    Remote sensing of Earth's atmosphere and surface using a digital array scanned interferometer: A new type of imaging spectrometer

    The capabilities of the digital array scanned interferometer (DASI) class of instruments for measuring terrestrial radiation fields over the visible to mid-infrared are evaluated. DASI's are capable of high throughput, sensitivity and spectral resolution and have the potential for field-of-view spatial discrimination (an imaging spectrometer). The simplicity of design and operation of DASI's make them particularly suitable for field and airborne platform based remote sensing. The long term objective is to produce a versatile field instrument which may be applied toward a variety of atmospheric and surface studies. The operation of DASI and its advantages over other spectrometers are discussed

    Addition of a sequence from α2-antiplasmin transforms human serum albumin into a blood clot component that speeds clot lysis

    <p>Abstract</p> <p>Background</p> <p>The plasma protein α<sub>2</sub>-antiplasmin (α<sub>2</sub>AP) is cross-linked to fibrin in blood clots by the transglutaminase factor XIIIa, and in that location retards clot lysis. Competition for this effect could be clinically useful in patients with thrombosis. We hypothesized that fusion of N-terminal portions of α<sub>2</sub>-antiplasmin to human serum albumin (HSA) and production of the chimeric proteins in <it>Pichia pastoris </it>yeast would produce a stable and effective competitor protein.</p> <p>Results</p> <p>Fusion protein α<sub>2</sub>AP(13-42)-HSA was efficiently secreted from transformed yeast and purified preparations contained within a mixed population the full-length intact form, while fusions with longer α<sub>2</sub>AP moieties were inefficiently secreted and/or degraded. The α<sub>2</sub>AP(13-42)-HSA protein, but not recombinant HSA, was cross-linked to both chemical lysine donors and fibrin or fibrinogen by factor XIIIa, although with less rapid kinetics than native α<sub>2</sub>AP. Excess α<sub>2</sub>AP(13-42)-HSA competed with α<sub>2</sub>AP for cross-linking to chemical lysine donors more effectively than a synthetic α<sub>2</sub>AP(13-42) peptide, and reduced the α<sub>2</sub>AP-dependent resistance to fibrinolysis of plasma clots equally effectively as the peptide. Native α<sub>2</sub>AP was found in <it>in vivo </it>clots in rabbits to a greater extent than α<sub>2</sub>AP(13-42), however.</p> <p>Conclusion</p> <p>In this first report of transfer of transglutamination substrate status from one plasma protein to another, fusion protein α<sub>2</sub>AP(13-42)-HSA was shown to satisfy initial requirements for a long-lasting, well-tolerated competitive inhibitor of α<sub>2</sub>-antiplasmin predicted to act in a clot-localized manner.</p

    Variants within the MMP3 gene are associated with achilles tendinopathy: possible interaction with the COL5A1 gene

    Objectives: Sequence variation within the COL5A1 and TNC genes are known to associate with Achilles tendinopathy. The primary aim of this case-control genetic association study was to investigate whether variants within the matrix metalloproteinase 3 (MMP3) gene also contributed to both Achilles tendinopathy and Achilles tendon rupture in a Caucasian population. A secondary aim was to establish whether variants within the MMP3 gene interacted with the COL5A1 rs12722 variant to raise risk of these pathologies. Methods: 114 subjects with symptoms of Achilles tendon pathology and 98 healthy controls were genotyped for MMP3 variants rs679620, rs591058 and rs650108. Results: As single markers, significant associations were found between the GG genotype of rs679620 (OR = 2.5, 95% CI 1.2 to 4.90, p = 0.010), the CC genotype of rs591058 (OR = 2.3, 95% CI 1.1 to 4.50, p = 0.023) and the AA genotype of rs650108 (OR = 4.9, 95% CI 1.0 to 24.1, p = 0.043) and risk of Achilles tendinopathy. The ATG haplotype (rs679620, rs591058, and rs650108) was under-represented in the tendinopathy group when compared to the control group (41% vs 53%, p = 0.038). Finally, the G allele of rs679620 and the T allele of COL5A1 rs12722 significantly interacted to raise risk of AT (p = 0.006). No associations were found between any of the MMP3 markers and Achilles tendon rupture. Conclusion: Variants within the MMP3 gene are associated with Achilles tendinopathy. Furthermore, the MMP3 gene variant rs679620 and the COL5A1 marker rs12722 interact to modify the risk of tendinopathy.     Large-scale Star Formation Triggering in the Low-mass Arp 82 System: A Nearby Example of Galaxy Downsizing Based on UV/Optical/Mid-IR Imaging

