    Crafting genetic diversity: unlocking the potential of protein evolution

    Genetic diversity is the foundation of evolutionary resilience, adaptive potential, and the flourishing vitality of living organisms, serving as the cornerstone for robust ecosystems and the continuous evolution of life on Earth. The landscape of directed evolution, a powerful biotechnological tool inspired by natural evolutionary processes, has undergone a transformative shift propelled by innovative strategies for generating genetic diversity. This shift is fuelled by several factors, encompassing the utilization of advanced toolkits like CRISPR-Cas and base editors, the enhanced comprehension of biological mechanisms, cost-effective custom oligo pool synthesis, and the seamless integration of artificial intelligence and automation. This comprehensive review looks into the myriad of methodologies employed for constructing gene libraries, both in vitro and in vivo, categorized into three major classes: random mutagenesis, focused mutagenesis, and DNA recombination. The objectives of this review are threefold: firstly, to present a panoramic overview of recent advances in genetic diversity creation; secondly, to inspire novel ideas for further innovation in genetic diversity generation; and thirdly, to provide a valuable resource for individuals entering the field of directed evolution

    Experiments of Interfacial Roughening in Hele-Shaw Flows with Weak Quenched Disorder

    We have studied the kinetic roughening of an oil--air interface in a forced imbibition experiment in a horizontal Hele--Shaw cell with quenched disorder. Different disorder configurations, characterized by their persistence length in the direction of growth, have been explored by varying the average interface velocity v and the gap spacing b. Through the analysis of the rms width as a function of time, we have measured a growth exponent beta ~= 0.5 that is almost independent of the experimental parameters. The analysis of the roughness exponent alpha through the power spectrum have shown different behaviors at short (alpha_1) and long (alpha_2) length scales, separated by a crossover wavenumber q_c. The values of the measured roughness exponents depend on experimental parameters, but at large velocities we obtain alpha_1 ~= 1.3 independently of the disorder configuration. The dependence of the crossover wavenumber with the experimental parameters has also been investigated, measuring q_c ~ v^{0.47} for the shortest persistence length, in agreement with theoretical predictions.Comment: 20 pages, 22 figure

    Optical activity of neutrinos and antineutrinos

    Using the one-loop helicity amplitudes for low-energy νγνγ\nu\gamma\to\nu\gamma and νˉγνˉγ\bar\nu\gamma\to\bar\nu\gamma scattering in the standard model with massless neutrinos, we study the optical activity of a sea of neutrinos and antineutrinos. In particular, we estimate the values of the index of refraction and rotary power of this medium in the absence of dispersion.Comment: Additional reference

    Conserving and Gapless Approximations for an Inhomogeneous Bose Gas at Finite Temperatures

    We derive and discuss the equations of motion for the condensate and its fluctuations for a dilute, weakly interacting Bose gas in an external potential within the self--consistent Hartree--Fock--Bogoliubov (HFB) approximation. Account is taken of the depletion of the condensate and the anomalous Bose correlations, which are important at finite temperatures. We give a critical analysis of the self-consistent HFB approximation in terms of the Hohenberg--Martin classification of approximations (conserving vs gapless) and point out that the Popov approximation to the full HFB gives a gapless single-particle spectrum at all temperatures. The Beliaev second-order approximation is discussed as the spectrum generated by functional differentiation of the HFB single--particle Green's function. We emphasize that the problem of determining the excitation spectrum of a Bose-condensed gas (homogeneous or inhomogeneous) is difficult because of the need to satisfy several different constraints.Comment: plain tex, 19 page

    Extension of the sum rule for the transition rates between multiplets to the multiphoton case

    The sum rule for the transition rates between the components of two multiplets, known for the one-photon transitions, is extended to the multiphoton transitions in hydrogen and hydrogen-like ions.     Upper critical field in dirty two-band superconductors: breakdown of the anisotropic Ginzburg-Landau theory

    We investigate the upper critical field in a dirty two-band superconductor within quasiclassical Usadel equations. The regime of very high anisotropy in the quasi-2D band, relevant for MgB2_{2}, is considered. We show that strong disparities in pairing interactions and diffusion constant anisotropies for two bands influence the in-plane Hc2H_{c2} in a different way at high and low temperatures. This causes temperature-dependent Hc2H_{c2} anisotropy, in accordance with recent experimental data in MgB2_{2}. The three-dimensional band most strongly influences the in-plane Hc2H_{c2} near TcT_{c}, in the Ginzburg-Landau (GL) region. However, due to a very large difference between the c-axis coherence lengths in the two bands, the GL theory is applicable only in an extremely narrow temperature range near TcT_c. The angular dependence of Hc2H_{c2} deviates from a simple effective-mass law even near TcT_c.Comment: 12 pages, 5 figures, submitted to Phys.Rev.

    Nucleon charge exchange on the deuteron: A critical review

    The existing experimental data on the d(n,p)nn and d(p,n)pp cross sections in the forward direction are reviewed in terms of the Dean sum rule. It is shown that the measurement of the ratio of the charge exchange on the deuteron to that on the proton might, if taken together with other experimental data, allow a direct construction of the np -> np scattering amplitude in the backward direction with few ambiguities.Comment: 7 pages with 3 figure
