Characterization of endophytic bacteriome diversity and associated beneficial bacteria inhabiting a macrophyte Eichhornia crassipes
IntroductionThe endosphere of a plant is an interface containing a thriving community of endobacteria that can affect plant growth and potential for bioremediation. Eichhornia crassipes is an aquatic macrophyte, adapted to estuarine and freshwater ecosystems, which harbors a diverse bacterial community. Despite this, we currently lack a predictive understanding of how E. crassipes taxonomically structure the endobacterial community assemblies across distinct habitats (root, stem, and leaf).MethodsIn the present study, we assessed the endophytic bacteriome from different compartments using 16S rRNA gene sequencing analysis and verified the in vitro plant beneficial potential of isolated bacterial endophytes of E. crassipes.Results and discussionPlant compartments displayed a significant impact on the endobacterial community structures. Stem and leaf tissues were more selective, and the community exhibited a lower richness and diversity than root tissue. The taxonomic analysis of operational taxonomic units (OTUs) showed that the major phyla belonged to Proteobacteria and Actinobacteriota (> 80% in total). The most abundant genera in the sampled endosphere was Delftia in both stem and leaf samples. Members of the family Rhizobiaceae, such as in both stem and leaf samples. Members of the family Rhizobiaceae, such as Allorhizobium- Neorhizobium-Pararhizobium-Rhizobium were mainly associated with leaf tissue, whereas the genera Nannocystis and Nitrospira from the families Nannocystaceae and Nitrospiraceae, respectively, were statistically significantly associated with root tissue. Piscinibacter and Steroidobacter were putative keystone taxa of stem tissue. Most of the endophytic bacteria isolated from E. crassipes showed in vitro plant beneficial effects known to stimulate plant growth and induce plant resistance to stresses. This study provides new insights into the distribution and interaction of endobacteria across different compartments of E. crassipes Future study of endobacterial communities, using both culture-dependent and -independent techniques, will explore the mechanisms underlying the wide-spread adaptability of E. crassipesto various ecosystems and contribute to the development of efficient bacterial consortia for bioremediation and plant growth promotion
Prediction of ESRD in IgA Nephropathy Patients from an Asian Cohort: A Random Forest Model
Background/Aims: There is an increasing risk of end-stage renal disease (ESRD) among Asian people with immunoglobulin A nephropathy (IgAN). A computer-aided system for ESRD prediction in Asian IgAN patients has not been well studied. Methods: We retrospectively reviewed biopsy-proven IgAN patients treated at the Department of Nephrology of the Second Xiangya Hospital from January 2009 to November 2013. Demographic and clinicopathological data were obtained within 1 month of renal biopsy. A random forest (RF) model was employed to predict the ESRD status in IgAN patients. All cases were initially trained and validated, taking advantage of the out-of-bagging(OOB) error. Predictors used in the model were selected according to the Gini impurity index in the RF model and verified by logistic regression analysis. The area under the receiver operating characteristic(ROC) curve (AUC) and F-measure were used to evaluate the RF model. Results: A total of 262 IgAN patients were enrolled in this study with a median follow-up time of 4.66 years. The importance rankings of predictors of ESRD in the RF model were first obtained, indicating some of the most important predictors. Logistic regression also showed that these factors were statistically associated with ESRD status. We first trained an initial RF model using gender, age, hypertension, serum creatinine, 24-hour proteinuria and histological grading suggested by the Clinical Decision Support System for IgAN (CDSS, www.IgAN.net). This 6-predictor model achieved a F-measure of 0.8 and an AUC of 92.57%. By adding Oxford-MEST scores, this model outperformed the initial model with an improved AUC (96.1%) and F-measure (0.823). When C3 staining was incorporated, the AUC was 97.29% and F-measure increased to 0.83. Adding the estimated glomerular filtration rate (eGFR) improved the AUC to 95.45%. We also observed improved performance of the model with additional inputs of blood urea nitrogen (BUN), uric acid, hemoglobin and albumin. Conclusion: In addition to the predictors in the CDSS, Oxford-MEST scores, C3 staining and eGFR conveyed additional information for ESRD prediction in Chinese IgAN patients using a RF model
Computed Tomography Screening for Early Lung Cancer, COPD and Cardiovascular Disease in Shanghai:Rationale and Design of a Population-based Comparative Study
Rationale and Objectives: To describe the rational and design of a population-based comparative study. The objective of the study is to assess the screening performance of volume-based management of CT-detected lung nodule in comparison to diameter-based management, and to improve the effectiveness of CT screening for chronic obstructive pulmonary disease (COPD) and cardiovascular disease (CVD), in addition to lung cancer, based on quantitative measurement of CT imaging biomarkers in a Chinese screening setting. Materials and Methods: A population-based comparative study is being performed, including 10,000 asymptomatic participants between 40 and 74 years old from Shanghai urban population. Participants in the intervention group undergo a low-dose chest and cardiac CT scan at baseline and 1 year later, and are managed according to NELCIN-B3 protocol. Participants in the control group undergo a low-dose chest CT scan according to the routine CT protocol and are managed according to the clinical practice. Epidemiological data are collected through questionnaires. In the fourth year from baseline, the diagnosis of the three diseases will be collected. Results: The unnecessary referral rate will be compared between NELCIN-B3 and standard protocol for managing early-detected lung nodules. The effectiveness of quantitative measurement of CT imaging biomarkers for early detection of lung cancer, COPD and CVD will be evaluated. Conclusion: We expect that the quantitative assessment of the CT imaging biomarkers will reduce the number of unnecessary referrals for early detected lung nodules, and will improve the early detection of COPD and CVD in a Chinese urban population
