    An X-ray Investigation of Three Supernova Remnants in the Large Magellanic Cloud

    We have investigated three SNRs in the LMC using multi-wavelength data. These SNRs are generally fainter than the known sample and may represent a previously missed population. One of our SNRs is the second LMC remnant analyzed which is larger than any Galactic remnant for which a definite size has been established. The analysis of such a large remnant contributes to the understanding of the population of highly evolved SNRs. We have obtained X-ray images and spectra of three of these recently identified SNRs using the XMM-Newton observatory. These data, in conjunction with pre-existing optical emission-line images and spectra, were used to determine the physical conditions of the optical- and X-ray-emitting gas in the SNRs. We have compared the morphologies of the SNRs in the different wavebands. The physical properties of the warm ionized shell were determined from the H-alpha surface brightness and the SNR expansion velocity. The X-ray spectra were fit with a thermal plasma model and the physical conditions of the hot gas were derived from the model fits. Finally, we have compared our observations with simulations of SNR evolution

    Supernova Remnants in the Magellanic Clouds. IV. X-Ray Emission from the Largest SNR in the LMC

    We present the first X-ray detection of SNR 0450-70.9 the largest known supernova remnant (SNR) in the Large Magellanic Cloud. To study the physical conditions of this SNR, we have obtained XMM-Newton X-ray observations, optical images and high-dispersion spectra, and radio continuum maps. Optical images of SNR 0450-70.9 show a large, irregular elliptical shell with bright filaments along the eastern and western rims and within the shell interior. The interior filaments have higher [S II]/Halpha ratios and form an apparent inner shell morphology. The X-ray emission region is smaller than the full extent of the optical shell, with the brightest X-ray emission found within the small interior shell and on the western rim of the large shell. The expansion velocity of the small shell is ~220 km/s, while the large shell is ~120 km/s. The radio image shows central brightening and a fairly flat radio spectral index over the SNR. However, no point X-ray or radio source corresponding to a pulsar is detected and the X-ray emission is predominantly thermal. Therefore, these phenomena can be most reasonably explained in terms of the advanced age of the large SNR. Using hydrodynamic models combined with a nonequilibrium ionization model for thermal X-ray emission, we derived a lower limit on the SNR age of about 45,000 yr, well into the later stages of SNR evolution. Despite this, the temperature and density derived from spectral fits to the X-ray emission indicate that the remnant is still overpressured, and thus that the development is largely driven by hot gas in the SNR interior.Comment: Accepted for publication in The Astrophysical Journa

    Increased Hematopoietic Cells in the mertk-/- Mouse Peritoneal Cavity: A Result of Augmented Migration

    The peritoneal cavity is recognized as an important site for autoreactive B cells prior to their transit to other immune tissues; however, little is known of the genes that may regulate this process. Mice lacking the receptor tyrosine kinase Mertk display a lupus-like autoimmune phenotype with splenomegaly and high autoantibodies titers. Here, we investigate whether Mertk regulates the composition of peritoneal cells that favor an autoimmune phenotype. We found an increase in the number of macrophages, DC, plasmacytoid DC, T cells and B cells in the peritoneal cavity of mertk−/− mice when compared to wild-type mice. This disparity in cell numbers was not due to changes in cell proliferation or cell death. In adoptive transfer experiments, we showed an increase in migration of labeled donor cells into the mertk−/− peritoneal cavity. In addition, bone marrow chimeric mice showed hematopoietic-derived factors were also critical for T cell migration. Consistent with this migration and the increase in the number of cells, we identified elevated expression of CXCL9, its receptor CXCR3, and IL-7 receptor on peritoneal cells from mertk−/− mice. To corroborate the migratory function of CXCR3 on cells, the depletion of CXCR3 donor cells significantly reduced the number of adoptively transferred cells that entered into the peritoneum of mertk−/− mice. This control of peritoneal cells numbers correlated with autoantibody production and was exclusively attributed to Mertk since mice lacking other family members, Axl or Tyro 3, did not display dysregulation in peritoneal cell numbers or the autoimmune phenotype

    TAM receptors are dispensable in the phagocytosis and killing of bacteria

    Many receptors that are employed for the engulfment of apoptotic cells are also used for the recognition and phagocytosis of bacteria. Tyro3, Axl, and Mertk (TAM) are important in the phagocytosis of apoptotic cells by macrophages. Animals lacking these receptors are hypersensitive to bacterial products. In this report, we examine whether the TAM receptors are involved in the phagocytosis of bacteria. We found that macrophages lacking Mertk, Axl, Tyro3 or all three receptors were equally efficient in the phagocytosis of Gram-negative E. coli. Similarly, the phagocytosis of E. coli and Gram-positive S. aureus bioparticles by macrophages lacking TAM receptors was equal to wild-type. In addition, we found that Mertk did not play a role in killing of extracellular E. coli or the replication status of intracellular F. tularensis. Thus, while TAM receptors may regulate signal transduction to bacterial components, they are not essential for the phagocytosis and killing of bacteria

    Molecular Analyses of Circadian Gene Variants Reveal Sex-Dependent Links Between Depression and Clocks

    An extensive literature links circadian irregularities and/or sleep abnormalities to mood disorders. Despite the strong genetic component underlying many mood disorders, however, previous genetic associations between circadian clock gene variants and major depressive disorder (MDD) have been weak. We applied a combined molecular/functional and genetic association approach to circadian gene polymorphisms in sex-stratified populations of control subjects and case subjects suffering from MDD. This approach identified significant sex-dependent associations of common variants of the circadian clock genes hClock, hPer3 and hNpas2 with major depression and demonstrated functional effects of these polymorphisms on the expression or activity of the hCLOCK and hPER3 proteins, respectively. In addition, hCLOCK expression is affected by glucocorticoids, consistent with the sex-dependency of the genetic associations and the modulation of glucocorticoid-mediated stress response, providing a mechanism by which the circadian clock controls outputs that may affect psychiatric disorders. We conclude that genetic polymorphisms in circadian genes (especially hClock and hPer3, where functional assays could be tested) influence risk of developing depression in a sex- and stress-dependent manner. These studies support a genetic connection between circadian disruption and mood disorders, and confirm a key connection between circadian gene variation and major depression
