    Phylogenetic Analyses of Andean and Amazonian Tree Communities in Ecuador

    The forests of Ecuador are known for their high levels of diversity and endemism, classifying the country as a biodiversity hotspot. Both the western Amazon and Andean montane forests are richly populated with tropical tree species that have been little studied in a community phylogenetic context. The implementation of elevational transects and trait based analyses having proven useful in gaining a better understanding ofhow environmental factors are affecting the tree community structure in these habitats. The goal of this research was to evaluate the magnitude ofDNA barcode diversity among Amazonian and Andean tree species. Specifically, the objectives were to (1) evaluate community phylogenetic structure and correlate phylogenetic analyses with diversity metrics among Andean tree species along an elevational gradient at Siempre Verde Reserve, Ecuador, and to (2) construct a tropical tree community phylogeny using DNA barcodes and to test for phylogenetic signal in the occurrence of phytochemicals among tree species within Yasuni National Park, Ecuador. In the montane forest at Siempre Verde, 595 individuals were tagged, collected and identified, comprising 36 families, 53 genera, and 88 species. Analyses revealed that species richness was decreasing with elevation but the number of stems of common species was increasing causing phylogenetic clumping at higher elevations. Evidence implies that habitat filtering of species due to cloud inundation is behind this observed pattern contributing to the community structure. In the upland Amazonian forest of Yasuni, 337 common tree species making up 181 genera and 56 families were sent for sequencing, and the trait distribution of phytochemical presence was determined. Metrics of phylogenetic trait distribution all supported a random distribution of the medicinal trait within the Yasuni tree community. In the future, having higher sequence recovery and resolution along with complete floristic sampling will improve statistical power and the ability to detect fine scale community structure patterns in both of these forests. Studies like this, which include taxonomic, functional, and phylogenetic diversity, will allow for more comparisons to better understand these unique biodiversity hotspots

    The Covid‐19 pandemic has revealed and accelerated an information crisis as well as a health one. What we discover about Covid 19, how it spreads, to whom and why and how best to mitigate it—all depend on information. The essays in this special section, which this article introduces, explore the importance of information and the fundamental role of fact checkers in understanding how information flows, why mistakes are made, and how to counteract them. Fact checking as an idea and a practice emerged in the early twenty‐first century, developed as a positive beacon to counteract a growing sense that information could no longer be trusted. Now, more than a decade after its creation, fact checking sits within a far more complex and chaotic media context, and its expertise and understanding has never been so important. We need to understand what fact checkers do because they are grappling with how to tether us to reality

    High-resolution coproecology: Using coprolites to reconstruct the habits and habitats of New Zealand’s extinct upland Moa (Megalapteryx didinus)

    Knowledge about the diet and ecology of extinct herbivores has important implications for understanding the evolution of plant defence structures, establishing the influences of herbivory on past plant community structure and composition, and identifying pollination and seed dispersal syndromes. The flightless ratite moa (Aves: Dinornithiformes) were New Zealand's largest herbivores prior to their extinction soon after initial human settlement. Here we contribute to the knowledge of moa diet and ecology by reporting the results of a multidisciplinary study of 35 coprolites from a subalpine cave (Euphrates Cave) on the South Island of New Zealand. Ancient DNA analysis and radiocarbon dating revealed the coprolites were deposited by the extinct upland moa (Megalapteryx didinus), and span from at least 6,368±31 until 694±30 ¹⁴C years BP; the approximate time of their extinction. Using pollen, plant macrofossil, and ancient DNA analyses, we identified at least 67 plant taxa from the coprolites, including the first evidence that moa fed on the nectar-rich flowers of New Zealand flax (Phormium) and tree fuchsia (Fuchsia excorticata). The plant assemblage from the coprolites reflects a highly-generalist feeding ecology for upland moa, including browsing and grazing across the full range of locally available habitats (spanning southern beech (Nothofagus) forest to tussock (Chionochloa) grassland). Intact seeds in the coprolites indicate that upland moa may have been important dispersal agents for several plant taxa. Plant taxa with putative anti-browse adaptations were also identified in the coprolites. Clusters of coprolites (based on pollen assemblages, moa haplotypes, and radiocarbon dates), probably reflect specimens deposited at the same time by individual birds, and reveal the necessity of suitably large sample sizes in coprolite studies to overcome potential biases in diet interpretation

