    Formal coach education programmes, driven by the national governing bodies of individual sports, have for many years been regarded as essential in the development of competent, qualified coaches. However, more recent opinion suggests that such coach education is actually ‘‘low impact’’ in comparison to the hours individual aspirant coach’s spend coaching, being coached or participating (Rossi & Cassidy, 1999: Learning and teaching in physical education. London: Falmer). It appears the literature is, in part, implying that experiential learning has greater significance over formal education programmes. The aim of this study was to investigate the educational journey of a sample of paddlesport coaches by addressing why coaches chose to enter formal coach education programmes

    Dynamic Modeling of Starting Aerodynamics and Stage Matching in an Axi-Centrifugal Compressor

    A DYNamic Turbine Engine Compressor Code (DYNTECC) has been modified to model speed transients from 0-100% of compressor design speed. The impetus for this enhancement was to investigate stage matching and stalling behavior during a start sequence as compared to rotating stall events above ground idle. The model can simulate speed and throttle excursions simultaneously as well as time varying bleed flow schedules. Results of a start simulation are presented and compared to experimental data obtained from an axi-centrifugal turboshaft engine and companion compressor rig. Stage by stage comparisons reveal the front stages to be operating in or near rotating stall through most of the start sequence. The model matches the starting operating line quite well in the forward stages with deviations appearing in the rearward stages near the start bleed. Overall, the performance of the model is very promising and adds significantly to the dynamic simulation capabilities of DYNTECC

    A NuRD Complex from Xenopus laevis Eggs Is Essential for DNA Replication during Early Embryogenesis.

    DNA replication in the embryo of Xenopus laevis changes dramatically at the mid-blastula transition (MBT), with Y RNA-independent random initiation switching to Y RNA-dependent initiation at specific origins. Here, we identify xNuRD, an MTA2-containing assemblage of the nucleosome remodeling and histone deacetylation complex NuRD, as an essential factor in pre-MBT Xenopus embryos that overcomes a functional requirement for Y RNAs during DNA replication. Human NuRD complexes have a different subunit composition than xNuRD and do not support Y RNA-independent initiation of DNA replication. Blocking or immunodepletion of xNuRD inhibits DNA replication initiation in isolated nuclei in vitro and causes inhibition of DNA synthesis, developmental delay, and embryonic lethality in early embryos. xNuRD activity declines after the MBT, coinciding with dissociation of the complex and emergence of Y RNA-dependent initiation. Our data thus reveal an essential role for a NuRD complex as a DNA replication factor during early Xenopus development

    Guidelines on the management of ascites in cirrhosis.

    The British Society of Gastroenterology in collaboration with British Association for the Study of the Liver has prepared this document. The aim of this guideline is to review and summarise the evidence that guides clinical diagnosis and management of ascites in patients with cirrhosis. Substantial advances have been made in this area since the publication of the last guideline in 2007. These guidelines are based on a comprehensive literature search and comprise systematic reviews in the key areas, including the diagnostic tests, diuretic use, therapeutic paracentesis, use of albumin, transjugular intrahepatic portosystemic stent shunt, spontaneous bacterial peritonitis and beta-blockers in patients with ascites. Where recent systematic reviews and meta-analysis are available, these have been updated with additional studies. In addition, the results of prospective and retrospective studies, evidence obtained from expert committee reports and, in some instances, reports from case series have been included. Where possible, judgement has been made on the quality of information used to generate the guidelines and the specific recommendations have been made according to the 'Grading of Recommendations Assessment, Development and Evaluation (GRADE)' system. These guidelines are intended to inform practising clinicians, and it is expected that these guidelines will be revised in 3 years' time

    Veterinary trypanocidal benzoxaboroles are peptidase-activated prodrugs

    Livestock diseases caused by Trypanosoma congolense, T. vivax and T. brucei, collectively known as nagana, are responsible for billions of dollars in lost food production annually. There is an urgent need for novel therapeutics. Encouragingly, promising antitrypanosomal benzoxaboroles are under veterinary development. Here, we show that the most efficacious subclass of these compounds are prodrugs activated by trypanosome serine carboxypeptidases (CBPs). Drug-resistance to a development candidate, AN11736, emerged readily in T. brucei, due to partial deletion within the locus containing three tandem copies of the CBP genes. T. congolense parasites, which possess a larger array of related CBPs, also developed resistance to AN11736 through deletion within the locus. A genome-scale screen in T. brucei confirmed CBP loss-of-function as the primary mechanism of resistance and CRISPR-Cas9 editing proved that partial deletion within the locus was sufficient to confer resistance. CBP re-expression in either T. brucei or T. congolense AN11736-resistant lines restored drug-susceptibility. CBPs act by cleaving the benzoxaborole AN11736 to a carboxylic acid derivative, revealing a prodrug activation mechanism. Loss of CBP activity results in massive reduction in net uptake of AN11736, indicating that entry is facilitated by the concentration gradient created by prodrug metabolism.</p