    As part of our Spitzer Spirals, Bridges, and Tails project to help understand the effects of galaxy interactions on star formation, we analyze GALEX ultraviolet, SARA optical, and Spitzer infrared images of the interacting galaxy pair Arp 82 (NGC 2535/6) and compare to a numerical simulation of the interaction. We investigate the multiwavelength properties of several individual star forming complexes (clumps). Using optical and UV colors, EW(Halpha), and population synthesis models we constrain the ages of the clumps and find that the median clump age is about 12 Myr. The clumps have masses ranging from a few times 10^6 to 10^9 solar masses. In general, the clumps in the tidal features have similar ages to those in the spiral region, but are less massive. The 8 micron and 24 micron luminosities are used to estimate the far-infrared luminosities and the star formation rates of the clumps. The total clump star formation rate is 2.0+/-0.8 solar masses per year, while the entire Arp 82 system is forming stars at a rate of 4.9+/-2.0 solar masses per year. We find, for the first time, stars in the HI arc to the southeast of the NGC 2535 disk. Population synthesis models indicate that all of the observed populations have young to intermediate ages. We conclude that although the gas disks and some old stars may have formed early-on, the progenitors are late-type or low surface brightness and the evolution of these galaxies was halted until the recent encounter.Comment: Accepted for publication in the AJ, 22 Figures, 5 Table

    A method and vehicle preferably a wheelchair (10) for transporting and transferring an invalid person is described. The wheelchair has first and second frames (12 and 48) with drive wheels (18) between which is provided a chair (82) or support (83). The first frame has top and bottom sliding mechanisms (34 and 36) which are mounted by I-beam extensions (38 and 40) to the bars (24 and 26) of the first frame. The second frame has top and bottom rails (54 and 56) which are slidably connected to the sliding mechanisms of the first frame. The rails have slots (78) through which extend the center portions (38C and 40C) of the extensions which allow the first frame to move along the length of the rails. The bottom rails have front wheels (60), intermediate wheels (68) and stabilizing wheels (70). The chair is connected to the second frame by belts (84) and ratchets (86) and has a lifting system (90). In use, the wheelchair is moved adjacent the open doorway (104) of the automotive vehicle (100) and the second frame with the chair and person are slid over the seat (102). As the second frame moves, the second frame tilts such that the intermediate wheels touch the ground surface (72). The seat is lowered using the ratchets and removed from the wheelchair

    EST Express: PHP/MySQL based automated annotation of ESTs from expression libraries

    <p>Abstract</p> <p>Background</p> <p>Several biological techniques result in the acquisition of functional sets of cDNAs that must be sequenced and analyzed. The emergence of redundant databases such as UniGene and centralized annotation engines such as Entrez Gene has allowed the development of software that can analyze a great number of sequences in a matter of seconds.</p> <p>Results</p> <p>We have developed "EST Express", a suite of analytical tools that identify and annotate ESTs originating from specific mRNA populations. The software consists of a user-friendly GUI powered by PHP and MySQL that allows for online collaboration between researchers and continuity with UniGene, Entrez Gene and RefSeq. Two key features of the software include a novel, simplified Entrez Gene parser and tools to manage cDNA library sequencing projects. We have tested the software on a large data set (2,016 samples) produced by subtractive hybridization.</p> <p>Conclusion</p> <p>EST Express is an open-source, cross-platform web server application that imports sequences from cDNA libraries, such as those generated through subtractive hybridization or yeast two-hybrid screens. It then provides several layers of annotation based on Entrez Gene and RefSeq to allow the user to highlight useful genes and manage cDNA library projects.</p

    Bacterial defences: mechanisms, evolution and antimicrobial resistance

    Throughout their evolutionary history, bacteria have faced diverse threats from other microorganisms, including competing bacteria, bacteriophages and predators. In response to these threats, they have evolved sophisticated defence mechanisms that today also protect bacteria against antibiotics and other therapies. In this Review, we explore the protective strategies of bacteria, including the mechanisms, evolution and clinical implications of these ancient defences. We also review the countermeasures that attackers have evolved to overcome bacterial defences. We argue that understanding how bacteria defend themselves in nature is important for the development of new therapies and for minimizing resistance evolution

    IL-21 receptor expression in human tendinopathy

    The pathogenetic mechanisms underlying tendinopathy remain unclear, with much debate as to whether inflammation or degradation has the prominent role. Increasing evidence points toward and early inflammatory infiltrate and associated inflammatory cytokine production in human and animal models of tendon disease. The IL-21/IL-21R axis is a proinflammatory cytokine complex that has been associated with chronic inflammatory diseases including rheumatoid arthritis and inflammatory bowel disease. This project aimed to investigate the role and expression of the cytokine/receptor pair IL-21/IL-21R in human tendinopathy. We found significantly elevated expression of IL-21 receptor message and protein in human tendon samples but found no convincing evidence of the presence of IL-21 at message or protein level. The level of expression of IL-21R message/protein in human tenocytes was significantly up regulated by proinflammatory cytokines (TNFα/IL-1β) in vitro. These findings demonstrate that IL-21R is present in early human tendinopathy mainly expressed by tenocytes and macrophages. Despite a lack of IL-21 expression these data again suggest that early tendinopathy has an inflammatory/cytokine phenotype, which may provide novel translational targets in the treatment of tendinopathy

    Searching for Planets in the Hyades. I. The Keck Radial Velocity Survey

    We describe a high-precision radial velocity search for jovian-mass companions to main sequence stars in the Hyades star cluster. The Hyades provides an extremely well controlled sample of stars of the same age, the same metallicity, and a common birth and early dynamical environment. This sample allows us to explore the dependence of the process of planet formation on only a single independent variable: the stellar mass.