Publisher website: http://www.nhm-wien.ac.at/Until now, kiwi (Apteryx, Apterygidae) have had no pre-Quaternary fossil record to inform on the timing of their arrival in New Zealand or on their inter-ratite relationships. Here we describe two fossils in a new genus of apterygid from Early Miocene sediments at St Bathans, Central Otago, minimally dated to 19–16 Ma. The new fossils indicate a markedly smaller and possibly volant bird, supporting a possible overwater dispersal origin to New Zealand of kiwi independent of moa. If the common ancestor of this early Miocene apterygid species and extant kiwi was similarly small and volant, then the phyletic dwarfing hypothesis to explain relatively small body size of kiwi compared with other ratites is incorrect. Apteryx includes five extant species distributed on North, South, Stewart and the nearshore islands of New Zealand. They are nocturnal, flightless and comparatively large birds, 1–3 kg, with morphological attributes that reveal an affinity with ratites, but others, such as their long bill, that differ markedly from all extant members of that clade. Although kiwi were long considered most closely related to sympatric moa (Dinornithiformes), all recent analyses of molecular data support a closer affinity to Australian ratites (Casuariidae). Usually assumed to have a vicariant origin in New Zealand (ca 80–60 Ma), a casuariid sister group relationship for kiwi, wherein the common ancestor was volant, would more easily allow a more recent arrival via overwater dispersal

    Inhibitor regulation of tissue kallikrein activity in the synovial fluid of patients with rheumatoid athritis

    Tissue kallikrein (TK) and 1-antitrypsin (AT)/TK complexes can be detected in SF from patients with RA if components of the fluids which interfere with the detection of TK are removed. 2-Macroglobulin (2-M) in SF was demonstrated to contain trapped proteases which were still active in amidase assays. Removal of 2-M from RA SF reduced their amidase activity. However, at least some of the remaining activity was due to TK because it was soya bean trypsin inhibitor resistant and trasylol sensitive and was partly removed by affinity chromatography on anti-TK sepharose. Removal of RF from the fluids reduced the values obtained for TK levels by ELISA. Addition of SF to human urinary kallikrein (HUK) considerably reduced the levels of TK detected suggesting the presence of a TK ELISA inhibitor in the fluids. Removal of components of >300 kDa from SF markedly reduced the TK ELISA inhibitory activity and increased the values for both the TK and l-AT/TK levels in fluids as measured by ELISA. It is considered this novel inhibitor does not bind to the active site of TK but rather binds to the site reactive with anti-TK antibodies

    Cost effectiveness analysis of larval therapy for leg ulcers

    Objective: To assess the cost effectiveness of larval therapy compared with hydrogel in the management of leg ulcers. Design: Cost effectiveness and cost utility analyses carried out alongside a pragmatic multicentre, randomised, open trial with equal randomisation. Population: Intention to treat population comprising 267 patients with a venous or mixed venous and arterial ulcers with at least 25% coverage of slough or necrotic tissue. Interventions: Patients were randomly allocated to debridement with bagged larvae, loose larvae, or hydrogel. Main outcome measure: The time horizon was 12 months and costs were estimated from the UK National Health Service perspective. Cost effectiveness outcomes are expressed in terms of incremental costs per ulcer-free day (cost effectiveness analysis) and incremental costs per quality adjusted life years (cost utility analysis). Results: The larvae arms were pooled for the main analysis. Treatment with larval therapy cost, on average, 96.70 pound ((sic)109.61; $140.57) more per participant per year (95% confidence interval -491.9 pound to 685.8) pound than treatment with hydrogel. Participants treated with larval therapy healed, on average, 2.42 days before those in the hydrogel arm (95% confidence interval -0.95 to 31. 91 days) and had a slightly better health related quality of life, as the annual difference in QALYs was 0.011 (95% confidence interval -0.067 to 0.071). However, none of these differences was statistically significant. The incremental cost effectiveness ratio for the base case analysis was estimated at 8826 pound per QALY gained and 40 pound per ulcer-free day. Considerable uncertainty surrounds the outcome estimates. Conclusions: Debridement of sloughy or necrotic leg ulcers with larval therapy is likely to produce similar health benefits and have similar costs to treatment with hydrogel.     Early Miocene fossil frogs (Anura: Leiopelmatidae) from New Zealand.

    Author version made available in accordance with publisher copyright policy.The first pre-Quaternary anurans from New Zealand are reported from the Early Miocene (19–16 Ma) St Bathans Fauna based on 10 fossil bones. Four bones representing two new species differing in size are described in Leiopelma: Leiopelmatidae, and are the first Tertiary records for the family. Six indeterminate frog fossils are morphologically similar to leiopelmatids and represent two species consistent in size with those known from diagnostic material. These records are highly significant, as minimally, they reduce the duration of the leiopelmatid ‘ghost lineage’ by c.20 million years and demonstrate that a diversity of leiopelmatids has long been present on New Zealand, supporting the ancient dichotomy of the extant species based on molecular data

    Shared reading of children's interactive picture books

    We report on a study of children and parents shared reading of interactive printed books. We investigated the differences between books with interactive features and books with expressive typography in order to evaluate which features within a book encouraged interaction between the reading participants and the book. 11 parent and child groups took part in the study that involved three observed reading sessions. From our observations we offer suggestions for the development of books and eBooks to encourage shared reading practices
