    Adipose mTORC2 is essential for sensory innervation in white adipose tissue and whole-body energy homeostasis

    Adipose tissue, via sympathetic and possibly sensory neurons, communicates with the central nervous system (CNS) to mediate energy homeostasis. In contrast to the sympathetic nervous system, the morphology, role and regulation of the sensory nervous system in adipose tissue are poorly characterized.; Taking advantage of recent progress in whole-mount three-dimensional imaging, we identified a network of calcitonin gene-related protein (CGRP)-positive sensory neurons in murine white adipose tissue (WAT). We found that adipose mammalian target of rapamycin complex 2 (mTORC2), a major component of the insulin signaling pathway, is required for arborization of sensory, but not of sympathetic neurons. Time course experiments revealed that adipose mTORC2 is required for maintenance of sensory neurons. Furthermore, loss of sensory innervation in WAT coincided with systemic insulin resistance. Finally, we established that neuronal protein growth-associated protein 43 (GAP43) is a marker for sensory neurons in adipose tissue.; Our findings indicate that adipose mTORC2 is necessary for sensory innervation in WAT. In addition, our results also suggest that WAT may affect whole-body energy homeostasis via sensory neurons

This selective loss of sensory innervation followed reduced expression of growth-associated protein 43 (GAP43) in CGRP-positive sensory neurons. Finally, we found that loss of sensory innervation in WAT correlated with systemic insulin resistance. Our findings suggest that adipose mTORC2 is necessary for sensory innervation in WAT which likely contributes to WAT-to-CNS communication

    From near and far: Stone procurement and exchange at Çukuriçi Höyük in Western Anatolia

    The focus of this paper are the stone tools of Çukuriçi Höyük, a prehistoric site situated at the central Aegean coast of Anatolia. The settlement was inhabited from the Neolithic, through the Late Chalcolithic and Early Bronze Age 1 periods, a period lasting from the early 7th to the early 3rd millennium BCE. A long-term interdisciplinary study of the excavated lithics with different scientific methods on various stone materials (thin section analysis, pXRF, NAA, LA-ICP-MS) offer new primary data about the procurement strategies of prehistoric societies from a diachronic perspective. The results will be presented for the first time with an overview of all source materials and their distinct use through time. The lithic assemblages from Çukuriçi Höyük consist of a considerable variety of small finds, grinding stones and chipped stone tools. The high variability of raw materials within the different categories of tools is remarkable. In addition to stone tools manufactured from sources in the immediate vicinity of the settlement (i.e., mica-schist, limestone, marble, amphibolite, serpentinite), others are of rock types such as chert, which indicate an origin within the broader region. Moreover, volcanic rocks, notably the exceptionally high amount of Melian obsidian found at Çukuriçi Höyük, attest to the supra-regional procurement of distinct rock types. Small stone axes made of jadeite presumably from the Greek island of Syros, also indicate these far-reaching procurement strategies. The systematic and diachronic analyses of the stone tools found at Çukuriçi Höyük has demonstrated that as early as the Neolithic period extensive efforts were made to supply the settlement with carefully selected raw materials or finished goods procured from distinct rock sources.   Erratum:• This article was submitted and initially published with only the following authors listed: Christoph Schwall, Michael Brandl, Tatjana M.     Isolation of Salmonella enterica in seropositive classified finishing pig herds

    The aim of this study was to assess the probability of detecting Salmonella from pen faecal samples in seropositive classified finishing pig herds. The study involved 77 herds from Denmark (20), the Netherlands (20), Greece (17) and Germany (20). The serological herd status was determined by the blood- sampling of 50 finishing pigs. Bacteriological sampling was performed by 20 pen faecal samples per herd. Over-all, 47 % of the blood samples had an OD% larger than 10 and 23 % larger than 40. Salmonella was isolated from 135 (9.3 %) pen faecal samples in 32 herds (42 %). Twenty-eight of these herds (87.5 %) had a within-herd seroprevalence larger than 50% at sample cut-off OD%\u3e10. A correlation coefficient of 0.62 was found between the proportion of culture positive- and seropositive samples in a herd at cut-off OD % \u3e 10 and of 0.58 at cut-off OD % \u3e 40. Due to the low sensitivity of culture methods, apparent ‘false positive’ serological results may well represent real infections not detected by bacteriological testing.     Myocardial disease and ventricular arrhythmia in Marfan syndrome : a prospective study

