio of the furniture is 0.30 m2/m3 and the vacant time is 100 days, the required pre-ventilation time approaches zero. Results further reveal that an air change rate of 2 h−1 is the most effective means for rapid removal of indoor formaldehyde for the cases studied. The proposed strategy should be helpful for achieving effective indoor pollution control