Vibrational spectroscopy and microwave dielectric properties of AY2Si3O10 (A=Sr, Ba) ceramics for 5G applications
AY2Si3O10 (AâŻ=âŻSr, Ba) trisilicate ceramics were synthesized by traditional high temperature solid state reaction method. X-ray diffraction patterns and Rietveld refinement revealed that AY2Si3O10 (AâŻ=âŻSr, Ba) ceramics belonged to triclinic and monoclinic crystal systems with PÄ« and P21/m space groups, respectively. The vibrational modes of [SiO4] tetrahedra, [YO6] octahedra and [(Sr/Ba)O8] polyhedra were analyzed by Raman spectroscopy. The infrared spectroscopy fitting analysis was used to determine intrinsic dielectric properties. Excellent microwave dielectric properties were measured for SrY2Si3O10 and BaY2Si3O10 with ÉrâŻ=âŻ9.3, QfâŻ=âŻ64100âŻGHz, ÏfâŻ=âŻâ31âŻppm/°C and ÉrâŻ=âŻ9.5, QfâŻ=âŻ65600âŻGHz, ÏfâŻ=âŻâ28âŻppm/°C, respectively. Both trisilicate ceramics are considered potential candidates for 5G and mm wave technology, provided Ïf can be further tuned
The DArk Matter Particle Explorer mission
The DArk Matter Particle Explorer (DAMPE), one of the four scientific space
science missions within the framework of the Strategic Pioneer Program on Space
Science of the Chinese Academy of Sciences, is a general purpose high energy
cosmic-ray and gamma-ray observatory, which was successfully launched on
December 17th, 2015 from the Jiuquan Satellite Launch Center. The DAMPE
scientific objectives include the study of galactic cosmic rays up to
TeV and hundreds of TeV for electrons/gammas and nuclei respectively, and the
search for dark matter signatures in their spectra. In this paper we illustrate
the layout of the DAMPE instrument, and discuss the results of beam tests and
calibrations performed on ground. Finally we present the expected performance

Liddle syndrome misdiagnosed as primary aldosteronism resulting from a novel frameshift mutation of SCNN1B
Liddle syndrome (LS), a monogenetic autosomal dominant disorder, is mainly characterized by early-onset hypertension and hypokalemia. Clinically, misdiagnosis or missing diagnosis is common, since clinical phenotypes of LS are variable and nonspecific. We report a family with misdiagnosis of primary aldosteronism (PA), but identify as LS with a pathogenic frameshift mutation of the epithelial sodium channel (ENaC) ÎČ subunit. DNA samples were collected from a 32-year-old proband and 31 other relatives in the same family. A designed panel including 41 genes associated with monogenic hypertension was screened using next-generation sequencing. The best candidate disease-causing variants were verified by Sanger sequencing. Genetic analysis of the proband revealed a novel frameshift mutation c.1838delC (p.Pro613Glnfs*675) in exon 13 of SCNN1B. This heterozygous mutation involved the deletion of a cytosine from a string of three consecutive cytosines located at codons 612 to 613 and resulted in deletion of the crucial PY motif and elongation of the ÎČ-ENaC protein. The identical mutation was also found in 12 affected family members. Amiloride was effective in alleviating LS for patients. There were no SCNN1A or SCNN1G mutations in this family. Our study emphasizes the importance of considering LS in the differential diagnosis of early-onset hypertension. The identification of a novel frameshift mutation of SCNN1B enriches the genetic spectrum of LS and has allowed treatment of this affected family to prevent severe complications
Direct detection of a break in the teraelectronvolt cosmic-ray spectrum of electrons and positrons
High energy cosmic ray electrons plus positrons (CREs), which lose energy
quickly during their propagation, provide an ideal probe of Galactic
high-energy processes and may enable the observation of phenomena such as
dark-matter particle annihilation or decay. The CRE spectrum has been directly
measured up to TeV in previous balloon- or space-borne experiments,
and indirectly up to TeV by ground-based Cherenkov -ray
telescope arrays. Evidence for a spectral break in the TeV energy range has
been provided by indirect measurements of H.E.S.S., although the results were
qualified by sizeable systematic uncertainties. Here we report a direct
measurement of CREs in the energy range by the
DArk Matter Particle Explorer (DAMPE) with unprecedentedly high energy
resolution and low background. The majority of the spectrum can be properly
fitted by a smoothly broken power-law model rather than a single power-law
model. The direct detection of a spectral break at TeV confirms the
evidence found by H.E.S.S., clarifies the behavior of the CRE spectrum at
energies above 1 TeV and sheds light on the physical origin of the sub-TeV

The rapid size- and shape-controlled continuous hydrothermal synthesis of metal sulphide nanomaterials
Continuous flow hydrothermal synthesis offers a cheap, green and highly scalable route for the preparation of inorganic nanomaterials which has predominantly been applied to metal oxide based materials. In this work we report the first continuous flow hydrothermal synthesis of metal sulphide nanomaterials. A wide range of binary metal sulphides, ZnS, CdS, PbS, CuS, Feâââá”ȘâS and BiâSâ, have been synthesised. By varying the reaction conditions two different mechanisms may be invoked; a growth dominated route which permits the formation of nanostructured sulphide materials, and a nucleation driven process which produces nanoparticles with temperature dependent size control. This offers a new and industrially viable route to a wide range of metal sulphide nanoparticles with facile size and shape control