    Background Aortic root dilatation and-dissection and mitral valve prolapse are established cardiovascular manifestations in Marfan syndrome (MFS). Heart failure and arrhythmic sudden cardiac death have emerged as additional causes of morbidity and mortality. Methods To characterize myocardial dysfunction and arrhythmia in MFS we conducted a prospective longitudinal case-control study including 86 patients with MFS (55.8% women, mean age 36.3 yr-range 13-70 yr-) and 40 age-and sex-matched healthy controls. Cardiac ultrasound, resting and ambulatory ECG (AECG) and NT-proBNP measurements were performed in all subjects at baseline. Additionally, patients with MFS underwent 2 extra evaluations during 30 +/- 7 months follow-up. To study primary versus secondary myocardial involvement, patients with MFS were divided in 2 groups: without previous surgery and normal/mild valvular function (MFS-1; N = 55) and with previous surgery or valvular dysfunction (MFS-2; N = 31). Results Compared to controls, patients in MFS-1 showed mild myocardial disease reflected in a larger left ventricular end-diastolic diameter (LVEDD), lower TAPSE and higher amount of (supra) ventricular extrasystoles [(S)VES]. Patients in MFS-2 were more severely affected. Seven patients (five in MFS-2) presented decreased LV ejection fraction. Twenty patients (twelve in MFS-2) had non-sustained ventricular tachycardia (NSVT) in at least one AECG. Larger LVEDD and higher amount of VES were independently associated with NSVT. Conclusion Our study shows mild but significant myocardial involvement in patients with MFS. Patients with previous surgery or valvular dysfunction are more severely affected. Evaluation of myocardial function with echocardiography and AECG should be considered in all patients with MFS, especially in those with valvular disease and a history of cardiac surgery

    The Third wave in globalization theory

    This essay examines a proposition made in the literature that there are three waves in globalization theory—the globalist, skeptical, and postskeptical or transformational waves—and argues that this division requires a new look. The essay is a critique of the third of these waves and its relationship with the second wave. Contributors to the third wave not only defend the idea of globalization from criticism by the skeptics but also try to construct a more complex and qualified theory of globalization than provided by first-wave accounts. The argument made here is that third-wave authors come to conclusions that try to defend globalization yet include qualifications that in practice reaffirm skeptical claims. This feature of the literature has been overlooked in debates and the aim of this essay is to revisit the literature and identify as well as discuss this problem. Such a presentation has political implications. Third wavers propose globalist cosmopolitan democracy when the substance of their arguments does more in practice to bolster the skeptical view of politics based on inequality and conflict, nation-states and regional blocs, and alliances of common interest or ideology rather than cosmopolitan global structures

    Metabolite Profiles of Sugarcane Culm Reveal the Relationship Among Metabolism and Axillary Bud Outgrowth in Genetically Related Sugarcane Commercial Cultivars

    Metabolic composition is known to exert influence on several important agronomic traits, and metabolomics, which represents the chemical composition in a cell, has long been recognized as a powerful tool for bridging phenotype–genotype interactions. In this work, sixteen truly representative sugarcane Brazilian varieties were selected to explore the metabolic networks in buds and culms, the tissues involved in the vegetative propagation of this species. Due to the fact that bud sprouting is a key trait determining crop establishment in the field, the sprouting potential among the genotypes was evaluated. The use of partial least square discriminant analysis indicated only mild differences on bud outgrowth potential under controlled environmental conditions. However, primary metabolite profiling provided information on the variability of metabolic features even under a narrow genetic background, typical for modern sugarcane cultivars. Metabolite–metabolite correlations within and between tissues revealed more complex patterns for culms in relation to buds, and enabled the recognition of key metabolites (e.g., sucrose, putrescine, glutamate, serine, and myo-inositol) affecting sprouting ability. Finally, those results were associated with the genetic background of each cultivar, showing that metabolites can be potentially used as indicators for the genetic background
